pH Regulation in Cells

Il pH è una misura della concentrazione di H+ in una soluzione e indica il suo livello di acidità o alcalinità.
La scala del pH va da 0 a 14, dove sette è considerato neutro, un valore inferiore a sette è acido e un valore superiore a sette è basico.
Il tipico pH citoplasmatico si trova in un intervallo leggermente alcalino compreso tra 7,0 e 7,4. Grandi variazioni di questo pH intracellulare possono influenzare i processi cellulari critici, come il metabolismo, il potenziale di membrana e le vie di segnalazione.
Ad esempio, la CO2 – il prodotto finale della respirazione cellulare, si combina con H2O per formare H2CO3 .
H2CO3 è un acido debole che si dissocia facilmente, acidificando lentamente il citoplasma e portando a stress cellulare.
Pertanto, le cellule regolano il loro pH interno utilizzando scambiatori di ioni per alterare la concentrazione di ioni idrogeno.
Lo scambiatore Na+/H+ aiuta a bilanciare il pH intracellulare accoppiando l'efflusso di H+ con l'afflusso di Na+.

Il pH svolge un ruolo fondamentale nel mantenimento delle normali attività cellulari. Aiuta a mantenere la struttura e la funzione di varie proteine, determina la carica sulle membrane cellulari ed è fondamentale per le reazioni metaboliche all'interno della cellula. Inoltre, le cellule utilizzano l'energia della forza motrice del protone per generare ATP.

pH citosolico

In condizioni fisiologiche, il pH citosolico è leggermente più acido del pH extracellulare. Tuttavia, le cellule devono prevenire un’ulteriore acidificazione del loro citosol per mantenere il loro potenziale di membrana e svolgere il normale funzionamento. Pertanto, impiegano diversi macchinari specializzati per la traslocazione dei protoni – antiportatori, simportatori e ATPasi di pompaggio di protoni – per regolare strettamente il pH allo stato stazionario.

Organelli cellulari e omeostasi del pH

La compartimentazione all'interno delle cellule eucariotiche aiuta a fornire condizioni ambientali distinte per svolgere funzioni specifiche all'interno di diversi organelli racchiusi da membrana. Il pH interno in questi diversi compartimenti è molto variabile, ma è un fattore determinante del loro efficace funzionamento.

Ad esempio, i citocromi situati sulla membrana mitocondriale interna emettono protoni utilizzando l'energia proveniente dal flusso di elettroni. Ciò aiuta a stabilire un gradiente protonico attraverso la membrana mitocondriale che viene utilizzato per generare energia chimica sotto forma di ATP. Allo stesso modo, i lisosomi mantengono un pH acido interno pompando protoni dal citosol utilizzando l’energia derivante dall’idrolisi dell’ATP. Questo è essenziale per il funzionamento degli enzimi lisosomiali.

Tuttavia, alcuni organelli, tra cui il nucleo, il reticolo endoplasmatico e i perossisomi, necessitano di mantenere il loro pH interno in equilibrio con il citoplasma. Pertanto, mancano di sistemi intrinseci di regolazione del pH e sono altamente permeabili ai protoni.

Disregolazione del pH intracellulare

Alcune condizioni metaboliche o genetiche possono alterare il pH stazionario del citoplasma o degli organelli all’interno della cellula e portare alla malattia. Allo stesso modo, le mutazioni nel canale CLCN5 ostacolano l’acidificazione del lume e la produzione di endosomi precoci nelle cellule epiteliali renali e possono eventualmente portare all’insufficienza renale.

Cell pH and organelle balance are essential for normal cell function. pH affects protein shape and activity, the charge on cell membranes, and many metabolic reactions inside the cell. Cells also use the proton motive force, which is the energy stored in a proton gradient, to make ATP, the cell’s main energy molecule.

The cytosol, or fluid inside the cell, is usually slightly more acidic than the fluid outside the cell. Even so, cells must stop the cytosol from becoming too acidic. They use proton-translocating machinery, including antiporters, symporters, and proton-pumping ATPases, to keep steady-state pH under control and preserve membrane potential.

Eukaryotic cells also separate functions into membrane-bound organelles with different internal pH levels. This compartmentalization creates the conditions needed for each organelle to work properly. In mitochondria, cytochromes on the inner membrane move protons using energy from electron flow. This builds the proton gradient that helps produce ATP. Lysosomes keep an acidic interior by pumping protons from the cytosol with energy from ATP hydrolysis, which is needed for lysosomal enzymes to function.

Some organelles need their internal pH to stay close to the cytoplasm. These include the nucleus, endoplasmic reticulum, and peroxisomes. They do not have their own pH-control systems and are highly permeable to protons, so their pH stays in equilibrium with the cytosol.

Problems with intracellular pH can cause disease. Changes in metabolism or gene function can disturb the steady-state pH of the cytoplasm or organelles. Mutations in chloride carriers such as CLCN6 and CLCN7, which are found on late endosomes and lysosomes, have been linked to osteoporosis and lysosomal storage disease. Mutations in the CLCN5 channel can block luminal acidification and early endosome formation in renal epithelial cells, and this can eventually lead to kidney failure.
