Viral Recombination

- [Instructeur] Sommige cellen kunnen tegelijkertijd geïnfecteerd zijn met twee verschillende virusstammen, waardoor een verwisseling van beide genetische sequenties ontstaat, in een proces dat virale recombinatie wordt genoemd. Meer specifiek, nadat de virussen het cytoplasma van de cel zijn binnengedrongen, worden ze gedemonteerd en beginnen ze hun genomen te repliceren. Gelijkaardige regio's van elk genoom kunnen samen binden en stukken uitwisselen, waardoor de strengen weer met elkaar verbonden worden.

Als alternatief kunnen de virussen een deel van hun segmenten ruilen in een proces dat "reassassortiment" wordt genoemd, waarbij nieuwe virussen die in de cel zijn gemaakt, een mengsel van de stammen kunnen hebben. Zoals de helft van hun segmenten van stam één en de andere helft van stam twee. Deze nieuw gevormde virussen zijn nu geëvolueerd uit de twee oorspronkelijke virussen en vormen populaties die in de loop van de tijd kunnen blijven veranderen.

Cellen worden soms door meer dan één virus tegelijk geïnfecteerd. Wanneer twee virussen uit elkaar worden gehaald om hun genomen bloot te stellen voor replicatie in dezelfde cel, kunnen vergelijkbare gebieden van hun genomen met elkaar paren en sequenties uitwisselen in een proces dat recombinatie wordt genoemd. Als alternatief kunnen virussen met gesegmenteerde genomen segmenten uitwisselen in een proces dat herschikking wordt genoemd.

<h4>Virale recombinatie kan nieuwe ziekten veroorzaken</h4>

Sommige ziekten kunnen meerdere soorten infecteren. Varkens kunnen bijvoorbeeld besmet zijn met bepaalde mens- en vogelvirussen, naast de virussen die varkens doorgaans infecteren. Omdat virussen zich kunnen recombineren wanneer ze dezelfde cel co-infecteren, kunnen varkens zich gedragen als ‘mengvaten’. die virussen van andere soorten recombineren om nieuwe virussen te creëren die soms mensen kunnen infecteren. Dit zorgwekkende fenomeen vertegenwoordigt een route waarlangs infectieus materiaal van andere soorten de menselijke bevolking kan binnendringen. Ziekten die van dier op mens overgaan, worden zoönosen genoemd. Mensen kunnen zeer vatbaar zijn voor dergelijke virussen, omdat we geen geschiedenis hebben van blootstelling die immuniteit zou hebben gegenereerd.

Influenza A is een goed voorbeeld van het ‘mengvat’. theorie van virale ziekten. Onderzoek heeft aangetoond dat varkens-, vogel- en menselijke influenza A-virussen zich opnieuw hebben gegroepeerd in de varkensgastheren. Viral gene swapping can happen when more than one virus infects the same cell. After the viruses break open and expose their genomes for replication, matching regions can pair up and exchange sequences. This process is called recombination. Viruses with segmented genomes can also swap whole segments, which is called reassortment.

These changes can help create new diseases. Some diseases can infect more than one species. Pigs can be infected by some human and bird viruses, along with the viruses that usually infect pigs. When different viruses co-infect the same pig cell, the pig can act as a mixing vessel and help produce new viruses that may infect humans.

This matters because diseases that move from animals to humans are called zoonoses. Humans can be highly susceptible to these viruses because we may not have previous exposure. Without that exposure, we would not have developed immunity. Influenza A is a clear example of the mixing vessel theory. Research has shown that pig, bird, and human influenza A viruses have reassorted inside pig hosts.

Some of those events produced double reassortant viruses with genes from human and bird viruses. Others produced triple-reassortant viruses with genes from human, bird, and pig viruses. In the Netherlands, a reassortant virus made from human and bird flu genes was passed from pigs to humans.

How Viruses Infect Cells and Multiply

Viruses infect cells and make new copies by using a host cell’s own machinery. A virus is a tiny infectious particle with an RNA or DNA genome inside a protective protein coat. It cannot reproduce on its own, so it must enter a cell to make more viruses. After infection, the virus sheds its protein coat and uses the host cell to transcribe and translate its genetic material. The cell then assembles many new viral particles. In many cases, these new viruses rupture the host cell and kill it.
