Enzymes

Enzymen zijn biologische katalysatoren die de snelheid van biochemische reacties versnellen. De meeste enzymen zijn eiwitten, opgebouwd uit aminozuren;terwijl sommige RNA-moleculen zijn die bekend staan als ribozymen. Enzymen werken door de activeringsenergie van een reactie te verlagen, waardoor de reactiesnelheid toeneemt.Ze kunnen zowel de synthese als de ontleding van chemische bindingen katalyseren, maar hebben geen invloed op de richting of het evenwicht van de reactie. Elk enzym bindt zich aan een specifieke reactant, het substraat genaamd, en katalyseert een bepaalde reactie. Het substraat bindt zich aan een apart katalytisch gebied van het enzym dat de actieve centrum wordt genoemd door interacties zoals intermoleculaire krachten en voorbijgaande covalente bindingen, wat resulteert in een enzym-substraatcomplex.Deze binding is zeer specifiek vanwege de conformationele complementariteit die vereist is tussen het enzym en zijn substraat. In levende organismen fungeren enzymen als katalysatoren voor veel biochemische reacties die betrokken zijn bij het cellulaire metabolisme. De rol van enzymen is het verminderen van de activeringsenergieën van biochemische reacties door complexen te vormen met de substraten ervan. Het verlagen van de activeringsenergieën bevordert een toename van de snelheid van biochemische reacties. Enzymtekorten kunnen zich vaak vertalen in levensbedreigende ziekten. Een genetische afwijking die resulteert in een tekort aan het enzym G6PD (glucose-6-fosfaatdehydrogenase) heeft bijvoorbeeld een nadelige invloed op de metabolische route die NADPH aan cellen levert. Een verstoring van deze metabolische route kan het glutathion in de rode bloedcellen verminderen, waardoor schade aan andere enzymen en eiwitten zoals hemoglobine ontstaat. De overmatige metabolisatie van hemoglobine verhoogt het bilirubineniveau, wat leidt tot geelzucht, een aandoening die ernstig kan worden. Daarom moeten mensen die lijden aan een G6PD-tekort bepaalde voedingsmiddelen en medicijnen vermijden die chemicaliën bevatten die schade aan hun glutathion-deficiënte rode bloedcellen kunnen veroorzaken. Enzymfunctie en structuur Enzymen zijn gegroepeerd in verschillende klassen op basis van de specifieke functie die ze vervullen. Oxoreductasen zijn bijvoorbeeld betrokken bij redoxreacties, terwijl transferasen de overdracht van functionele groepen katalyseren. Voor de vorming van bindingen met ATP-hydrolyse zijn ligasen nodig, terwijl hydrolysereacties en de vorming van dubbele bindingen worden gekatalyseerd door respectievelijk hydrolasen en lyasen. Isomerase-enzymen katalyseren gewoonlijk isomerisatiereacties. Enzymen bezitten over het algemeen actieve sites. Dit zijn specifieke gebieden op het molecuul met een conformatie die ervoor zorgt dat het enzym zich aan een specifiek substraat (een reactantmolecuul) bindt om een enzym-substraatcomplex of het reactietussenproduct te vormen. Twee modellen – het lock-and-key-model en het geïnduceerde-fit-model – proberen de werking van een actieve site te verklaren (Figuur 1). De meest simplistische slot-en-sleutelhypothese suggereert dat de actieve plaats en de moleculaire vorm van het substraat complementair zijn en in elkaar passen als een sleutel in een slot (Figuur 1a). Aan de andere kant suggereert de geïnduceerde-fit-hypothese dat het enzymmolecuul flexibel is en van vorm verandert om een binding met het substraat mogelijk te maken (Figuur 1b). Enzym-remmers De activiteit van enzymen kan ook worden onderbroken door het proces van enzymremming. Er zijn verschillende veel voorkomende vormen van enzymremming. Tijdens competitieve remming bindt een ander molecuul (natuurlijk of synthetisch) dan het substraat rechtstreeks aan de actieve plaats van het enzym. De structurele en chemische gelijkenis van de remmer met het substraat vergemakkelijkt de binding ervan aan de actieve plaats. Dergelijke competitieve remmers concurreren dus met substraten en voorkomen dat ze aan het enzym binden. Meestal kan een toenemende substraatconcentratie de effecten van competitieve remming onderdrukken. Bij niet-competitieve remming bindt een molecuul (natuurlijk of synthetisch) aan een allosterisch (ander) gebied van het enzym, anders dan de actieve plaats ervan. De remmerbinding veroorzaakt een conformationele verandering in de actieve plaats van het enzym, wat resulteert in een afname van het vermogen van het enzym om de reactie te katalyseren. In tegenstelling tot competitieve remming vermindert een verhoging van de substraatconcentratie de remmende effecten van niet-competitieve remming niet.

Enzymes are biological catalysts that speed up many reactions in living organisms. They help control cellular metabolism by lowering the activation energy needed for biochemical reactions. When activation energy drops, reaction rates can increase.

Enzyme problems can also lead to serious disease. A genetic abnormality that causes G6PD, or glucose-6-phosphate dehydrogenase, deficiency can disrupt the pathway that supplies NADPH to cells. In red blood cells, this can reduce glutathione and damage other enzymes and proteins such as hemoglobin. As hemoglobin is broken down too much, bilirubin builds up and can cause jaundice. People with G6PD deficiency often need to avoid certain foods and medicines that may trigger damage to their red blood cells.

Enzymes are grouped by the reactions they carry out. Oxidoreductases take part in redox reactions. Transferases move functional groups from one molecule to another. Ligases form bonds and use ATP hydrolysis, while hydrolases drive hydrolysis reactions and lyases help form double bonds. Isomerases catalyze isomerization reactions.

Most enzymes have an active site, which is the region that binds a specific substrate, or reactant molecule. The enzyme and substrate form an enzyme-substrate complex, which acts as the reaction intermediate. Two models explain how this binding works. In the lock-and-key model, the active site and substrate fit together like a key in a lock. In the induced-fit model, the enzyme changes shape to fit the substrate. Both models explain enzyme specificity, meaning an enzyme usually binds only certain substrates and catalyzes only one particular reaction.

Enzyme activity can also be blocked by inhibitors. In competitive inhibition, an inhibitor that is similar to the substrate binds directly to the active site and competes with the substrate. Increasing substrate concentration can often reduce this effect. In non-competitive inhibition, the inhibitor binds to an allosteric site, which is a different region of the enzyme. This binding changes the shape of the active site and lowers enzyme activity. Unlike competitive inhibition, adding more substrate does not fix non-competitive inhibition.
