La differenza relativa nella carica elettrica, o voltaggio, tra l'interno e l'esterno di una membrana cellulare, è chiamata potenziale della membrana. È generato da differenze nella permeabilità della membrana a vari ioni e dalle concentrazioni di questi ioni attraverso la membrana.

<h4>L'interno di un neurone è più negativo</h4>

Il potenziale di membrana di una cellula può essere misurato inserendo un microelettrodo in una cellula e confrontando la carica con un elettrodo di riferimento nel fluido extracellulare. Il potenziale di membrana di un neurone a riposo, cioè un neurone che attualmente non riceve o invia messaggi, è negativo, tipicamente intorno a -70 millivolt (mV). Questo è chiamato il potenziale della membrana a riposo. Il valore negativo indica che l'interno della membrana è relativamente più negativo rispetto all'esterno, è polarizzato. Il potenziale di riposo deriva da due fattori principali: permeabilità selettiva della membrana e differenze nella concentrazione di ioni all'interno della cellula rispetto all'esterno.

<h4>Permeabilità della membrana</h4>

Le membrane cellulari sono permeabili selettivamente perché la maggior parte degli ioni e delle molecole non può attraversare il bistrato lipidico senza aiuto, spesso da proteine del canale ionico che attraversano la membrana. Questo perché gli ioni caricati non possono diffondersi attraverso l'interno idrofobico non caricato delle membrane. Gli ioni intra ed extracellulari più comuni che si trovano nel tessuto nervoso sono il potassio(K)), il sodio(Na), il cloruro (Cl-) e il calcio (Ca2). Quando un neurone è a riposo, i canali di potassio(K)sono il tipo principale di canale ionico che è aperto, permettendo a K- di migrare attraverso la membrana. Questa permeabilità, insieme alle grandi concentrazioni intracellulari, rende il potenziale di membrana a riposo del neurone determinato principalmente dal movimento diK.

<h4>Pompe Creano Gradienti di Concentrazione</h4>

Le differenze nella concentrazione di ioni tra l'interno e l'esterno dei neuroni sono principalmente dovute all'attività della pompa sodio-potassio(Na, /K) , una proteina transmembrana che pompa continuamente treioni Na fuori dalla cellula per ogni due K: ioni all'interno. Questo stabilisce gradienti di concentrazione, con una maggiore concentrazione di ioni di Na, al di fuori dei neuroni e una maggiore concentrazione diioni K all'interno.

Dal momento che la membrana è principalmente permeabile a K, a riposo, a causa deicanali K aperti, Kpuò diffondere il suo gradiente di concentrazione nella regione di minore concentrazione, fuori dalla cellula. <h4>Attenzione al pesce Palla</h4>

Il pesce palla non si trova spesso in molti menu di pesce al di fuori del Giappone, in parte perché contengono una potente neurotossina. La tetrodotossina (TTX) è un bloccante del canale di sodio molto selettivo che è letale in dosi minime. La dose letale mediana (LD50) per i topi è di 334 g/kg, rispetto agli 8,5 mg/kg per il cianuro di potassio. Ha anche servito come strumento essenziale nella ricerca sulle neuroscienze. Quando si apre il canale, la tossina blocca il flusso di Nanella cella. Pertanto, interrompe i potenziali di azione, ma non il potenziale della membrana a riposo, e può essere utilizzato per silenziare l'attività neuronale. The Resting Membrane Potential

Neurons keep a negative resting charge across their membrane. This electrical difference, called the membrane potential, comes from how ions move and how the membrane lets them pass. In a resting neuron, the inside is typically around -70 millivolts and is more negative than the outside, so the cell is polarized.

Two main factors create this resting state. First, the membrane is selectively permeable, which means most ions and molecules cannot cross the lipid bilayer without help from ion channels. Second, ion concentrations are different inside and outside the cell. In nervous tissue, common ions include potassium (K+), sodium (Na+), chloride (Cl-), and calcium (Ca2+).

At rest, potassium channels are the main channels that are open. This makes potassium the ion that most strongly shapes the resting membrane potential. Potassium can move across the membrane more easily than many other ions, and its movement helps set the neuron’s negative charge.

The sodium-potassium pump creates the ion gradients that support this process. This transmembrane protein pumps three Na+ ions out of the cell for every two K+ ions it pumps in. As a result, Na+ builds up outside the neuron, and K+ builds up inside.

Because the membrane is mostly permeable to K+ at rest, potassium diffuses out of the cell down its concentration gradient. As positive charges leave, the inside becomes more negative. Negatively charged proteins inside the cell also contribute to this effect. Eventually, the outward movement of K+ is balanced by the electrical push from the positive charge outside the cell, and electrochemical equilibrium is reached.

This resting potential is important because changes in membrane potential are the basis of neural signaling. One example is the action potential, which depends on these voltage changes. The pufferfish toxin tetrodotoxin (TTX) blocks voltage-gated sodium channels and stops Na+ from entering when the channel opens. It disrupts action potentials, but it does not change the resting membrane potential, so it can be used to silence neuronal activity.
