Factors Influencing Microbial Growth: pH

Les micro-organismes sont classés comme acidophiles, neutrophiles ou alcalinophiles en fonction de leur pH préféré pour la croissance. Les neutrophiles, tels que E. coli, se développent entre 5,5 et 8,0. Contrairement aux bactéries, les moisissures et les levures préfèrent un pH plus bas entre 5 et 6. Les acidophiles prospèrent à un pH encore plus acide entre 0 et 5,5, avec Picrophilus oshimae survivant à un pH de 0,7. Les alcaliphiles prospèrent à un pH de 8 ou plus, habitant généralement les lacs de soude et les sols riches en carbonate. De même, la plupart des microbes marins sont adaptés à un environnement légèrement alcalin, autour d’un pH de 8,1. La survie microbienne dépend du maintien d’un pH intracellulaire stable pour la stabilité macromoléculaire en réponse aux variations des conditions de pH externes. Les neutrophiles maintiennent l’homéostasie du pH grâce à des mécanismes de transport de protons tels que les échangeurs K-H et le tampon cytoplasmique. Les acidophiles s’appuient sur des membranes imperméables et des pompes à protons pour limiter l’afflux de protons. Les alcalinophiles maintiennent un pH interne presque neutre en échangeant des ions sodium contre des protons externes. Dans les cultures de laboratoire, des tampons de phosphate sont ajoutés au milieu pour empêcher les changements de pH et maintenir des conditions de croissance optimales.

Microbial growth depends on pH, the measure of how acidic or alkaline an environment is. Different microorganisms grow best in different pH ranges. Their ability to keep a stable internal pH helps protect enzymes and other macromolecules. External pH changes can challenge that balance.

Neutrophiles grow best near neutral pH, usually between 5.5 and 8.0. Escherichia coli is a common example. These microbes often live in neutral or slightly acidic places. They can use potassium-proton exchange and intracellular buffering to help control internal pH. Molds and yeasts are also classed as neutrophiles, but they can tolerate more acidic conditions and grow best around pH 5 to 6.

Acidophiles prefer very acidic environments, with an optimal pH range from 0 to 5.5. Picrophilus oshimae can survive at a pH as low as 0.7. These organisms use special adaptations to stay stable in acid. Their membranes limit proton entry, and proton pumps remove excess hydrogen ions to keep internal pH in balance.

Alkaliphiles grow best in alkaline conditions above pH 8. They are often found in soda lakes and carbonate-rich soils. Many of these microbes exchange external protons for sodium ions to keep their cytoplasm near neutral. Some marine microbes also fit this pattern because ocean water has an average pH of about 8.1.

In the lab, scientists often add buffers such as phosphate salts to culture media. Buffers help stabilize pH and keep growth conditions within the right range. This prevents sudden pH changes that could disturb cell function. These pH-based adaptations show how microbes survive in very different environments and how researchers can better grow them for scientific and industrial use.
