Stereo-isomeren van coördinatieverbindingen zijn moleculen met dezelfde chemische formule en dezelfde connectiviteit van de liganden met het metaalatoom, maar verschillen in de rangschikking van de ligandmoleculen rond het centrale atoom. Op basis van de moleculaire structuur worden stereo-isomeren van coördinatieverbindingen geclassificeerd als geometrische isomeren of optische isomeren. Geometrische isomeren zijn stereo-isomeren met volledig verschillende moleculaire geometrieën en met verschillende fysische en chemische eigenschappen.Cis-trans-isomeren zijn een voorbeeld van geometrische isomeren. Ze komen voor in vierkant-vlakke complexen met twee sets van twee identieke liganden, of MA₂B₂, en in octaëdrische complexen met een set van vier identieke liganden en een paar van een andere ligand, of MA₄B₂. In het cis-isomeer van het vierkant-vlakke complex diamminedichloorplatina(II)bekend als cisplatina, bevinden de chlorideliganden zich bijvoorbeeld naast elkaar en aan dezelfde kant van het molecuul.In het trans-isomeer, transplatina, bevinden de chlorideliganden zich aan de tegenoverliggende zijden van het molecuul. Octaëdrische complexen met twee sets van drie identieke liganden, of MA₃B₃, kunnen een ander type geometrische isomerie vertonen, bekend als fac-mer isomerie. In de fac-isomeer van triamminetrichlorokobalt(III)bevinden de drie amineliganden en drie chloride-liganden zich bijvoorbeeld aan weerszijden van het molecuul en vormen ze dus tegenoverliggende zijden van de octaëder.In het mer-isomeer vormen drie van dezelfde liganden een boog rond het centrale metaalatoom. De andere klasse stereo-isomeren wordt gedefinieerd als optische isomeren. Net als onze handen zijn optische isomeren spiegelbeelden van elkaar en kunnen ze niet over elkaar heen worden gelegd.Deze isomeren, ook wel enantiomeren genoemd, worden chiraal genoemd en vertonen weinig verschillen in hun fysische en chemische eigenschappen. Optische isomeren onderscheiden zich door hun interactie met vlak gepolariseerd licht. Gepolariseerd licht heeft elektrische veldvectoren die oscilleren in een enkel vlak, dat een bepaalde hoeveelheid wordt geroteerd wanneer het gepolariseerde licht door een oplossing van een enantiomeer gaat.Een enantiomeer van tris(ethyleendiamine)kobalt(III)roteert bijvoorbeeld het vlak naar rechts en wordt rechtsdraaiend of het d optische isomeer genoemd. Het andere enantiomeer roteert het vlak naar links en wordt linksdraaiend of het l optische isomeer genoemd.

