Structure of Lipids

Lipiden zijn een groep hydrofobe moleculen die triglyceriden bevatten, die worden gebruikt om energie op te slaan, en fosfolipiden, die de belangrijkste structurele componenten van celmembranen zijn. Triglyceriden en de meeste fosfolipiden bestaan uit glycerol-en vetzuurketens. Vetzuren hebben methylgroepen aan het ene uiteinde van de keten en carboxylgroepen aan het andere uiteinde.De koolstof die met de carboxylgroep is verbonden, wordt de alfakoolstof genoemd en de koolstof uit de methylgroep staat bekend als de omega-koolstof. Vetzuren variëren in lengte en de aanwezigheid van dubbele bindingen in de koolwaterstofketen. Die met dubbele bindingen zijn onverzadigde vetzuren en die met alleen enkele bindingen zijn verzadigde vetzuren omdat ze verzadigd zijn met het grootst mogelijke aantal waterstofatomen.Vetzuren hebben vaak gewone namen, maar kunnen systematisch worden benoemd op basis van het aantal koolstofatomen en het aantal en de positie van de dubbele bindingen in de koolstofketen. Er zijn verschillende veelgebruikte nummeringsmethoden. Het carboxyl-referentiesysteem telt de positie van alle dubbele bindingen van de carboxylkoolstof, die is genummerd als 1.Het omega-referentiesysteem telt de positie van de dubbele binding die zich het dichtst bij de omega-koolstof bevindt, waarbij de omega-koolstof genummerd is als 1. Linoleenzuur, een omega 3-vetzuur, heeft bijvoorbeeld 18 koolstofatomen en dubbele bindingen op de posities 9, 12 en 15 vanaf het carboxyluiteinde. Dit wordt 18:3 Δ9, 12, 15 genoemd volgens het carboxyl-referentiesysteem en 18:3 volgens het omega-referentiesysteem.Onverzadigde vetzuren kunnen in twee configuraties voorkomen:cis en trans. In de cis-configuratie bevinden de waterstofatomen op de koolstofatomen die betrokken zijn bij de dubbele binding zich aan dezelfde kant van de binding, terwijl in de trans-configuratie de waterstofatomen zich aan de tegenoverliggende zijden bevinden. Structureel hebben cis-vetzuren gebogen ketens, terwijl transvetzuren rechte ketens hebben.De consumptie van transvetten blijkt verantwoordelijk te zijn voor verschillende hart-en vaatziekten, terwijl de consumptie van cis-vetzuren, zoals omega-3 en omega-6, goed is voor de gezondheid. Triglyceriden, algemeen bekend als vetten, worden voornamelijk gebruikt voor de opslag van energie. Ze zijn samengesteld uit drie vetzuren die aan glycerol zijn gekoppeld via esterbindingen tussen de hydroxyluiteinden van glycerol en de carboxyluiteinden van de vetzuren, wat resulteert in de vorming van een niet-polair molecuul.Als alle drie de vetzuren van hetzelfde type zijn, staan ze bekend als eenvoudige triglyceriden. Als de drie vetzuren verschillen, staan ze bekend als gemengde triglyceriden. Een ander type lipide, fosfolipiden, is een belangrijk structureel kenmerk van biologische membranen.Ze hebben hydrofiele koppen bestaande uit fosfaatgroepen gemodificeerd met een hydrofiele alcoholgroep en hydrofobe vetzuurstaarten. Glycerofosfolipiden zijn een veelgebruikt type fosfolipide dat bestaat uit twee vetzuren en een sterk polaire groep die aan elk koolstofatoom van glycerol is gehecht via respectievelijk ester-en fosfodiësterbindingen.

Lipiden omvatten een diverse groep verbindingen die grotendeels niet-polair van aard zijn. Dit komt omdat het koolwaterstoffen zijn die voornamelijk niet-polaire koolstof-koolstof- of koolstof-waterstofbindingen bevatten. Niet-polaire moleculen zijn hydrofoob (“watervrezend”) of onoplosbaar in water. Lipiden vervullen veel verschillende functies in een cel. Cellen slaan energie op voor langdurig gebruik in de vorm van vetten. Lipiden zorgen ook voor isolatie van de omgeving voor planten en dieren. Ze helpen bijvoorbeeld watervogels en zoogdieren droog te houden wanneer ze een beschermende laag over vacht of veren vormen vanwege hun waterafstotende, hydrofobe karakter. Lipiden zijn ook de bouwstenen van veel hormonen en vormen een belangrijk bestanddeel van alle celmembranen. Lipiden omvatten vetten, oliën, wassen, fosfolipiden en steroïden. 

 Algemene structuur van lipiden 

 Een vetmolecuul bestaat uit twee hoofdcomponenten: glycerol en vetzuren. Glycerol is een organische verbinding (alcohol) met drie koolstofatomen, vijf waterstofatomen en drie hydroxylgroepen (OH). Vetzuren hebben een lange keten van koolwaterstoffen waaraan een carboxylgroep is bevestigd, vandaar de naam ‘vetzuur’. Het aantal koolstofatomen in het vetzuur kan variëren van 4 tot 36. De meest voorkomende zijn die met 12-18 koolstofatomen. In een vetmolecuul hechten de vetzuren zich aan elk van de drie koolstofatomen van het glycerolmolecuul met een esterbinding via een zuurstofatoom. Door drie vetzuren aan een glycerolskelet te verbinden in een dehydratatiereactie, ontstaat triacylglycerol. De drie vetzuren in de triacylglycerol kunnen vergelijkbaar of verschillend zijn. 

