Peptidoglycan Synthesis

Il peptidoglicano, un componente chiave della parete cellulare batterica, è costituito da unità alternate di N-acetilglucosamina, o NAG, e di acido N-acetilmuramico o NAM unite da legami glicosidici β-1,4. La sintesi del peptidoglicano prevede tre fasi. Nel citoplasma, le molecole di uridina difosfato (UDP) sono attaccate covalentemente al NAG. UDP-NAG viene convertito enzimaticamente in UDP-NAM. Quindi, una catena di pentapeptide viene aggiunta a UDP-NAM per formare un pentapeptide UDP-NAM. Nella fase associata alla membrana, il NAM-pentapeptide è legato al bactoprenolo fosfato, formando il lipide I sul lato citoplasmatico della membrana plasmatica. L'aggiunta di un'unità NAG al Lipidio I forma il Lipidi II, che viene traslocato attraverso la membrana fino al periplasma da una flippasi. Queste catene zuccherine sono reticolate da brevi catene peptidiche, formando un polimero a rete che circonda la membrana plasmatica batterica.
Fase citoplasmatica &#8211; sintesi dei precursori
La biosintesi del peptidoglicano inizia nel citoplasma con la formazione di precursori solubili. In questa fase, la uridina difosfato (UDP), un estere dell&#8217;acido pirofosforico, si lega a NAG e NAM formando rispettivamente UDP-NAG e UDP-NAM. L&#8217;UDP-NAG viene poi convertito enzimaticamente in UDP-NAM. A quest&#8217;ultimo viene aggiunta sequenzialmente una catena pentapeptidica (tipicamente composta da L-alanina, D-acido glutammico, acido meso-diaminopimelico (o L-lisina) e due residui di D-alanina) formando UDP-NAM-pentapeptide. Questo composto funge da unit&#224; di base per le fasi successive della sintesi del peptidoglicano.
Fase associata alla membrana &#8211; trasferimento con vettore lipidico
Nella fase associata alla membrana, l&#8217;UDP-NAM-pentapeptide viene trasferito al bactoprenolo fosfato, un vettore lipidico inserito nel foglietto interno della membrana plasmatica. Questo trasferimento porta alla formazione del Lipide I. Successivamente, un&#8217;unit&#224; NAG viene aggiunta al Lipide I, generando il Lipide II, che ora contiene la struttura completa disaccaride-pentapeptide. Un enzima flippasi trasloca quindi il Lipide II attraverso la membrana, portando il precursore sul lato periplasmatico per l&#8217;incorporazione nella parete cellulare in crescita.
Fase periplasmatica &#8211; polimerizzazione e reticolazione
Nella fase finale, situata nel periplasma, le glicosiltransferasi incorporano le unit&#224; disaccaride-pentapeptide nella rete di peptidoglicano preesistente catalizzando la formazione di legami glicosidici. L&#8217;ultimo passaggio, noto come transpeptidazione, &#232; catalizzato da enzimi transpeptidasi. Questa reazione rilascia la subunit&#224; dal bactoprenolo e forma legami crociati peptidici tra peptidi stelo adiacenti, rafforzando l&#8217;integrit&#224; strutturale dello strato di peptidoglicano e completando il processo di biosintesi.

Peptidoglycan builds the bacterial cell wall and gives the cell its shape and strength. It is a mesh-like polymer around the bacterial plasma membrane. The polymer is made of repeating sugar units, N-acetylglucosamine (NAG) and N-acetylmuramic acid (NAM), linked by β-1,4 glycosidic bonds. Short peptide chains cross-link the sugar chains and make the wall more rigid.

Peptidoglycan biosynthesis starts in the cytoplasm with soluble precursors. Uridine diphosphate, or UDP, binds to NAG and NAM to form UDP-NAG and UDP-NAM. UDP-NAG is then converted into UDP-NAM by enzymes. A pentapeptide chain is added step by step to UDP-NAM. This chain usually includes L-alanine, D-glutamic acid, meso-diaminopimelic acid or L-lysine, and two D-alanine residues. The product, UDP-NAM-pentapeptide, becomes the core building block for the later stages.

Next, the pathway moves to the membrane-associated phase. UDP-NAM-pentapeptide is transferred to bactoprenol phosphate, a lipid carrier in the inner leaflet of the plasma membrane, and this forms Lipid I. A NAG unit is then added to make Lipid II. Lipid II contains the complete disaccharide-pentapeptide structure. A flippase enzyme then moves Lipid II across the membrane to the periplasmic side, where the cell wall grows.

The final stage happens in the periplasm. Glycosyltransferases add the disaccharide-pentapeptide units to the growing peptidoglycan mesh by forming glycosidic bonds. The last step is transpeptidation, which is carried out by transpeptidase enzymes. This reaction releases the subunit from bactoprenol and creates peptide cross-links between neighboring stem peptides. These links strengthen the peptidoglycan layer and complete the synthesis process.
