- [Instructeur] Het CRISPR-Cas9-systeem is een DNA-bewerkingstool die staat voor Clustered Regularly Interspaced Short Palindromic Repeats en CRISPR-geassocieerd eiwit 9. CRISPR-Cas9 werd voor het eerst waargenomen in bacteriën en is een verdedigingsmiddel tegen virussen. Als vreemd, viraal DNA een bacterie binnendringt, wordt het verwerkt tot kleinere fragmenten, die kunnen worden ingebracht in een regio van het bacteriële genoom, een CRISPR-locus genaamd.

Wanneer het gebied wordt getranscribeerd, associeert het product met kleinere RNA's, tracrRNA's genaamd, die kunnen helpen om zowel het Cas9-eiwit als RNAse op het molecuul te oriënteren, waarvan de laatste het transcript splitst. Het eindresultaat is verschillende complexen, elk bestaande uit een Cas9-eiwit, tracrRNA en een CRISPR-RNA, afgeleid van DNA in de sprinkhaan. Het CRISPR-RNA in deze structuren herkent en begeleidt Cas9 naar viraal DNA, dat vervolgens wordt gesplitst en vernietigd.

Wetenschappers benutten CRISPR-Cas9 door individuele RNA-moleculen te synthetiseren die tracrRNA en CRISPR RNA nabootsen, die op een interessant gen kunnen richten. Wanneer bijvoorbeeld twee van dergelijke geleidende RNA's in cellen met Cas9 worden geïntroduceerd en beide op hetzelfde gen richten, kan een sequentie worden uitgesneden. Nadat dit doelgebied is verwijderd, worden de afgesneden uiteinden opnieuw verbonden en worden de effecten op de cellen waargenomen.

Het CRISPR-Cas9-systeem is dus gemodificeerd vanuit een bacterieel mechanisme en kan worden gebruikt voor een reeks technieken voor genbewerking.

Technologieën voor genoombewerking stellen wetenschappers in staat het DNA van een organisme te wijzigen door het toevoegen, verwijderen of herschikken van genetisch materiaal op specifieke genomische locaties. Dit soort technieken kunnen mogelijk worden gebruikt om genetische aandoeningen zoals hemofilie en sikkelcelanemie te genezen. Een populair en veelgebruikt onderzoeksinstrument voor DNA-bewerking dat zou kunnen leiden tot veilige en effectieve behandelingen voor genetische aandoeningen is het CRISPR-Cas9-systeem. CRISPR-Cas9 staat voor Clustered Regularly Interspaced Short Palindromic Repeats en CRISPR-associated Protein 9. Een basis CRISPR-Cas9-systeem bestaat uit een Cas9-endonuclease en een klein RNA dat Cas9 naar het doel-DNA leidt.

<h4>Oorsprong</h4>

CRISPR-sequenties werden voor het eerst waargenomen in bacteriën en later geïdentificeerd in archaea. Onderzoekers ontdekten dat het CRISPR-Cas9-systeem een adaptieve immuunverdediging biedt tegen binnendringende virussen. Veel bacteriën en de meeste archaea vangen korte sequenties van het virale DNA op om een bibliotheek van virus-DNA-segmenten of CRISPR-arrays te creëren. Wanneer de prokaryoten opnieuw worden blootgesteld aan hetzelfde virus of dezelfde klasse virussen, worden CRISPR-arrays gebruikt om kleine RNA-segmenten te transcriberen die virale indringers helpen herkennen en vervolgens viraal DNA vernietigen met Cas9 of een vergelijkbaar endonuclease.

<h4>Met behulp van CRISPR-Cas9-technologie</h4>

CRISPR-Cas9 wordt in het laboratorium vaak gebruikt om DNA te verwijderen en daarvoor in de plaats een nieuwe DNA-sequentie in te voegen. Om dit te bereiken moeten onderzoekers eerst een klein RNA-fragment maken, het gids-RNA, met een korte sequentie, de gidssequentie genaamd, die zich bindt aan een specifieke doelsequentie op genomisch DNA. Het gids-RNA kan ook associëren met Cas9 (of andere endonucleasen zoals Cpf1). Het gids-RNA en het Cas9-eiwit worden toegediend aan een cel van belang, waar het gids-RNA de doel-DNA-sequentie identificeert en Cas9 deze splitst.

De machinerie van de cel repareert vervolgens de gebroken strengen door willekeurige nucleotiden in te voegen of te verwijderen, waardoor het doelgen inactief wordt. Als alternatief kan een aangepaste DNA-sequentie samen met het gids-RNA en Cas9 in de cel worden geïntroduceerd, dat als sjabloon voor de reparatiemachines dient en de uitgesneden sequentie vervangt. Dit is een zeer effectieve manier voor onderzoekers om ‘knock-out’ te zijn. een gen om de effecten ervan te bestuderen of een gemuteerd gen te vervangen door een normaal exemplaar in de hoop een ziekte te genezen.

<h4>Ethische en haalbaarheidsoverwegingen bij mensen</h4>

CRISPR-Cas9 is a genome editing tool that lets scientists change DNA at a specific location. It can add, remove, or rearrange genetic material. Researchers study it because it may help treat genetic disorders such as hemophilia and sickle cell anemia.

CRISPR stands for Clustered Regularly Interspaced Short Palindromic Repeats, and Cas9 is a CRISPR-associated protein 9. The basic system includes a Cas9 endonuclease, which is an enzyme that cuts DNA, and a small RNA that guides Cas9 to the target sequence. Together, they make CRISPR-Cas9 a precise research tool for DNA editing.

CRISPR sequences were first seen in bacteria and later found in archaea. Scientists discovered that this system acts like an adaptive immune defense against viruses. Many bacteria and most archaea store short pieces of viral DNA in CRISPR arrays. When the same virus returns, these RNA segments help identify it, and Cas9 or a similar endonuclease destroys the viral DNA.

In the lab, CRISPR-Cas9 is often used to remove DNA and replace it with a new sequence. Researchers first design a guide RNA, which contains a guide sequence that matches a target sequence on genomic DNA. The guide RNA and Cas9 are then placed in the cell of interest. The guide RNA finds the target, Cas9 cuts the DNA, and the cell repairs the break by adding or deleting random nucleotides, which can turn the gene off.

Scientists can also add a custom DNA template with the guide RNA and Cas9. The repair machinery can use that template to replace the removed sequence. This makes CRISPR useful for gene knockouts and for replacing a mutated gene with a normal copy in hopes of treating disease. Other endonucleases, such as Cpf1, can also work with guide RNA.

CRISPR-Cas9 has also raised major ethical and safety questions, especially about embryo editing. A Chinese scientist claimed to have used CRISPR to create genome-edited babies by disabling a gene linked to HIV infection. That claim led to global concern because many scientists thought the work was premature and worried about off-target genomic effects. CRISPR has many possible biotech uses, but future challenges still need careful attention.

EXPLANATION: The utilization of new technologies in any field of science is dependent on both economic and social factors. In addition to evaluating these factors, scientists must also consider long-term consequences that may not be initially apparent. The emphasis should be on the construction of a rationale argument that supports a position on the use of an application with ethical and social impact. Students should begin to appreciate the differences in ethical values that exist, and recognize that discussion of these values is imperative as knowledge and technology continue to advance, even when resolutions of differences can be rare. After investigation of a specific biotechnology application, students can write a position paper and/or participate in a classroom debate.