 Een fosfolipide is een ander veel voorkomend type lipide. Het is een amfipatisch molecuul, wat betekent dat het een hydrofoob en een hydrofiel deel heeft. De vetzuurketens zijn hydrofoob en kunnen geen interactie aangaan met water; terwijl de fosfaathoudende groep hydrofiel is en een interactie aangaat met water. De hydrofiele hoofdgroepen van de fosfolipiden zijn naar de waterige oplossing gericht. De hydrofobe staarten worden in het midden van de dubbellaag afgezonderd. 

 Vetzuren 

 Vetzuren kunnen verzadigd of onverzadigd zijn. Als er in een vetzuurketen slechts enkele bindingen zijn tussen aangrenzende koolstofatomen in de koolwaterstofketen, is het vetzuur verzadigd. Stearinezuur is een voorbeeld van een verzadigd vetzuur. 

 Wanneer de koolwaterstofketen een dubbele binding bevat, is het vetzuur onverzadigd. Oliezuur is een voorbeeld van een onverzadigd vetzuur. De meeste onverzadigde vetten zijn vloeibaar bij kamertemperatuur en worden oliën genoemd. Als er één dubbele binding in het molecuul zit, dan is het een enkelvoudig onverzadigd vet (bijvoorbeeld olijfolie), en als er meer dan één dubbele binding is, dan is het een meervoudig onverzadigd vet (bijvoorbeeld koolzaadolie). Lange rechte vetzuren met enkele bindingen zijn over het algemeen stevig verpakt en zijn vast bij kamertemperatuur. Dierlijke vetten met stearinezuur en palmitinezuur (veel voorkomend in vlees) en het vet met boterzuur (veel voorkomend in boter) zijn voorbeelden van verzadigde vetten. 

 Vetzuren kunnen bovendien worden ingedeeld in Cis en trans. Cis en trans geven de configuratie van het molecuul rond de dubbele binding aan. Als waterstofatomen in hetzelfde vlak aanwezig zijn, is het een cis-vet. Als de waterstofatomen zich op twee verschillende vlakken bevinden, is er sprake van een transvet. De dubbele cis-binding veroorzaakt een buiging of een “knik” waardoor de vetzuren niet goed kunnen worden opgepakt, waardoor ze bij kamertemperatuur vloeibaar blijven. Olijfolie, maïsolie, koolzaadolie en levertraan zijn voorbeelden van onverzadigde vetten. Onverzadigde vetten helpen het cholesterolgehalte in het bloed te verlagen; terwijl verzadigde vetten bijdragen aan de vorming van plaques in de slagaders. 

 Transvetten 

 De voedingsindustrie maakt oliën kunstmatig waterstof om ze halfvast te maken en een consistentie te krijgen die wenselijk is voor veel bewerkte voedingsproducten. Tijdens dit proces kunnen dubbele bindingen van de cis–conformatie in de koolwaterstofketen worden omgezet in dubbele bindingen in de trans–conformatie. 

 Margarine, sommige soorten pindakaas en bakvet zijn voorbeelden van kunstmatig gehydrogeneerde transvetten. Recente onderzoeken hebben aangetoond dat een toename van transvetten in de menselijke voeding kan leiden tot hogere niveaus van lipoproteïnen met lage dichtheid (LDL), of ‘slechte’ cholesterol, wat op zijn beurt kan leiden tot afzetting van plaques in de bloedvaten, wat kan leiden tot hartziekten. 

 Omega-vetzuren 

 Essentiële vetzuren zijn de vetzuren die het menselijk lichaam nodig heeft, maar niet zelf synthetiseert. Daarom moeten ze worden aangevuld via inname via de voeding. Omega-3-vetzuren vallen in deze categorie en zijn een van de slechts twee die bij de mens bekend zijn (de andere is omega-6-vetzuur). Dit zijn meervoudig onverzadigde vetzuren en zijn omega-3 omdat een dubbele binding de derde koolstof vanaf het uiteinde van de koolwaterstofketen verbindt met de aangrenzende koolstof. 

 Alfa-linoleenzuur is een voorbeeld van een omega-3-vetzuur. Het heeft drie cis dubbele bindingen en daardoor een gebogen vorm. Zalm, forel en tonijn zijn goede bronnen van omega-3-vetzuren. Onderzoek wijst uit dat omega-3-vetzuren het risico op een plotselinge dood door hartaanvallen verminderen, de triglyceriden in het bloed verlagen, de bloeddruk verlagen en trombose voorkomens door de bloedstolling te remmen. Ze verminderen ook ontstekingen en kunnen het risico op bepaalde vormen van kanker bij dieren helpen verlagen.

Lipid molecules are built from hydrocarbon chains and a few key structural parts. Their shape helps explain why they behave differently from many other biological molecules. In high school biology, this structure is often linked to how lipids store energy and help form cell membranes.

Some lipids are made from fatty acids joined to other small molecules. Triglycerides contain glycerol and three fatty acids, while phospholipids contain glycerol, two fatty acids, and a phosphate group. These structural differences give each type a different role in living things.

Fatty acids can also differ from one another. Saturated fatty acids have only single bonds between carbon atoms, while unsaturated fatty acids have one or more double bonds. These bond patterns change the way lipid molecules pack together and help explain their physical properties.
