Blood Withdrawal I

Alle hier beschreven dierprocedures moeten worden uitgevoerd in overeenstemming met institutionele richtlijnen voor dierenetiek en goedgekeurd door IACUC. Alle procedures moeten volgen de principes van de 3R's - Vervanging, Vermindering en Verfijning - en moeten worden uitgevoerd door getraind personeel.
1. Bloedafname uit de staartvene

	<li>Bloedafname
	<ol>
		<li>Het dier wordt verdovd met een inhalatieanestheticum, zoals isofluraan, in een klokglas of gasanesthesie-inductiekamer, tot effect.</li>
		<li>Zodra het dier volledig verdoofd is, wordt het verwijderd en in laterale liggende positie geplaatst.</li>
		<li>Het oog wordt uitgerekt door een vinger op de bovenkant van het hoofd en langs de kaaklijn te plaatsen en de huid naar achteren en naar beneden te trekken.</li>
		<li>Vermijd druk op de luchtpijp uit te oefenen, omdat dit de luchtweg kan samenvallen of verstoppen, wat kan leiden tot de dood door verstikking.</li>

	<li>Bloedafname
	<ol>
		<li>De staart wordt gewassen met warm water (20-30°C) om vaatverwijding te bevorderen. GEBRUIK GEEN heet water.</li>
		<li>De staart wordt vervolgens afgeveegd met 70% ethanol om het oppervlak te reinigen en de zichtbaarheid van de ader verder te verbeteren.</li>
		<li>Een van de laterale staartaders wordt geselecteerd voor bemonstering. Het onderste deel van de staart wordt voorzichtig vastgegrepen en gestabiliseerd. Een bril-vergrooter kan worden gebruikt om de zichtbaarheid van de ader te verbeteren.</li>
		<li>Een 22G vlindernaald wordt in de laterale staartader ingebracht ongeveer halverwege vanaf het uiteinde van de staart onder een hoek van ongeveer 10°, met de naald gericht naar de basis van de staart voor herhaalde bemonstering.</li>
		<li>Bloed wordt verzameld in een vacuumboedbuis door het rubberen uiteinde van de naald in de opvangbuis te steken.</li>
		<li>Meerdere monsters kunnen uit de staart worden verkregen in overeenstemming met goedgekeurde protocollen. Voor bloedmicrosampling wordt de laterale staartader gepuncteerd met behulp van een fijne naald om een kleine, gecontroleerde hoeveelheid bloed te verkrijgen.</li>

		<li>De muis wordt voorzichtig in een gedesinfecteerde vasthoudende houder geplaatst.</li>
		<li>De vasthoudende houder wordt vastgezet om beweging tijdens de procedure te minimaliseren.</li>
		<li>De staart wordt gepositioneerd om gemakkelijk toegang tot het puntje mogelijk te maken.</li>

	</li>
	<li>Bloedafname
	<ol>
		<li>De staart wordt gereinigd met vers isopropylalcohol. Een topische verdovende crème wordt indien nodig op het staartpuntje aangebracht.</li>
		<li>Met behulp van scherpe schaar (of bij voorkeur een scalpelkling) wordt ongeveer 0,5-1 mm van het staartpuntje verwijderd.</li>
		<li>Bloed wordt direct verzameld in hepariniseerde capillaire buisjes (ongeveer 80 µL).</li>
		<li>Het verzamelde bloed wordt overgebracht met behulp van een kropje in gelabelde kalium EDTA microtainerbuisjes. De buisjes worden voorzichtig geïnverteerd (~20 keer) om coagulatie te voorkomen.</li>
	</ol>
	</li>
	<li>Hemostase en herstel
	<ol>
		<li>Bloeden wordt onder controle gehouden door voorzichtig druk uit te oefenen op het staartpuntje met behulp van steriele gaasjes totdat het bloeden stopt.</li>
		<li>Indien nodig kunnen aanvullende methoden zoals cauteriseren of styptische middelen voorzichtig worden gebruikt om hemostase te bereiken.</li>
		<li>Dieren moeten worden gemonitord om hemostase te waarborgen voor en na terugkeer naar de thuiscamer.</li>
	</ol>
	</li>

3. Hartbloedafname
<ol>
	<li>Materiaal
	<ol>
		<li>Voor een muis wordt een 3 cc spuit met een 22-25 gauge x 1" naald het beste gebruikt. Kleinere spuiten hebben niet dezelfde tegendruk en kunnen bloedafname moeilijker maken. Nadelen kleiner dan 25 gauge beperken de bloedstroom, wat leidt tot verhoogde stollingsneiging en schade aan de bloedcellen. Nadelen korter dan 1" kunnen het hartniveau niet bereiken wanneer vanaf het diafragma wordt genaderd.</li>
		<li>Voor een rat wordt een 10-12 cc spuit met een 18 gauge x 1,5" naald het beste gebruikt. Afhankelijk van de grootte van de rat, kan een kleinere spuit niet het volledige te verzamelen bloedvolume bevatten, en dus zou de spuit tijdens de procedure moeten worden vervangen. Nadelen kleiner dan 20 gauge beperken de bloedstroom, wat leidt tot verhoogde stollingsneiging. Nadelen korter dan 1,5" kunnen

