Długoterminowe wzmocnienie lub LTP to proces wzmacniania synaptycznego, który zachodzi z czasem między pre i postsynaptycznymi połączeniami neuronalnymi.

W jednym mechanizmie, gdy neurony presynaptyczne wielokrotnie uruchamiają i stymulują komórkę postsynaptyczną, działanie to wywołuje zmiany w rodzaju i liczbie kanałów jonowych w błonie postsynaptycznej, takich jak jedna klasa receptorów glutaminianu zwana N-metylo-D-asparaginianem lub NMDA.

Receptory NMDA są zwykle inaktywowane przez jony magnezu, jednak przy silnej depolaryzacji spowodowanej wielokrotną stymulacją jony magnezu są przemieszczane, umożliwiając wnikanie jonów wapnia.

Ten napływ wapnia inicjuje kaskadę sygnalizacyjną, której kulminacją jest druga klasa receptorów glutaminianu. Kwas alfa-amino-trzy-hydroksy-pięciometylo-czteroizoksazolepropionowy, w skrócie AMPA, wprowadzany do membrany. Długoterminowe wzmocnienie, czyli LTP, to jeden ze sposobów, w jaki plastyczność synaptyczna — zmiany w sile synaps chemicznych — mogą wystąpić w mózgu. LTP to proces wzmacniania synaptycznego, który zachodzi w czasie pomiędzy przed- i postsynaptycznymi połączeniami neuronowymi. Wzmocnienie synaptyczne LTP działa w opozycji do osłabienia synaptycznego w przypadku długotrwałej depresji (LTD) i razem stanowią główne mechanizmy leżące u podstaw uczenia się i pamięci.

<h4>Hebbian LTP</h4>

LTP może wystąpić, gdy neurony presynaptyczne wielokrotnie uruchamiają i stymulują neuron postsynaptyczny. Nazywa się to Hebbian LTP, ponieważ wynika z postulatu Donalda Hebba z 1949 r., że “neurony, które wyzwalają się razem, łączą się ze sobą”. Powtarzająca się stymulacja neuronów presynaptycznych indukuje zmiany w rodzaju i liczbie kanałów jonowych w błonie postsynaptycznej.

Dwa typy receptorów postsynaptycznych pobudzającego neuroprzekaźnika glutaminianu biorą udział w LTP: 1) receptory N-metylo-D-asparaginianu lub NMDA i 2) kwas α-amino-3-hydroksy-5-metylo-4-izoksazolopropionowy lub receptory AMPA. Alternatywnie kaskada sygnalizacyjna może fosforylować receptory glutaminianu—umożliwiając im pozostanie otwartymi przez dłuższy czas i zwiększając przewodnictwo dodatnio naładowanych jonów do komórki. W rezultacie ta sama stymulacja presynaptyczna będzie teraz wywoływać silniejszą odpowiedź postsynaptyczną, biorąc pod uwagę, że aktywowanych zostanie więcej receptorów glutaminianowych i więcej dodatnio naładowanych jonów dostanie się do neuronu postsynaptycznego. Występujące wzmocnienie nazywa się wzmocnieniem lub wzmocnieniem synaptycznym.

Powiedzenie “praktyka czyni mistrza” można częściowo wytłumaczyć LTP. Kiedy uczymy się nowego zadania, nowe obwody neuronowe są wzmacniane za pomocą LTP. Po każdej iteracji ćwiczenia siła synaptyczna w obwodach nerwowych staje się silniejsza i wkrótce zadanie będzie można wykonać poprawnie i efektywnie. Nowo wzmocnione połączenia mogą trwać od minut do tygodni lub dłużej, jeśli stymulacja presynaptyczna utrzymuje się, co oznacza, że przy każdym kolejnym wykonaniu zadania LTP jest utrzymywane.

<h4>LTP i choroba</h4>

Kiedy LTP działa normalnie, możemy z łatwością uczyć się i tworzyć wspomnienia. Jednakże nieprawidłowości w LTP powiązano z wieloma zaburzeniami neurologicznymi i poznawczymi, takimi jak choroba Alzheimera, autyzm, uzależnienia, schizofrenia i stwardnienie rozsiane. Lepsze zrozumienie mechanizmów stojących za LTP może ostatecznie doprowadzić do opracowania terapii.

Long-term potentiation, or LTP, is a form of synaptic strengthening in the brain. It changes the strength of chemical synapses over time between pre- and postsynaptic neurons. LTP works against long-term depression, or LTD, which weakens synapses. Together, LTP and LTD are major mechanisms behind learning and memory.

LTP can happen when a presynaptic neuron fires again and again and keeps stimulating a postsynaptic neuron. This is called Hebbian LTP, based on Donald Hebb’s idea that neurons that fire together wire together. Repeated stimulation changes the types and number of ion channels in the postsynaptic membrane. Two glutamate receptors are involved: NMDA receptors and AMPA receptors.

NMDA receptors open when glutamate binds, but magnesium ions usually block the pore. At the same time, glutamate can activate AMPA receptors on the postsynaptic cell. This causes sodium ions to enter and depolarize the membrane. When many presynaptic inputs arrive in quick succession, the magnesium block is removed from the NMDA receptor. Sodium and calcium ions then enter the neuron.

The calcium influx starts a signaling cascade inside the cell. This cascade leads to more AMPA receptors being inserted into the plasma membrane. It can also phosphorylate glutamate receptors, which helps them stay open longer and lets more positively charged ions enter the cell. The same presynaptic signal then creates a stronger postsynaptic response. This increase is called synaptic strengthening, or potentiation.

LTP also helps explain why practice makes perfect. When a new task is learned, new neural circuits are reinforced by LTP. With each round of practice, the synaptic strength in those circuits becomes stronger. Over time, the task can be performed more correctly and efficiently. These strengthened connections can last from minutes to weeks or longer if presynaptic stimulation continues, so later practice can maintain the LTP.

When LTP works normally, learning and memory are easier. Problems with LTP have been linked to Alzheimer’s disease, autism, addiction, schizophrenia, and multiple sclerosis. Understanding how LTP works may help guide future therapies for these neurological and cognitive disorders.
