Oxygen Requirements and Growth Patterns

In base al fabbisogno di ossigeno, i microrganismi sono classificati in varie classi. Gli aerobi obbligati, come Mycobacteria, hanno bisogno di ossigeno, mentre gli anaerobi obbligati, come Clostridia, ne sono danneggiati e si affidano alla respirazione anaerobica per l'energia. Gli anaerobi facoltativi come E. coli passano dal metabolismo aerobico a quello anaerobico, ma crescono più velocemente in presenza di ossigeno. I microaerofili, come Helicobacter, richiedono ossigeno a concentrazioni inferiori a quelle atmosferiche. Gli anaerobi aerotolleranti, come Lactobacilli, non usano ossigeno ma possono tollerarlo. In un mezzo liquido, l'ossigeno forma un gradiente di concentrazione dall'alto verso il basso, guidando i modelli di crescita. Gli aerobi obbligati occupano la superficie, gli anaerobi si depositano sul fondo, gli anaerobi facoltativi si raggruppano vicino alla parte superiore, i microaerofili formano uno strato sotterraneo e gli organismi aerotolleranti si distribuiscono uniformemente. Durante il metabolismo dell'ossigeno, vengono prodotti sottoprodotti tossici come il perossido di idrogeno e i radicali superossido. <ul>
<li>Gli aerobi obbligati, come il Mycobacterium tuberculosis, necessitano di ossigeno per la produzione di energia, in quanto esso funge da accettore finale di elettroni nella respirazione aerobica. Questi organismi prosperano in ambienti ricchi di ossigeno.</li>
<li>Gli anaerobi obbligati, come Clostridium botulinum, sono altamente sensibili all&#8217;ossigeno poich&#233; risultano del tutto privi di enzimi in grado di neutralizzare le ROS, quali catalasi, superossido dismutasi e perossidasi, oppure li producono in quantit&#224; insufficienti. In assenza di un&#8217;adeguata difesa enzimatica, questi microrganismi non sono in grado di neutralizzare in modo efficace le specie reattive dell&#8217;ossigeno (ROS), come il perossido di idrogeno e i radicali superossido generati durante la respirazione.</li>
<li>L&#8217;Escherichia coli e altri anaerobi facoltativi sono versatili. Utilizzano l&#8217;ossigeno quando disponibile, ma passano alla fermentazione o alla respirazione anaerobica in sua assenza.</li>
<li>I microaerofili, come l&#8217;Helicobacter pylori, richiedono ossigeno a basse concentrazioni. Questi microrganismi possiedono enzimi sensibili all&#8217;O_2, rendendo meno efficiente la detossificazione delle ROS a concentrazioni di ossigeno elevato.</li>

Modelli di crescita e tecniche di coltura
In ambito laboratoristico, la crescita dei microrganismi in condizioni di ossigeno variabili &#232; spesso dimostrata utilizzando il terreno tioglicolato, un mezzo contenente agenti riducenti come il tioglicolato o la cisteina. Questi agenti esauriscono l&#8217;ossigeno disciolto riducendolo chimicamente in acqua, creando un gradiente di ossigeno nel mezzo:

<li>Gli aerobi obbligati crescono in superficie, dove la concentrazione di ossigeno &#232; massima.</li>
<li>Gli anaerobi obbligati crescono sul fondo, dove l&#8217;ossigeno &#232; assente.</li>
<li>Gli anaerobi facoltativi crescono in tutto il mezzo ma proliferano vicino alla superficie.</li>

Tossicit&#224; dell&#8217;ossigeno e meccanismi di difesa
La presenza di ossigeno porta alla formazione di ROS, che possono danneggiare componenti cellulari critici. I radicali idrossilici causano rotture e mutazioni nel DNA. Le ROS ossidano e denaturano gli enzimi, compromettendone la funzionalit&#224;. La perossidazione danneggia le membrane cellulari, compromettendone l&#8217;integrit&#224;.
Tuttavia, i microrganismi aerobi e aerotolleranti mitigano gli effetti delle ROS grazie a diversi enzimi, tra cui:
<ul>
<li>Catalasi: decompone il perossido di idrogeno in acqua e ossigeno.</li>
<li>Superossido dismutasi (SOD): converte i radicali superossido in perossido di idrogeno.</li>
<li>Perossidasi: riduce ulteriormente il perossido di idrogeno in acqua.</li>
</ul>
Negli anaerobi obbligati, l&#8217;assenza di questi enzimi li rende estremamente sensibili all&#8217;esposizione all&#8217;ossigeno.
Conclusione

Microbial oxygen needs and tolerance shape how bacteria grow and survive. Oxygen can support energy production in some microorganisms, but it can also become harmful in certain environments. These differences help explain why microbes live in very specific places.

Microorganisms are grouped by how they use oxygen. Obligate aerobes, such as Mycobacterium tuberculosis, need oxygen because it serves as the terminal electron acceptor in aerobic respiration. Obligate anaerobes, such as Clostridium botulinum, are highly sensitive to oxygen because they lack enough protective enzymes or may not have them at all.

Facultative anaerobes, such as Escherichia coli, are flexible. They use oxygen when it is present, but they can switch to fermentation or anaerobic respiration when oxygen is missing. Microaerophiles, such as Helicobacter pylori, need oxygen at low levels. Aerotolerant anaerobes, such as most Lactobacillus species, do not use oxygen, but they can survive in it.

In the lab, thioglycollate broth is often used to show these growth patterns. This medium contains reducing agents such as thioglycollate or cysteine, which remove dissolved oxygen by reducing it to water. That creates an oxygen gradient in the tube. Obligate aerobes grow near the surface, obligate anaerobes grow at the bottom, facultative anaerobes grow throughout but most near the top, microaerophiles grow just below the surface, and aerotolerant anaerobes grow evenly.

Other anaerobic culture methods also help keep oxygen out. These include gas-generating systems and sealed chambers. Scientists use these tools to study microbes that need low-oxygen or oxygen-free conditions.

Oxygen can also cause toxicity because it leads to reactive oxygen species, or ROS. These harmful molecules can damage DNA, enzymes, and cell membranes. Hydroxyl radicals can cause strand breaks and mutations, ROS can oxidize and denature enzymes, and peroxidation can damage membranes.

Many aerobic and aerotolerant microorganisms defend themselves with enzymes. Catalase breaks hydrogen peroxide into water and oxygen. Superoxide dismutase, or SOD, converts superoxide radicals into hydrogen peroxide. Peroxidase then reduces hydrogen peroxide into water. Obligate anaerobes are especially vulnerable to oxygen because they lack these defenses or make too little of them.

Understanding microbial oxygen requirements matters in medicine and industrial microbiology. It helps explain why anaerobic infections need special treatment. It also helps scientists choose the best growth conditions for production and research.
