- [Instructeur] Gliale cellen zijn de ondersteunende cellen in het zenuwstelsel. Het centrale zenuwstelsel, of CZS, bevat een verscheidenheid aan glia. De grootste en meest voorkomende soort zijn astrocyten.

Deze cellen hebben vele functies, waarvan er één is om de bloed-hersenbarrière in stand te houden. Een ander soort, oligodendrocyten, met kleinere cellichamen en minder processen, vormt de myelineschede over de lengte van de axonen. De minst talrijke en kleinste cellen, microglia, zijn betrokken bij fagocytische processen, die ziekteverwekkers en cellulair puin overspoelen.

Bovendien omvat het perifere zenuwstelsel, of PNS, satellietcellen die de neuroncellichamen omgeven en functioneren als astrocyten in het CZS. Op dezelfde manier zijn schwanncellen de cellen die zich gedragen als oligodendrocyten, die helpen de omhulsels rond de perifere axonen te vormen. Naarmate het onderzoek vordert, is het duidelijk dat gliale cellen meer zijn dan alleen maar lijm in het zenuwstelsel.

Gliacellen zijn een van de twee belangrijkste soorten cellen in het zenuwstelsel. Glia-cellen omvatten astrocyten, oligodendrocyten, microglia en ependymale cellen in het centrale zenuwstelsel, en satelliet- en Schwann-cellen in het perifere zenuwstelsel. Deze cellen communiceren niet via elektrische signalen zoals neuronen dat doen, maar dragen bij aan vrijwel elk ander aspect van de werking van het zenuwstelsel. Bij mensen is het aantal gliacellen ongeveer gelijk aan het aantal neuronen in de hersenen.

<h4>Gliacellen van het centrale zenuwstelsel</h4>

Glia in het centrale zenuwstelsel (CZS) omvat astrocyten, oligodendrocyten, microglia en ependymale cellen. Astrocyten zijn het meest voorkomende type gliacellen en worden aangetroffen in georganiseerde, niet-overlappende patronen door de hersenen, waar ze nauw verbonden zijn met neuronen en haarvaten. Astrocyten spelen talloze rollen in de hersenfunctie, waaronder het reguleren van de bloedstroom en metabolische processen, synaptische ionen- en pH-homeostase, en het onderhoud van de bloed-hersenbarrière.

Een andere gespecialiseerde gliacel, de oligodendrocyt, vormt de myelineschede die neuronale axonen in het centrale zenuwstelsel omringt. Oligodendrocyten verlengen lange cellulaire processen die meerdere keren rond axonen wikkelen om deze coating te vormen. Myeline-omhulsel is vereist voor een goede geleiding van neuronale signalen en verhoogt de snelheid waarmee deze berichten zich verspreiden aanzienlijk.

Microglia – bekend als de macrofagen van het CZS – zijn het kleinste gliaceltype en zijn gespecialiseerd in fagocytose van zowel ziekteverwekkers als puin. Ze beschermen het centrale zenuwstelsel tegen infectieuze agentia en toxines en snoeien synapsen tijdens de ontwikkeling. Hoewel microglia als gliacellen worden beschouwd, hebben ze een unieke en aparte oorsprong vergeleken met andere gliale celtypen. Astrocyten en oligodendrocyten worden geproduceerd door radiale glia, terwijl microglia afkomstig zijn uit de dooierzak en vroeg in de embryonale ontwikkeling naar het embryo migreren.

Ten slotte zijn ependymcellen kubusvormige cellen met cilia-achtige uitsteeksels die de ventrikels bekleden, waar ze hersenvocht (CSF) produceren. Ependymcellen vormen een barrière tussen de hersenen en het hersenvocht en filteren potentieel schadelijke stoffen eruit. Net als astrocyten en oligodendrocyten zijn ependymcellen afkomstig van radiale glia die nabij de laterale ventrikels worden aangetroffen.