Bloedafname bij dieren is een veelvoorkomende vereiste voor verschillende onderzoeksstudies. De keuze van de methode voor bloedafname bij deze dieren is afhankelijk van vele factoren, zoals het volume bloed dat nodig is, de frequentie van de bemonstering en de gezondheidsstatus van het dier dat moet worden bebloed.                
Hier zullen we deze overwegingen beoordelen en bloedafnameprocedures beschrijven, inclusief bloedafname uit de staarvene, staar snip, intracardiale bloedafname en retro-orbitale oogbloedafname. Voor andere methoden, zie de tweede video in deze serie.        
Voordat we ingaan op de bloedafnameprotocollen, laten we eerst enkele algemene overwegingen beoordelen, inclusief monstertype, naaldselectie en het maximale bloedvolume dat kan worden verzameld.  
Voordat bloed wordt afgenomen van een muis of een rat, moet het type bloedmonster dat vereist is worden bepaald. Experimentele procedures kunnen vol bloed, plasma of serum vereisen.  
Indien vol bloed wordt afgenomen, moet een anticoagulans aan het monster worden toegevoegd om stolling te voorkomen. Veelgebruikte anticoagulantia zijn heparine, natriumcitraat en ethyleendiaminetetraazijnzuur of EDTA. Anticoagulantia kunnen direct in de spuit worden geladen om de oppervlakken te bedekken.  
Dit maakt contact van het anticoagulans mogelijk zodra het bloed wordt afgenomen, wat helpt bij het voorkomen van stolsels. Omdat knaagdierbloed snel stol, is het essentieel dat de juiste verhouding van anticoagulans tot bloed wordt gebruikt.  
Plasma-verzameling vereist centrifugeren van het volledige bloed met een anticoagulans. Na het draaien is de doorzichtige vloeistof boven de WBC en bloedplaatjeslaag plasma. Het bevat fibrinogeen en andere stollingsfactoren.  
Aan de andere kant wordt serum verzameld uit een volledige bloedmonster zonder anticoagulantia. En omdat het monster is gestold, bevat het serum, dat de bovenste laag is, geen fibrinogeen of andere stollingsfactoren.  
Naaldselectie is gebaseerd op de grootte van het dier en de plaats van de venepunctie. Over het algemeen veroorzaken grote naalden minder schade aan bloedcellen en maken ze een snellere bloedafname mogelijk, maar zijn ze ook eerder geneigd om vaatbeschadiging te veroorzaken.  
De naaldlengte moet ook worden overwogen. Als een naald te lang is, kan het lastig zijn om te gebruiken, of kan het bloed beginnen te stollen terwijl het nog in de naald zit. Naaldselectie is gebaseerd op de grootte van het dier en de bemonsteringsplaats.  
Ten slotte, vanwege de kleine grootte van knaagdieren, is er een maximale hoeveelheid bloed die kan worden afgenomen uit een enkele bloedafname, wat geen ernstige schade aan het organisme zal veroorzaken. Bloedafname kan zijn zonder of met vloeistoofvervanging - meestal gedaan met 0,9% fysiologisch zoutje.  
De bovengrens in elk geval is vermeld in het onderstaande tekstprotocol. Bovendien moet voor experimenten die herhaalde bemonstering vereisen, voldoende hersteltijd worden geboden tussen de verzamelingen, en moet vloeistoofvervanging worden overwogen om de dierenwelvaart en aanvulling van bloedcellen te ondersteunen.  
Nadat we enkele algemene overwegingen hebben besproken, laten we de specifieke bloedafnametechnieken bekijken, te beginnen met de staarvenebloedafname.  
Om een bloedmonster te verkrijgen, wast u de staar met warm water bij 20 tot 30 graden Celsius, verwarmt u de staar indien nodig om de laterale staarvene te visualiseren en gebruikt u de minst noodzakelijke opwarming. Selecteer de rechter of linker staarvene voor bloedafname.  
Grijp het onderste deel van de staar voorzichtig en houd de staar recht tijdens het verzamelen van het bloedmonster. Steek de naaldpunt in een van de laterale staarvenen rond de helft vanaf het uiteinde van de staar onder een hoek van ongeveer 10 graden, bewegend naar de basis van de staar.  
Verzamel bloed in de vacuümverzamelbuis door het rubberen uiteinde van de naald in de vacuümbloedverzamelbuis in te steken.                                                    
Venenpunctie is een andere methode die microsampling van bloed mogelijk maakt. Hiervoor wordt de staarvene met een naald gepuncteerd. Zodra de vene is bereikt, wordt het bloed langzaam verzameld met een spuit of een capillairbuisje totdat het gewenste volume is verkregen.  
Een minder gebruikelijke alternatief voor de staarvenebloedafname is de staarsnip, die herhaalde bloedafname over tijd mogelijk maakt.  
Neem een anesthesieerde muis. Gebruik scherpe chirurgische schaar om ongeveer 5 tot een millimeter van het uiteinde van de staar te snippen en verzamel een klein volume perifeer bloed in geheparineerde capillairbuisjes.  
Staarsnip mag alleen worden gebruikt wanneer dit wetenschappelijk gerechtvaardigd is en wanneer minder invasieve alternatieven niet haalbaar zijn, in overeenstemming met institutionele dierenwelvaartrichtlijnen. Bewaak dieren na de bloedafname op eventuele problemen en zorg voor hen, of euthaniseer ze humaan indien nodig.                    
Voor onderzoek dat grote, niet-overlevende bloedmonsters vereist, wordt intracardiale punctie uitgevoerd. Dit is een terminale procedure en moet worden uitgevoerd onder diepe anesthesie, gevolgd door euthanasie of in overeenstemming met institutionele richtlijnen.
