Structural Protein Function

Structural proteins are a class of proteins that support cell shape and movement. Outside of the cell, these proteins are present in structural body parts, such as bones, cartilage, and skin.&#160;
Collagen, the most abundant protein in mammals, is a structural protein found throughout the body in the extracellular matrix.&#160;
This protein winds into a triple helix that polymerizes into elongated fibrils, which then further assemble into larger fibers. Among its many structural functions, it contributes to cell and tissue shape.
The cytoskeleton is predominantly composed of structural proteins and forms the basic framework of a cell. It contains three main types of filaments: microfilaments, intermediate filaments, and microtubules.
Actin is a structural protein that makes up the microfilaments. This protein self-polymerizes into filaments and contributes to cell shape, organization, movement, and division.
Intermediate filaments can be composed of different proteins depending on the cell type. For example, keratin intermediate filaments are found in epithelial cells, whereas peripherin intermediate filaments are found in the peripheral neurons. The primary structural function of these filaments is to reinforce cells and organize them into tissues.&#160;
The microtubules consist of a class of structural proteins called tubulins. Tubulins self-assemble to form microtubules that contribute to the organization of the cytoplasm including the location of the organelles.&#160;
Structural proteins are necessary for the health of an organism, and mutations in the genes that code for these proteins can result in various diseases, including skeletal and cardiovascular abnormalities.

Structurele eiwitten zijn een categorie eiwitten die verantwoordelijk zijn voor functies variërend van celvorm en beweging tot het bieden van ondersteuning aan belangrijke structuren zoals botten, kraakbeen, haar en spieren. Deze groep omvat eiwitten zoals collageen, actine, myosine en keratine.

Collageen, het meest voorkomende eiwit bij zoogdieren, wordt door het hele lichaam aangetroffen. In bindweefsel, zoals de huid, ligamenten en pezen, zorgt het voor treksterkte en elasticiteit. In botten en tanden mineraliseert het om harde weefsels te vormen en draagt het bij aan het draagvermogen ervan. Naast structurele ondersteuning kan collageen ook interageren met celoppervlakreceptoren en andere tussenliggende moleculen om cellulaire processen te reguleren, zoals groei en migratie, die veranderingen in de cel- en weefselvorm met zich meebrengen.

Structurele eiwitten vormen het basisraamwerk van het celcytoskelet. Het cytoskelet bestaat uit drie soorten filamenten: microfilamenten, intermediaire filamenten en microtubuli, en elk is samengesteld uit verschillende structurele eiwitten. Het microfilament wordt gevormd wanneer actine zelfpolymeriseert tot lange, repetitieve structuren. Deze actinefilamenten dragen bij aan de celvorm en organisatie; bovendien kunnen microfilamenten ook bijdragen aan celbeweging en -deling, wanneer het samenwerkt met myosine. De samenstelling van tussenliggende filamenten varieert afhankelijk van het celtype. Er zijn ongeveer 70 verschillende genen die coderen voor verschillende tussenliggende filamenten. Tussenliggende filamenten in epitheelcellen bevatten keratine, perifere neuronen bevatten periferine en de sarcomeer in spiercellen bevat desmine. De primaire structurele functie van deze filamenten is het versterken van cellen en het organiseren ervan in weefsels. Microtubuli bestaan uit structurele eiwitten die tubulinen worden genoemd. Tubulinen assembleren zichzelf en vormen microtubuli die bijdragen aan de organisatie van het cytoplasma, inclusief de locatie van de organellen. Microtubuli zijn ook essentieel voor mitose en celdeling.

Omdat structurele eiwitten wijdverspreid zijn, kan een mutatie in een gen dat codeert voor een van deze eiwitten ernstige schadelijke gevolgen hebben. Een mutatie in een gen dat codeert voor collageen kan bijvoorbeeld resulteren in een aandoening die bekend staat als osteogenesis imperfecta, die wordt gekenmerkt door zwakke botten en misvormingen in bindweefsel. Verschillende mutaties in een collageengen kunnen resulteren in het Alport-syndroom, dat wordt gekenmerkt door problemen in organen zoals nieren, ogen en oren.