<h4>Gliacellen van het perifere zenuwstelsel</h4>

In het perifere zenuwstelsel (PNS) bestaan vergelijkbare maar verschillende typen gliacellen. Functies die worden uitgevoerd door astrocyten van het CZS worden bijvoorbeeld voornamelijk in het PZS tot stand gebracht door satellietcellen, gliacellen die structuur, demping en voedingsstoffen verschaffen aan de neuronale lichamen waarmee ze associëren. Een andere gliacel van het PZS, de Schwann-cel, functioneert als oligodendrocyten van het CZS door een myeline-omhulsel rond neuronale axonen te vormen. Net als myelinisatie in het centrale zenuwstelsel zorgt axonmyelinisatie van het PZS voor de noodzakelijke isolatie en geleidbaarheid voor de juiste overdracht van elektrische signalen.

<h4>Belang van Glia bij gezondheid en ziekte</h4>

Gliacellen zijn cruciale beschermers en toezichthouders van het zenuwstelsel. Ze handhaven niet alleen homeostatische omstandigheden en dragen bij aan de routinematige hersenfunctie, maar reageren ook op letsel aan het zenuwstelsel, infecties en ziekten. Bovendien vervullen glia cruciale functies tijdens de embryonale ontwikkeling van het zenuwstelsel. Deze cellen dragen zelfs bij aan het verwijderen van onnodige neuronale verbindingen, een proces dat synaptisch snoeien wordt genoemd. Vanwege het belang van glia in talrijke aspecten van de hersenfunctie kunnen defecten in een of meer gliale celpopulaties leiden tot ernstige en invaliderende neurologische aandoeningen, waaronder ontwikkelingsstoornissen, de ziekte van Alzheimer, de ziekte van Parkinson, multiple sclerose en vele andere. .

Tijdens de ontwikkeling vormen gliacellen een steiger voor neuronen om op de juiste manier naar hun axonen te migreren en deze uit te laten groeien. Later in het leven kunnen trauma of neurodegeneratieve ziekten leiden tot verlies van neuronale verbindingen die niet kunnen worden geregenereerd, wat kan leiden tot verminderd functioneren of verlamming.

Glial cells support the nervous system and work alongside neurons. They are one of the two main cell types in the nervous system. Unlike neurons, glia do not send electrical signals, but they help with nearly every other part of nervous system function. In humans, glial cells are about as numerous as neurons in the brain.

In the central nervous system, glia include astrocytes, oligodendrocytes, microglia, and ependymal cells. Astrocytes are the most common glial cells. They form organized, non-overlapping patterns in the brain and stay close to neurons and capillaries. They help regulate blood flow, support metabolism, maintain ion and pH balance around synapses, and help maintain the blood-brain barrier.

Oligodendrocytes make the myelin sheath in the central nervous system. This sheath wraps around neuronal axons many times and acts as insulation. Myelin helps nerve signals travel correctly and much faster. Microglia are the smallest glial cells and are known as the macrophages of the CNS. They remove pathogens and debris by phagocytosis and also help prune synapses during development. Unlike other glia, microglia come from the yolk sac and enter the embryo early in development.

Ependymal cells are cube-shaped cells with cilia-like projections that line the ventricles. They produce cerebrospinal fluid, or CSF, and form a barrier between the brain and the CSF. This barrier helps filter out harmful substances. Like astrocytes and oligodendrocytes, ependymal cells come from radial glia near the lateral ventricles.

The peripheral nervous system also has glial cells. Satellite cells support the neuronal cell bodies they surround by providing structure, cushioning, and nutrients. Schwann cells do for peripheral axons what oligodendrocytes do in the CNS. They form myelin around axons and help insulation and conductivity for electrical signaling.

Glia are important for health, development, and disease. They protect the nervous system, keep internal conditions stable, and respond to injury, infection, and disease. They also help guide neuron migration and axon growth during embryonic development. Because they help remove extra neuronal connections through synaptic pruning, problems in glial cells can contribute to developmental disorders, Alzheimer’s disease, Parkinson’s disease, multiple sclerosis, and other serious conditions. Damage later in life can also lead to lost connections that are difficult or impossible to replace, which may cause impaired function or paralysis.
