Potenziamento a lungo termine, o LTP, è uno dei modi con cui la plasticità sinaptica - cambiamenti nella forza delle sinapsi chimiche - può verificarsi nel cervello. LTP è il processo di rafforzamento sinaptico che si verifica nel tempo tra connessioni neuronali pre e post-sinaptiche. Il rafforzamento sinaptico di LTP funziona in opposizione all'indebolimento sinaptico della depressione a lungo termine (LTD) e insieme sono i principali meccanismi che sono alla base dell'apprendimento e della memoria.

<h4>Hebbian LTP</h4>

LTP può verificarsi quando i neuroni presinaptici "sparano" ripetutamente e stimolano il neurone post-sinaptico. Questo si chiama Hebbian LTP poiché ne consegue che il postulato di Donald Hebb del 1949 "neuroni che sparano insieme potenziano insieme". La stimolazione ripetuta da neuroni presinaptici induce cambiamenti nel tipo e nel numero di canali ionici nella membrana post-sinaptica.

Due tipi di recettori postsinaptici del glutammato del neurotrasmettitore eccitatorio sono coinvolti in LTP: 1) recettori N-metil-D-aspartato o NMDA e 2) recettori dell'acido o AMPA di-amino-3-idrossili-5-metili-4-isoxazolepropionici. Anche se i recettori NMDA si aprono su legame glutammato, il loro poro è di solito bloccato da ioni di magnesio che impediscono ad altri ioni caricati positivamente di entrare nel neurone. Tuttavia, il glutammato rilasciato dai neuroni presininaptici può legarsi ai recettori AMPA postsinaptici, causando un afflusso di ioni di sodio che si traduce nella depolarizzazione della membrana. Quando la membrana post-sinaptica è depolarizzata da molteplici ingressi presnapsitici frequenti, lo ione di magnesio che blocca il poro del recettore NMDA viene spostato, permettendo ai ioni di sodio e calcio di fluire nel neurone.

L'aumento dell'afflusso di ioni di calcio avvia quindi una cascata di segnalazione che culmina in più recettori AMPA inseriti nella membrana plasmatica. In alternativa, la cascata di segnalazione può fosforilare i recettori del glutammato, consentendo loro di rimanere aperti per una durata più lunga e migliorando la conduttanza degli ioni caricati positivamente nella cellula. Di conseguenza, la stessa stimolazione presinaptica ora evocherà una risposta post-sinaptica più forte dato che più recettori del glutammato saranno attivati e ioni più caricati positivamente entreranno nel neurone post-sinaptico. L'amplificazione che si verifica è conosciuta come rinforzo sinaptico o potenziamento.

IL detto "la pratica rende perfetto" può essere in parte spiegato da LTP. Quando si impara un nuovo compito, nuovi circuiti neurali vengono rinforzati utilizzando LTP. Dopo ogni iterazione di pratica, la forza sinaptica nei circuiti neurali diventa più forte e presto il compito può essere eseguito in modo corretto ed efficiente. Le connessioni appena rafforzate possono durare da minuti a settimane o più se la stimolazione pressinaptica persiste, il che significa che ogni volta che l'attività viene eseguita viene mantenuta.

<h4>LTP e malattie</h4>

Quando LTP funziona normalmente, possiamo imparare e formare i ricordi con facilità. Tuttavia, anomalie in LTP sono stati implicati in molti disturbi neurologici e cognitivi come il morbo di Alzheimer, autismo, dipendenza, schizofrenia, e sclerosi multipla. Long-term potentiation, or LTP, is a form of synaptic strengthening in the brain. It changes the strength of chemical synapses over time between pre- and postsynaptic neurons. LTP works against long-term depression, or LTD, which weakens synapses. Together, LTP and LTD are major mechanisms behind learning and memory.

LTP can happen when a presynaptic neuron fires again and again and keeps stimulating a postsynaptic neuron. This is called Hebbian LTP, based on Donald Hebb’s idea that neurons that fire together wire together. Repeated stimulation changes the types and number of ion channels in the postsynaptic membrane. Two glutamate receptors are involved: NMDA receptors and AMPA receptors.

NMDA receptors open when glutamate binds, but magnesium ions usually block the pore. At the same time, glutamate can activate AMPA receptors on the postsynaptic cell. This causes sodium ions to enter and depolarize the membrane. When many presynaptic inputs arrive in quick succession, the magnesium block is removed from the NMDA receptor. Sodium and calcium ions then enter the neuron.

The calcium influx starts a signaling cascade inside the cell. This cascade leads to more AMPA receptors being inserted into the plasma membrane. It can also phosphorylate glutamate receptors, which helps them stay open longer and lets more positively charged ions enter the cell. The same presynaptic signal then creates a stronger postsynaptic response. This increase is called synaptic strengthening, or potentiation.

LTP also helps explain why practice makes perfect. When a new task is learned, new neural circuits are reinforced by LTP. With each round of practice, the synaptic strength in those circuits becomes stronger. Over time, the task can be performed more correctly and efficiently. These strengthened connections can last from minutes to weeks or longer if presynaptic stimulation continues, so later practice can maintain the LTP.

When LTP works normally, learning and memory are easier. Problems with LTP have been linked to Alzheimer’s disease, autism, addiction, schizophrenia, and multiple sclerosis. Understanding how LTP works may help guide future therapies for these neurological and cognitive disorders.
