De studie van individuele genen heeft geleid tot buitengewone inzichten over bijna alle aspecten van de biologie, van ziekte en erfelijkheid tot agricultuur en evolutie. Maar veel vragen over de functie en evolutie van genen kunnen alleen worden beantwoord door alle DNA van een organisme— zijn genoom—in zijn geheel te bestuderen. De studie van complete genomen is genomica.

Hierbij worden gehele genomen gesequencet om hun opbouw, functie, regulering en evolutie te onderzoeken. Genomica bevindt zich op de kruising van genetica, moleculaire biologie en computationele wetenschap. Nadat in 1953 de structuur van DNA was geïdentificeerd begonnen wetenschappers technieken te ontwikkelen om individuele genen te sequencen.

Een belangrijke doorbraak in het sequencen van DNA kwam toen in 1977 Frederick Sanger een methode ontwikkelde die hele genomen kon sequencen. Dit leidde tot het sequencen van genomen van biergist en de fruitvlieg met behulp van shotgun sequencing. Het menselijkgenoomproject dat in 1990 begon was een internationale inspanning van 13 jaar voor $2,7 miljard waardoor het eerste menselijke genoom in 2003 werd gesequencet.

Dit project leidde tot nieuwe sequencetechnieken en nu kunnen menselijke genomen in één dag voor minder dan $1000 gesequencet worden. De grote hoeveelheid data heeft geleid tot de opkomst van het veld van bioinformatica waar mathematische en computerwetenschappelijke technieken worden gecombineerd om de enorme hoeveelheden genomisch data te organiseren, analyseren en vergelijken. Genomica is nu een centraal bestanddeel van gepersonaliseerde geneeskunde, bijvoorbeeld voor de behandeling van kanker, en het blijft een sleutelrol spelen in de antropologie, evolutie, forensische wetenschap, agricultuur en vele andere velden.

Genomics

Genomica is de wetenschap van genomen: het is de studie van al het genetische materiaal van een organisme. Bij mensen bestaat het genoom uit informatie die wordt vervoerd in 23 paar chromosomen in de kern, evenals uit mitochondriaal DNA. In de genomica wordt zowel coderend als niet-coderend DNA gesequenced en geanalyseerd. Genomica maakt een beter begrip mogelijk van alle levende wezens, hun evolutie en hun diversiteit. Het heeft talloze toepassingen: bijvoorbeeld om fylogenetische bomen te bouwen, de productiviteit en duurzaamheid van gewassen te verbeteren, strafrechtelijk onderzoek te ondersteunen, genen te identificeren die relatie houden met ziekten, of zich te richten op een optimale behandeling van kankerpatiënten.

De afgelopen dertig jaar hebben er belangrijke technologische ontwikkelingen op het gebied van DNA-sequencing plaatsgevonden, vooral dankzij de inspanningen van het internationale Human Genome Project. Er bestaat nu technologie om 60 miljard DNA-basen te sequencen, wat twintig keer zo groot is als een menselijk genoom. Met deze overvloed aan informatie en big data werken computerwetenschappers en bio-informatici hand in hand met biologen om de DNA-sequentieresultaten te verzamelen, op te slaan en te analyseren, terwijl de gegevens veilig toegankelijk zijn.

Het succes van genomics heeft de weg geopend voor andere grootschalige methoden om biologische systemen in hun geheel te onderzoeken. Genomische studies kunnen nu worden voltooid door andere 'omics'-onderzoekers. studies om gegevens te verzamelen over levende wezens op alle niveaus: soorten, populaties, individuen, cellen, eiwitten, RNA, DNA. Transcriptomics is de studie van hoe de algehele expressie van genen varieert onder verschillende experimentele of pathologische omstandigheden. Proteomics is de studie van alle eiwitten waaruit een organisme of systeem bestaat. Metabolomics is de studie van alle metabolieten (suiker, vetten, andere moleculen) in een bepaald biologisch systeem en hun interacties. Onlosmakelijk verbonden met bio-informatica, zijn de 'omics'-systemen maken een globaal beeld mogelijk van complexe leefomgevingen in hun omgeving, en maken als zodanig de weg vrij voor gepersonaliseerde geneeskunde.

Genomics studies the full set of genetic material in an organism. In humans, that genome includes the DNA carried in 23 pairs of chromosomes in the nucleus, along with mitochondrial DNA. Genomics looks at both coding DNA and non-coding DNA, which helps scientists study how living things are built and how they differ.

This field is useful in many areas of biology and medicine. Scientists use genomics to build phylogenetic trees, improve crop productivity and sustainability, help with criminal investigations, find genes linked to disease, and support better cancer treatment choices. It also gives researchers a clearer view of evolution and biological diversity.

DNA sequencing has changed genomics over the past 30 years. Major progress came from the international Human Genome Project, which helped drive faster and more powerful sequencing methods. Today, technology can sequence about 60 billion DNA bases, or around 20 times the size of a human genome.

That amount of data requires bioinformatics, the use of computers to manage and study biological information. Computer scientists and bioinformaticians work with biologists to collect, store, analyze, and secure DNA sequencing results. Their teamwork makes it possible to handle large-scale genomic data.

Genomics also led to other large-scale &quot;omics&quot; studies. Transcriptomics examines how gene expression changes under different experimental or disease conditions. Proteomics studies all the proteins in an organism or system, and metabolomics studies the metabolites, such as sugars, fats, and other molecules, and how they interact.

Together, these omics fields give a broad view of living systems at the level of species, populations, individuals, cells, proteins, RNA, and DNA. Because they rely on bioinformatics, they help scientists understand complex biology in context. They also support the growth of personalized medicine.
