Ricordiamo che in una reazione di sostituzione nucleofila, un nucleofilo dona i suoi elettroni a un elettrofilo.
Gli elettrofili sono reagenti a ricerca di elettroni, neutri o caricati positivamente, contenenti un orbitale atomico vuoto o un orbitale di antilegame a bassa energia.
Un elettrofilo positivo, come il protone, con un orbitale 1s vuoto e a bassa energia, è molto reattivo. Di conseguenza, un nucleofilo come lo ione idrossido attacca il protone, neutralizzando la carica e formando acqua.
Un altro elettrofilo positivo, il carbocatione, ha un orbitale p vuoto, che lo rende reattivo nei confronti di un attacco nucleofilo. 
Un elettrofilo neutro, come il trifluoruro di boro dell'acido di Lewis, ha un orbitale p vuoto che può accettare elettroni dal nucleofilo, formando così un legame e risultando in un complesso stabile.
In una molecola neutra come il clorobutano, il centro elettrofilo risulta dall'effetto induttivo di ritiro degli elettroni del sostituente più elettronegativo attaccato alla catena molecolare.
In un elettrofilo organico con un atomo elettronegativo a doppio legame, come il gruppo carbonilico, il dipolo del legame C=O fornisce una carica positiva parziale all'atomo di carbonio.
In una reazione, il nucleofilo deposita i suoi elettroni nell'orbitale di antilegame a bassa energia π dell'elettrofilo. Di conseguenza, il legame C-O π si rompe e gli elettroni si spostano verso l'atomo di ossigeno. 
In un elettrofilo, come l'HCl, che consiste in un atomo elettronegativo a legame singolo, il dipolo del legame σ costringe gli elettroni nucleofili a spostarsi nell'orbitale σ di antilegame HCl a bassa energia, e quindi rompe il legame.
Anche molecole come gli alogeni, con legami σ e senza dipoli, sono buoni elettrofili. Nel bromo, ad esempio, scarse sovrapposizioni tra gli orbitali atomici degli atomi di bromo risultano in un debole legame Br-Br.
Così, un nucleofilo attacca l'orbitale di antilegame σ energia più bassa, rompendo il legame Br-Br e creando un nuovo legame.

Questa lezione spiega la definizione, la classificazione e le caratteristiche di un elettrofilo che sono le caratteristiche chiave delle reazioni di sostituzione nucleofila. Ciò spiega la carica positiva parziale sull'atomo di carbonio in un gruppo carbonilico.

Nel contesto di una reazione chimica è necessario un quadro completo del trasferimento di elettroni in questo processo. Un nucleofilo deposita i suoi elettroni nell'orbitale π di antilegame a bassa energia dell'elettrofilo. Al contrario, il dipolo del legame σ forza gli elettroni nucleofili a spostarsi nell’orbitale σ di antilegame a bassa energia, con conseguente rottura del legame. Questi fenomeni sono spesso spiegati utilizzando esempi di gruppi carbonilici e HCl. Tipicamente, gli orbitali molecolari occupati più bassi (LUMO) negli elettrofili organici sono orbitali di antilegame a bassa energia, poiché sono associati ad atomi elettronegativi. Questi possono essere orbitali π* o orbitali σ*.

Alcune molecole, come gli alogeni, sono anche buoni elettrofili. Qui, nonostante l'assenza di un dipolo, una scarsa sovrapposizione tra gli orbitali atomici dei due alogenuri indebolisce il legame rendendolo più incline ad un attacco nucleofilo. Electrophile reactivity in organic reactions depends on how a species attracts electrons. Electrophiles are electron-poor and are key partners in nucleophilic substitution reactions. Their charge and orbital structure help explain why they react the way they do.

Electrophiles can be positive or neutral species. A positive electrophile such as a proton reacts because it has a vacant, low-energy 1s orbital. Other positive electrophiles, such as carbocations, are reactive because they have an empty p orbital.

Neutral electrophiles act like Lewis acids. They have empty p orbitals that can accept electrons from a nucleophile and form stable complexes. In some neutral molecules, an electrophilic center appears because an electronegative substituent pulls electron density away through the inductive effect. This explains the partial positive charge on the carbon atom in a carbonyl group.

During a reaction, electron transfer must be viewed as a whole. A nucleophile donates electrons into the lower-energy antibonding pi orbital of the electrophile. In other cases, the dipole of a sigma bond drives the nucleophilic electrons into a lower-energy antibonding sigma orbital, which leads to bond breaking. Carbonyl groups and HCl are common examples of these ideas.

In organic molecules, the lowest unoccupied molecular orbitals, or LUMOs, are often low-energy antibonding orbitals. These are usually pi-star or sigma-star orbitals, especially when electronegative atoms are involved. Halogens can also act as electrophiles. Even without a dipole, poor overlap between the two halide atomic orbitals weakens the bond and makes it easier for a nucleophile to attack. In many cases, molecules with a single or double bond linked to O, N, Cl, or Br make good electrophiles.
