- [Verteller] Langetermijnpotentiëring, of LTP, is een proces van synaptische versterking die optreedt na verloop van tijd tussen pre- en postsynaptische neuronale verbindingen. Bij één mechanisme, wanneer presynaptische neuronen herhaaldelijk vuren en de postsynaptische cel stimuleren, leidt deze actie tot veranderingen in het type en het aantal ionenkanalen in het postsynaptische membraan, zoals een klasse van glutamaatreceptoren genaamd N-methyl-D-aspartaat, of NMDA. NMDA-receptoren worden meestal geblokkeerd door magnesiumionen.

Bij sterke depolarisatie door herhaalde stimulatie worden de magnesiumionen echter verdrongen, waardoor calciumionen kunnen binnendringen. Deze calciuminflux initieert een signaalcascade die uitmondt in een tweede klasse glutamaatreceptoren, alfa-amino-3-hydroxy-5-methyl-4-isoxazolepropionzuur, kortweg AMPA, die in het membraan wordt ingebracht. In dit geval vloeien er meer positieve ionen in het neuron, waardoor een sterke postsynaptische reactie op dezelfde presynaptische stimulatie ontstaat.

LTP is essentieel voor leren en is een van de manieren om het gezegde 'oefening baart kunst' uit te leggen omdat de nieuwe versterkte reactie kan duren van minuten tot weken of langer als de presynaptische stimulatie aanhoudt.

De synaptische versterking van LTP werkt in tegenstelling tot de synaptische verzwakking van langdurige depressie (LTD) en vormt samen de belangrijkste mechanismen die ten grondslag liggen aan leren en geheugen.

<h4>Hebbian LTP</h4>

LTP kan optreden wanneer presynaptische neuronen herhaaldelijk het postsynaptische neuron vuren en stimuleren. Dit wordt Hebbian LTP genoemd, omdat het voortvloeit uit het postulaat van Donald Hebb uit 1949 dat "neuronen die samen vuren, met elkaar verbonden zijn." De herhaalde stimulatie van presynaptische neuronen induceert veranderingen in het type en aantal ionkanalen in het postsynaptische membraan.

Bij LTP zijn twee soorten postsynaptische receptoren van de exciterende neurotransmitter glutamaat betrokken: 1) N-methyl-D-aspartaat- of NMDA-receptoren en 2) α-amino-3-hydroxy-5-methyl-4-isoxazolpropionzuur- of AMPA-receptoren . Hoewel NMDA-receptoren zich openen bij glutamaatbinding, wordt hun porie gewoonlijk geblokkeerd door magnesiumionen die voorkomen dat andere positief geladen ionen het neuron binnendringen. Glutamaat dat vrijkomt uit presynaptische neuronen kan zich echter binden aan postsynaptische AMPA-receptoren, waardoor een instroom van natriumionen ontstaat die resulteert in membraandepolarisatie. Wanneer het postsynaptische membraan wordt gedepolariseerd door meerdere frequente presynaptische inputs, wordt het magnesiumion dat de NMDA-receptorporie blokkeert, verplaatst, waardoor natrium- en calciumionen in het neuron kunnen stromen.

De verhoogde instroom van calciumionen initieert vervolgens een signaalcascade die culmineert in het inbrengen van meer AMPA-receptoren in het plasmamembraan. Als alternatief kan de signaalcascade glutamaatreceptoren fosforyleren—waardoor ze langer open kunnen blijven en de geleiding van positief geladen ionen in de cel wordt verbeterd. Als gevolg hiervan zal dezelfde presynaptische stimulatie nu een sterkere postsynaptische respons oproepen, aangezien meer glutamaatreceptoren zullen worden geactiveerd en meer positief geladen ionen het postsynaptische neuron zullen binnendringen. De versterking die optreedt, staat bekend als synaptische versterking of potentiëring.

Het adagium ‘oefening baart kunst’ kan gedeeltelijk worden verklaard door LTP. Wanneer een nieuwe taak wordt geleerd, worden nieuwe neurale circuits versterkt met behulp van LTP. Na elke herhaling van de oefening wordt de synaptische kracht in de neurale circuits sterker, en al snel kan de taak correct en efficiënt worden uitgevoerd. De nieuw versterkte verbindingen kunnen minuten tot weken of langer duren als de presynaptische stimulatie aanhoudt, wat betekent dat elke volgende keer dat de taak wordt uitgevoerd de LTP behouden blijft.

<h4>LTP en ziekte</h4>

Wanneer LTP normaal functioneert, kunnen we gemakkelijk herinneringen leren en vormen. Afwijkingen in LTP zijn echter betrokken bij veel neurologische en cognitieve stoornissen zoals de ziekte van Alzheimer, autisme, verslaving, schizofrenie en multiple sclerose. Een beter begrip van de mechanismen achter LTP zou uiteindelijk tot therapieën kunnen leiden.

Long-term potentiation, or LTP, is a form of synaptic strengthening in the brain. It changes the strength of chemical synapses over time between pre- and postsynaptic neurons. LTP works against long-term depression, or LTD, which weakens synapses. Together, LTP and LTD are major mechanisms behind learning and memory.

LTP can happen when a presynaptic neuron fires again and again and keeps stimulating a postsynaptic neuron. This is called Hebbian LTP, based on Donald Hebb’s idea that neurons that fire together wire together. Repeated stimulation changes the types and number of ion channels in the postsynaptic membrane. Two glutamate receptors are involved: NMDA receptors and AMPA receptors.

NMDA receptors open when glutamate binds, but magnesium ions usually block the pore. At the same time, glutamate can activate AMPA receptors on the postsynaptic cell. This causes sodium ions to enter and depolarize the membrane. When many presynaptic inputs arrive in quick succession, the magnesium block is removed from the NMDA receptor. Sodium and calcium ions then enter the neuron.

The calcium influx starts a signaling cascade inside the cell. This cascade leads to more AMPA receptors being inserted into the plasma membrane. It can also phosphorylate glutamate receptors, which helps them stay open longer and lets more positively charged ions enter the cell. The same presynaptic signal then creates a stronger postsynaptic response. This increase is called synaptic strengthening, or potentiation.

LTP also helps explain why practice makes perfect. When a new task is learned, new neural circuits are reinforced by LTP. With each round of practice, the synaptic strength in those circuits becomes stronger. Over time, the task can be performed more correctly and efficiently. These strengthened connections can last from minutes to weeks or longer if presynaptic stimulation continues, so later practice can maintain the LTP.

When LTP works normally, learning and memory are easier. Problems with LTP have been linked to Alzheimer’s disease, autism, addiction, schizophrenia, and multiple sclerosis. Understanding how LTP works may help guide future therapies for these neurological and cognitive disorders.
