Quando un nucleofilo si avvicina all'elettrofilo dallo stesso lato del gruppo uscente, si trova di fronte a un nodo, il che provoca la sovrapposizione dell'HOMO con il LUMO di legame e antilegame. Tuttavia, non si forma alcun legame poiché la sovrapposizione dell'antilegame annulla la sovrapposizione del legame.
Al contrario, l'approccio posteriore del nucleofilo si sovrappone in modo efficiente all'HOMO con il LUMO dell'elettrofilo, portando alla formazione di legami.
Pertanto, entrambe le teorie supportano che il meccanismo di reazione SN2 sia concertato in cui il nucleofilo attacca dal lato posteriore mentre contemporaneamente sposta il gruppo uscente e causa un'inversione di configurazione.

La presenza dell'alogeno più elettronegativo nel substrato crea un legame polarizzato di alogenuro di carbonio. L'alogenuro attira la nuvola di elettroni generando un centro elettrofilo nell'atomo di carbonio. Pertanto, l'atomo di carbonio porta una carica parziale positiva mentre l'alogenuro ha una carica parziale negativa. Il carbonio elettrofilo attrae il nucleofilo con la sua coppia solitaria di elettroni.

Tuttavia, l’elevata densità elettronica attorno all’alogenuro blocca efficacemente l’attacco dallo stesso lato da parte del nucleofilo. Pertanto, il nucleofilo si avvicina all'elettrofilo dal lato povero di elettroni del substrato, provocando un attacco dal lato posteriore. Il nucleofilo dona così il suo doppietto al carbonio elettrofilo, a 180° di distanza dal gruppo uscente.

Quando l'alogenuro si allontana dal carbonio elettrofilo, lo fa con la coppia di elettroni legata al carbonio. Ciò si traduce in uno stato di transizione con un legame parzialmente formato tra il nucleofilo e il substrato e un legame parzialmente rotto tra il substrato e il gruppo uscente.

Con tre legami solidi e due parziali al carbonio, lo stato di transizione è altamente instabile. Pertanto, il gruppo uscente si allontana con la coppia di elettroni legata al carbonio, portando all'inversione della configurazione del substrato. (Figura 1)

Inoltre, la teoria dell'orbitale molecolare supporta anche l'attacco dorsale. Quando l'orbitale di legame del nucleofilo, cioè l'orbitale molecolare più occupato, o HOMO, si avvicina all'orbitale molecolare non occupato più basso, o LUMO, del substrato dallo stesso lato del gruppo uscente, si trova di fronte ad un nodo che annulla sia l'orbitale di legame sia quello di antilegame. Al contrario, un attacco dorsale da parte del nucleofilo si sovrappone efficacemente al LUMO del substrato, determinando la formazione di un legame.

Pertanto, il meccanismo SN2 avviene in un unico passaggio, quando il nucleofilo entrante reagisce con il substrato da una direzione opposta al gruppo uscente, che viene spostato.

SN2 reactions happen in one step and end with inversion of configuration. The nucleophile and the substrate both take part in the rate-determining step, and no intermediate forms.

The substrate contains a polarized carbon-halide bond. The more electronegative halogen pulls the electron cloud toward itself. That leaves the carbon atom electrophilic, or electron-poor, with a partial positive charge. The halide has a partial negative charge.

The electrophilic carbon attracts the nucleophile, which brings a lone pair of electrons. But the electron-rich halide blocks attack from the same side. For that reason, the nucleophile approaches from the opposite side of the leaving group. This is called a backside attack.

In the transition state, the nucleophile is partly bonded to the carbon while the carbon-halide bond is partly broken. The carbon has three solid bonds and two partial bonds, so this state is very unstable. As the halide leaves with the electron pair, the substrate flips and its configuration inverts.

Molecular orbital theory also supports this path. The nucleophile’s highest occupied molecular orbital, or HOMO, matches poorly with the substrate’s lowest unoccupied molecular orbital, or LUMO, from the same side because of a node. A backside attack gives better overlap and allows bond formation. So the SN2 mechanism is a single-step reaction in which the nucleophile attacks opposite the leaving group and displaces it.