<h3>Isomerie in complexen</h3> Isomeren zijn verschillende chemische soorten met dezelfde chemische formule. Overgangsmetaalcomplexen bestaan vaak als geometrische isomeren, waarin dezelfde atomen verbonden zijn via dezelfde soorten bindingen, maar met verschillen in hun oriëntatie in de ruimte. Coördinatiecomplexen met twee verschillende liganden in de cis- en trans-posities van een benodigd ligand, vormen isomeren. Het octaëdrische [Co(NH3)4Cl2]+-ion heeft bijvoorbeeld twee isomeren (Figuur 1). In de cis-configuratie grenzen de twee chlorideliganden aan elkaar. Het andere isomeer, de trans-configuratie, heeft de twee chlorideliganden direct tegenover elkaar. <img alt="Image1" src="/files/ftp_upload/11459/11459_Image1.svg" style="text-align: center; height: 200px;"> Figuur 1. De cis- en trans-isomeren van [Co(H2O)4Cl2]+ bevatten dezelfde liganden die aan hetzelfde metaalion zijn gebonden, maar de ruimtelijke ordening zorgt ervoor dat deze twee verbindingen zeer verschillende eigenschappen hebben. Verschillende geometrische isomeren van een stof zijn verschillende chemische verbindingen. Ze vertonen verschillende eigenschappen, ook al hebben ze dezelfde formule. De twee isomeren van [Co(NH3)4Cl2]NO3 verschillen bijvoorbeeld in kleur; de cis-vorm is violet en de trans-vorm is groen. Bovendien hebben deze isomeren verschillende dipoolmomenten, oplosbaarheden en reactiviteiten. Als voorbeeld van hoe de opstelling in de ruimte de moleculaire eigenschappen kan beïnvloeden, beschouwen we de polariteit van de twee [Co(NH3)4Cl2]NO3-isomeren. Bedenk dat de polariteit van een molecuul of ion wordt bepaald door de bindingsdipolen (die te wijten zijn aan het verschil in elektronegativiteit van de bindende atomen) en hun rangschikking in de ruimte. In het ene isomeer veroorzaken cis-chlorideliganden meer elektronendichtheid aan de ene kant van het molecuul dan aan de andere kant, waardoor het polair wordt. Voor het trans-isomeer ligt elk ligand direct tegenover een identiek ligand, dus de dipolen van de binding heffen elkaar op en het molecuul is niet-polair. Een ander belangrijk type isomeren zijn optische isomeren, of enantiomeren, waarbij twee objecten exacte spiegelbeelden van elkaar zijn, maar niet zo kunnen worden uitgelijnd dat alle onderdelen bij elkaar passen. Dit betekent dat optische isomeren niet-superponeerbare spiegelbeelden zijn. Een klassiek voorbeeld hiervan is een paar handen, waarbij de rechter- en linkerhand spiegelbeelden van elkaar zijn, maar niet over elkaar heen kunnen worden gelegd. Optische isomeren zijn erg belangrijk in de organische en biochemie, omdat levende systemen vaak het ene specifieke optische isomeer bevatten en niet het andere. In tegenstelling tot geometrische isomeren hebben paren optische isomeren vrijwel identieke eigenschappen (kookpunt, polariteit, oplosbaarheid, etc.). Optische isomeren verschillen alleen in de manier waarop ze gepolariseerd licht beïnvloeden en hoe ze reageren met andere optische isomeren. Voor coördinatiecomplexen vormen veel coördinatieverbindingen zoals [M(en)3]n+ [waarin Mn+ een centraal metaalion is zoals ijzer(III) of kobalt(II)] enantiomeren, zoals weergegeven in figuur 2. Deze twee isomeren zullen anders reageren met andere optische isomeren. DNA-helices zijn bijvoorbeeld optische isomeren, en de vorm die in de natuur voorkomt (rechtshandig DNA) zal slechts aan één isomeer van [M(en)3]n+ binden en niet aan de andere. <img alt="Image2" src="/files/ftp_upload/11459/11459_Image_2.svg" style="text-align: center; height: 200px;"> Figuur 2. Isomer shapes in coordination complexes can change a compound’s properties even when the chemical formula stays the same. Isomers are different chemical species with the same formula. In transition metal complexes, the atoms stay connected through the same types of bonds, but their arrangement in space can differ.

One common form is geometric isomerism. In these complexes, ligands (the ions or molecules attached to the metal) can sit in cis or trans positions. The octahedral [Co(NH3)4Cl2]+ ion has two geometric isomers. In the cis form, the two chloride ligands are next to each other. In the trans form, the chlorides are directly across from one another.

These geometric isomers can behave very differently. The two isomers of [Co(NH3)4Cl2]NO3 have different colors. The cis form is violet, and the trans form is green. They also differ in dipole moment, solubility, and reactivity. Polarity depends on bond dipoles and how they are arranged in space. In the cis isomer, the chloride ligands create more electron density on one side of the ion, so it is polar. In the trans isomer, matching ligands are opposite each other, so the dipoles cancel and the ion is nonpolar.

Another important category is optical isomerism, also called enantiomerism. Enantiomers are mirror images that cannot be superposed, like right and left hands. These optical isomers are important in organic chemistry and biochemistry because living systems often use one optical isomer but not the other. Unlike geometric isomers, enantiomers usually have nearly identical boiling points, polarity, and solubility. They differ in how they affect polarized light and how they react with other optical isomers.

Many coordination compounds also form enantiomers. A complex such as [M(en)3]n+, where M is a metal ion like iron(III) or cobalt(II) and en is ethylenediamine, can exist as a pair of non-superposable mirror images. These forms react differently with other optical isomers. For example, DNA helices are optical isomers, and natural right-handed DNA binds to only one isomer of [M(en)3]n+.

Some complexes show both geometric and optical isomerism. The [Co(en)2Cl2]+ ion has cis and trans forms. Its trans isomer has the chlorines 180° apart and has very different properties from the cis form. The cis isomer also exists as a pair of optical isomers. These mirror-image forms behave the same in most ways, except when they react with other enantiomers.
