장기강화(long-term potentiation, 줄여서 LTP; 장기 상승 작용)는 시냅스 가소성(synaptic plasticity; 화학 시냅스의 강도 변화)이 뇌에서 발생할 수 있는 방법의 하나입니다. LTP는 시냅스 전후의 신경 연결 사이에 시간이 지남에 따라 발생하는 시냅스 강화 과정입니다. LTP의 시냅스 강화는 장기저하(long-term depression, 줄여서 LTD; 장기 억압 작용)의 시냅스 약화에 반대하여 작용하며 함께 학습과 기억의 기초가 되는 주요 기작입니다.
 
 <h4>헵 LTP</h4>
 
 LTP는 시냅스이전 뉴런(presynaptic neuron)이 반복적으로 발화해 시냅스이후 뉴런(postsynaptic neuron)을 자극할 때 발생할 수 있습니다. 이것은 도널드 헵(Donald Hebb)의 1949년 논문에서 “함께 발화하는 뉴런은 같이 연결되어있다”라고 가정하기 때문에 헵 LTP(Hebbian LTP)라고도 합니다. 시냅스이전 뉴런에서 반복되는 자극은 시냅스이후 막의 이온 채널의 종류와 수에 변화를 유도합니다.
 
 LTP에는 다음과 같은 흥분성 신경전달물질(neurotransmitter)인 글루탐산(glutamate; 글루타메이트)의 두 가지 유형의 시냅스후 수용체(postsynaptic receptor)가 관여합니다: (1) NMDA 수용체 (N-methyl-D-aspartate receptor, 줄여서 NMDA receptor), (2) AMPA 수용체 (α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor, 줄여서 AMPA receptor). NMDA 수용체는 글루탐산 결합 시 열리지만, 수용체의 구멍은 일반적으로 다른 양전하 이온이 뉴런에 들어가는 것을 막는 마그네슘 이온에 의해 차단됩니다. 한편 시냅스이전 뉴런에서 방출된 글루탐산은 시냅스후 AMPA 수용체에 결합하여 막의 탈분극(depolarization)을 일으키는 나트륨 이온의 유입을 일으킬 수 있습니다. 시냅스후 막이 여러 번의 시냅스전 입력에 의해 탈분극될 때, NMDA 수용체 구멍을 막는 마그네슘 이온이 변위되어 나트륨과 칼슘 이온이 뉴런으로 흘러 들어갈 수 있습니다.
 
 칼슘 이온 유입이 증가하면 원형질막(plasma membrane)에 더 많은 AMPA 수용체가 삽입되는 신호의 연쇄반응(signaling cascade)이 시작되거나, 신호의 연쇄반응이 글루탐산 수용체를 인산화(phosphorylation)하여 더 긴 기간 동안 개방 상태를 유지하게 만들어 세포 내 양전하 이온의 전도성을 향상할 수 있습니다. 결과적으로, 동일한 양의 시냅스전 자극은 반복적으로 발화될 시 더 많은 글루탐산 수용체가 활성화되고 더 많은 양전하 이온이 시냅스이후 뉴런에 들어간다는 점에서 더 강한 시냅스후 반응을 유발합니다. 이렇게 발생하는 증폭을 시냅스 강화(synaptic strengthening) 또는 강화(potentiation)라고 합니다.
 
 “연습이 완벽을 만든다”는 격언은 LTP를 통해 부분적으로 설명될 수 있습니다. 새로운 일이 학습되고 있을 때 새로운 신경 회로가 LTP를 통해 강화됩니다. 연습을 반복할 때마다 신경 회로의 시냅스 강도가 강해지며 곧 작업이 정확하고 효율적으로 수행될 수 있게 만듭니다. 새로 강화된 연결은 시냅스전 자극이 지속되면 몇 분에서 몇 주 또는 그 이상 지속될 수 있으며, 이는 작업이 수행될 때마다 LTP가 유지된다는 것을 의미합니다.

 LTP가 정상적으로 기능할 때 쉽게 배우고 기억을 형성할 수 있습니다. 그러나 LTP의 이상은 알츠하이머병, 자폐증, 중독, 정신분열증, 다발성 경화증과 같은 많은 신경인지장애와 관련이 있습니다. LTP 뒤에 숨겨진 기작에 대한 더 나은 이해는 치료로 이어질 수 있습니다.

Long-term Potentiation

Long-term potentiation, or LTP, is a form of synaptic strengthening in the brain. It changes the strength of chemical synapses over time between pre- and postsynaptic neurons. LTP works against long-term depression, or LTD, which weakens synapses. Together, LTP and LTD are major mechanisms behind learning and memory.

LTP can happen when a presynaptic neuron fires again and again and keeps stimulating a postsynaptic neuron. This is called Hebbian LTP, based on Donald Hebb’s idea that neurons that fire together wire together. Repeated stimulation changes the types and number of ion channels in the postsynaptic membrane. Two glutamate receptors are involved: NMDA receptors and AMPA receptors.

NMDA receptors open when glutamate binds, but magnesium ions usually block the pore. At the same time, glutamate can activate AMPA receptors on the postsynaptic cell. This causes sodium ions to enter and depolarize the membrane. When many presynaptic inputs arrive in quick succession, the magnesium block is removed from the NMDA receptor. Sodium and calcium ions then enter the neuron.

The calcium influx starts a signaling cascade inside the cell. This cascade leads to more AMPA receptors being inserted into the plasma membrane. It can also phosphorylate glutamate receptors, which helps them stay open longer and lets more positively charged ions enter the cell. The same presynaptic signal then creates a stronger postsynaptic response. This increase is called synaptic strengthening, or potentiation.

LTP also helps explain why practice makes perfect. When a new task is learned, new neural circuits are reinforced by LTP. With each round of practice, the synaptic strength in those circuits becomes stronger. Over time, the task can be performed more correctly and efficiently. These strengthened connections can last from minutes to weeks or longer if presynaptic stimulation continues, so later practice can maintain the LTP.

When LTP works normally, learning and memory are easier. Problems with LTP have been linked to Alzheimer’s disease, autism, addiction, schizophrenia, and multiple sclerosis. Understanding how LTP works may help guide future therapies for these neurological and cognitive disorders.
