Względna różnica ładunku elektrycznego, czyli napięcia, pomiędzy wnętrzem i zewnętrzem błony komórkowej, nazywana jest potencjałem błony. Jest on generowany przez różnice w przepuszczalności membrany dla różnych jonów i stężeniach tych jonów przez membranę.

<h4>Wnętrze neuronu jest bardziej ujemne</h4>

Potencjał błonowy komórki można zmierzyć, umieszczając mikroelektrodę w komórce i porównując ładunek z elektrodą odniesienia w płynie zewnątrzkomórkowym. Potencjał błonowy neuronu w stanie spoczynku—czyli neuronu, który aktualnie nie odbiera ani nie wysyła wiadomości—jest ujemny i zwykle wynosi około -70 miliwoltów (mV). Nazywa się to spoczynkowym potencjałem błonowym. Wartość ujemna wskazuje, że wnętrze membrany jest stosunkowo bardziej ujemne niż na zewnątrz — jest spolaryzowane. Potencjał spoczynkowy wynika z dwóch głównych czynników: selektywnej przepuszczalności błony oraz różnic w stężeniu jonów wewnątrz komórki w porównaniu z otoczeniem.

<h4>Przepuszczalność membrany</h4>

Błony komórkowe są selektywnie przepuszczalne, ponieważ większość jonów i cząsteczek nie może przejść przez dwuwarstwę lipidową bez pomocy, często z białek kanałów jonowych rozciągających się przez błonę. Dzieje się tak, ponieważ naładowane jony nie mogą dyfundować przez nienaładowane hydrofobowe wnętrze membran. Najczęstszymi jonami wewnątrz- i zewnątrzkomórkowymi występującymi w tkance nerwowej są: potas (K+), sód (Na+), chlorek (Cl- >) i wapń (Ca2+). Kiedy neuron jest w spoczynku, głównym rodzajem kanałów jonowych, które są otwarte—są kanały potasowe (K+), które umożliwiają migrację K+ przez błonę. Ta przepuszczalność, w połączeniu z dużymi stężeniami wewnątrzkomórkowymi, sprawia, że spoczynkowy potencjał błonowy neuronu jest zależny głównie od ruchu K+.

<h4>Pompy tworzą gradienty stężeń</h4>

Różnice w stężeniu jonów pomiędzy neuronami wewnątrz i na zewnątrz wynikają przede wszystkim z aktywności pompy sodowo-potasowej (Na+/K+) — transbłonowej białko, które w sposób ciągły pompuje trzy jony Na+ z komórki na każde dwa jony K+, które wpompowuje. W ten sposób ustala się gradienty stężeń, z wyższym stężeniem Na+ na zewnątrz neuronów i wyższe stężenie jonów K+ wewnątrz.

Ponieważ membrana jest przepuszczalna głównie dla K+ w stanie spoczynku—ze względu na otwarte kanały K+—K+ może dyfundować w dół gradient stężenia do obszaru o niższym stężeniu, poza komórką. Te dodatnie ładunki opuszczające komórkę, w połączeniu z faktem, że wewnątrz komórki znajduje się wiele ujemnie naładowanych białek, powodują, że wnętrze komórki jest stosunkowo bardziej ujemne.

Ostatecznie dyfuzja K+ na zewnątrz jest równoważona przez odpychanie elektrostatyczne ładunków dodatnich gromadzących się na zewnątrz ogniwa i osiągnięta zostaje równowaga elektrochemiczna. Efektem netto jest zaobserwowany ujemny potencjał spoczynkowy. Potencjał spoczynkowy jest bardzo ważny w układzie nerwowym, ponieważ zmiany potencjału błonowego — takie jak potencjał czynnościowy — są podstawą sygnalizacji nerwowej.

<h4>Uważaj na rybę rozdymkowatą</h4>

Rozdymka nie jest często spotykana w wielu menu z owocami morza poza Japonią, po części dlatego, że zawiera silną neurotoksynę. Tetrodotoksyna (TTX) to bardzo selektywny bloker kanałów sodowych bramkowany napięciem, który w minimalnych dawkach jest śmiertelny. Średnia dawka śmiertelna (LD50) dla myszy wynosi 334 μg/kg w porównaniu z 8,5 mg/kg w przypadku cyjanku potasu. Służył również jako podstawowe narzędzie w badaniach neurologicznych. The Resting Membrane Potential

Neurons keep a negative resting charge across their membrane. This electrical difference, called the membrane potential, comes from how ions move and how the membrane lets them pass. In a resting neuron, the inside is typically around -70 millivolts and is more negative than the outside, so the cell is polarized.

Two main factors create this resting state. First, the membrane is selectively permeable, which means most ions and molecules cannot cross the lipid bilayer without help from ion channels. Second, ion concentrations are different inside and outside the cell. In nervous tissue, common ions include potassium (K+), sodium (Na+), chloride (Cl-), and calcium (Ca2+).

At rest, potassium channels are the main channels that are open. This makes potassium the ion that most strongly shapes the resting membrane potential. Potassium can move across the membrane more easily than many other ions, and its movement helps set the neuron’s negative charge.

The sodium-potassium pump creates the ion gradients that support this process. This transmembrane protein pumps three Na+ ions out of the cell for every two K+ ions it pumps in. As a result, Na+ builds up outside the neuron, and K+ builds up inside.

Because the membrane is mostly permeable to K+ at rest, potassium diffuses out of the cell down its concentration gradient. As positive charges leave, the inside becomes more negative. Negatively charged proteins inside the cell also contribute to this effect. Eventually, the outward movement of K+ is balanced by the electrical push from the positive charge outside the cell, and electrochemical equilibrium is reached.

This resting potential is important because changes in membrane potential are the basis of neural signaling. One example is the action potential, which depends on these voltage changes. The pufferfish toxin tetrodotoxin (TTX) blocks voltage-gated sodium channels and stops Na+ from entering when the channel opens. It disrupts action potentials, but it does not change the resting membrane potential, so it can be used to silence neuronal activity.
