Quando un alogenuro alchilico reagisce con un nucleofilo, il nucleofilo può spostare l'alogeno per dare il prodotto di sostituzione o può astrarre un idrogeno vicino per formare un alchene attraverso una reazione di eliminazione.
Nelle reazioni di eliminazione, i nucleofili funzionano come basi di Lewis donando una coppia di elettroni a un protone. Alcune delle basi comuni utilizzate per promuovere le reazioni di eliminazione includono idrossidi come l'idrossido di sodio, alcossidi come il terz-butossido di potassio o alcoli come l'etanolo.
Le reazioni di eliminazione comportano tipicamente la perdita di piccoli frammenti molecolari da un substrato per formare almeno un legame π. Negli alogenuri alchilici, la reazione di eliminazione procede con la perdita di un atomo di idrogeno e di un atomo di alogeno, da cui il nome deidroalogenazione.
Poiché il carbonio legato al gruppo uscente è un carbonio α e l'idrogeno sul carbonio adiacente è un idrogeno β, queste reazioni sono spesso chiamate reazioni di β-eliminazione o reazioni di eliminazione 1,2.
La maggior parte delle reazioni di eliminazione avviene tramite un meccanismo E2 o E1.
Per le reazioni E2 vengono utilizzate basi forti come l'etossido di sodio. Il meccanismo concertato è avviato dalla deprotonazione del carbonio β seguita dalla partenza del gruppo uscente dall'alogenuro, portando alla formazione di un legame π tra le posizioni α e β.
Al contrario, la reazione E1 procede in due fasi. Il primo prevede la partenza del gruppo uscente per formare un carbocatione intermedio, seguito dalla deprotonazione del carbocatione da parte della base per formare un legame π.
Con alogenuri alchilici contenenti due diversi atomi di carbonio β, le reazioni di eliminazione possono produrre più di un alchene. Qui, l'alchene più sostituito è il più stabile ed è chiamato prodotto di Zaitsev, mentre l'alchene meno sostituito è chiamato prodotto di Hofmann. Elimination Reactions

Un nucleofilo può reagire con un alogenuro alchilico per dare il prodotto di sostituzione spostando l'alogeno. Oppure può funzionare come base per dare il prodotto di eliminazione mediante deprotonazione del carbonio vicino per formare un alchene. In una reazione di eliminazione, il substrato perde due gruppi di atomi di carbonio adiacenti formando almeno un legame π. Il carbonio attaccato all'alogeno è chiamato carbonio α, mentre il carbonio adiacente è chiamato carbonio β; quindi, queste reazioni sono chiamate reazioni di eliminazione β o reazioni di eliminazione 1,2.

Il nucleofilo agisce come una base di Lewis donando una coppia di elettroni ad un protone. Le basi comuni utilizzate per promuovere le reazioni di eliminazione includono idrossidi (OH−), alcossidi (OR−) e ammidi (NH2−). In presenza di una base forte, l'alogenuro alchilico perde un protone dal carbonio β e l'alogeno dal carbonio α, consentendo la formazione di un legame π tra i due atomi di carbonio.

Meccanismo delle reazioni di eliminazione

Le reazioni di eliminazione si verificano comunemente tramite i meccanismi E2 o E1. Il meccanismo E2 avviene in un unico passaggio concertato: l'estrazione dell'idrogeno β da parte della base è accompagnata dalla scissione del legame carbonio α-alogeno. Pertanto, la reazione E2 procede attraverso uno stato di transizione.

La reazione E1 avviene in due fasi. Innanzitutto, l'alogenuro alchilico subisce la ionizzazione formando un carbocatione intermedio e uno ione alogenuro. Successivamente, la deprotonazione del carbocatione da parte della base dà luogo ad un legame π. Pertanto, nelle reazioni E1, l'intermedio carbocationico si forma attraverso uno stato di transizione ed esiste un secondo stato di transizione per la fase di deprotonazione.

Regio- e stereoselettività

Quando l'alogenuro alchilico ha due diversi carboni β, la reazione di eliminazione può produrre più di un alchene. In questi casi, si osserva generalmente l'alchene più sostituito (e più stabile), noto come prodotto di Zaitsev. Tuttavia, in alcuni casi, si ottiene l'alchene meno sostituito (prodotto di Hofmann). La scelta della base gioca un ruolo importante nel decidere quale prodotto regioselettivo si formerà. Le reazioni di eliminazione favoriscono anche la formazione di trans-alcheni rispetto agli isomeri cis, rendendoli stereoselettivi.

Elimination reactions of alkyl halides form alkenes by removing groups from adjacent carbons. A base can act on the substrate instead of causing substitution. It deprotonates the neighboring carbon, and the reaction creates at least one pi bond. The carbon bonded to the halogen is the alpha carbon, and the adjacent carbon is the beta carbon. These are also called beta elimination or 1,2-elimination reactions.

Strong bases help drive this process. Common examples include hydroxides, alkoxides, and amides. The base donates an electron pair to a proton, so it behaves as a Lewis base. In the presence of a strong base, the alkyl halide loses a proton from the beta carbon and the halogen from the alpha carbon. This lets a pi bond form between the two carbon atoms.

Elimination reactions often follow either the E2 or E1 mechanism. The E2 pathway is a single concerted step. The base removes the beta hydrogen at the same time that the alpha carbon-halogen bond breaks. This means E2 has one transition state.

The E1 pathway happens in two steps. First, the alkyl halide ionizes to form a carbocation intermediate and a halide ion. Then the base removes a proton from the carbocation, which forms the pi bond. In E1 reactions, the first step has one transition state, and the deprotonation step has a second transition state.

When an alkyl halide has two different beta carbons, more than one alkene can form. The more substituted and usually more stable alkene is called the Zaitsev product. In some cases, the less substituted Hofmann product forms instead, and the base helps decide which product is favored. Elimination reactions also tend to give trans-alkenes over cis isomers, so they are stereoselective.
