Stereoizomery związków koordynacyjnych to cząsteczki, które mają ten sam wzór chemiczny i tę samą łączność ligandów z atomem metalu, ale różnią się ułożeniem cząsteczek liganda wokół atomu centralnego.
Ze względu na strukturę molekularną stereoizomery związków koordynacyjnych klasyfikuje się jako izomery geometryczne lub izomery optyczne.
Izomery geometryczne to stereoizomery, które mają zupełnie inną geometrię molekularną i mają różne właściwości fizyczne i chemiczne.
Izomery cis-trans są przykładem izomerów geometrycznych. Występują w kompleksach kwadratowo-płaskich z dwoma zestawami dwóch identycznych ligandów, czyli MA2B2 oraz w kompleksach oktaedrycznych z zestawem czterech identycznych ligandów i parą innego liganda, czyli MA4B2. 
Na przykład w izomerze cis kwadratowo-płaskiego kompleksu diaminodichloroplatyny(II), znanego jako cisplatyna, ligandy chlorkowe sąsiadują ze sobą i znajdują się po tej samej stronie cząsteczki. W izomerze trans, transplatynie, ligandy chlorkowe znajdują się po przeciwnych stronach cząsteczki. 
Kompleksy oktaedryczne z dwoma zestawami trzech identycznych ligandów lub MA3B3 mogą wykazywać inny rodzaj izomerii geometrycznej, znany jako izomeria fac-mer. 
Na przykład w izomerze fac triammintrichlorokobaltu(III) trzy ligandy aminowe i trzy ligandy chlorkowe znajdują się po przeciwnych stronach cząsteczki, a zatem tworzą przeciwległe ściany ośmiościanu. W izomerze mera mera trzy takie same ligandy tworzą łuk wokół centralnego atomu metalu.
Druga klasa stereoizomerów jest zdefiniowana jako izomery optyczne. Podobnie jak nasze dłonie, izomery optyczne są swoimi lustrzanymi odbiciami i nie nakładają się na siebie. Izomery te, zwane również enancjomerami, są opisywane jako chiralne i mają bardzo niewiele różnic w swoich właściwościach fizycznych i chemicznych.
Izomery optyczne wyróżniają się oddziaływaniem ze światłem spolaryzowanym płaszczyznowo. Światło spolaryzowane ma wektory pola elektrycznego oscylujące w jednej płaszczyźnie, która jest obracana o pewną wartość, gdy spolaryzowane światło przechodzi przez roztwór enancjomeru. 
Na przykład jeden enancjomer tris(etylenodiaminy)kobaltu(III) obraca płaszczyznę w prawo i nazywa się go prawoskrętnym lub izomerem optycznym d. Drugi enancjomer obraca płaszczyznę w lewo i nazywa się lewoskrętnym lub l izomerem optycznym.

Izomery to różne gatunki chemiczne, które mają ten sam wzór chemiczny.

Kompleksy metali przejściowych często występują w postaci izomerów geometrycznych, w których te same atomy są połączone tymi samymi rodzajami wiązań, ale różnią się orientacją w przestrzeni. Kompleksy koordynacyjne z dwoma różnymi ligandami w pozycjach cis i trans z danego liganda tworzą izomery. Na przykład oktaedryczny jon [Co(NH3)4Cl2]+ ma dwa izomery (Rysunek 1). W konfiguracji cis dwa ligandy chlorkowe sąsiadują ze sobą. Drugi izomer, konfiguracja trans, ma dwa ligandy chlorkowe bezpośrednio naprzeciw siebie.

Różne izomery geometryczne substancji to różne związki chemiczne. Wykazują różne właściwości, mimo że mają tę samą formułę. Na przykład dwa izomery [Co(NH3)4Cl2]NO3 różnią się kolorem: forma cis jest fioletowa, a forma trans jest zielona. Ponadto izomery te mają różne momenty dipolowe, rozpuszczalność i reaktywność. Jako przykład tego, jak układ w przestrzeni może wpływać na właściwości molekularne, rozważmy polarność dwóch izomerów [Co(NH3)4Cl2]NO3. Należy pamiętać, że o polarności cząsteczki lub jonu decydują dipole wiązań (które wynikają z różnicy elektroujemności atomów wiążących) i ich rozmieszczenie w przestrzeni. W jednym izomerze ligandy chlorku cis powodują większą gęstość elektronów po jednej stronie cząsteczki niż po drugiej, czyniąc ją polarną. W przypadku izomeru trans każdy ligand znajduje się bezpośrednio naprzeciw identycznego liganda, więc dipole wiązań znoszą się, a cząsteczka jest niepolarna.

Innym ważnym typem izomerów są izomery optyczne, czyli enancjomery, w których dwa obiekty są swoimi dokładnymi odbiciami lustrzanymi, ale nie można ich ustawić tak, aby wszystkie części do siebie pasowały. Oznacza to, że izomery optyczne są nienakładającymi się na siebie odbiciami lustrzanymi. Klasycznym tego przykładem jest para rąk, w której prawa i lewa ręka są swoimi lustrzanymi odbiciami, ale nie można ich nałożyć na siebie. Izomery optyczne są bardzo ważne w chemii organicznej i biochemii, ponieważ systemy żywe często zawierają jeden konkretny izomer optyczny, a nie dwa. W przeciwieństwie do izomerów geometrycznych pary izomerów optycznych mają prawie identyczne właściwości (temperatura wrzenia, polarność, rozpuszczalność itp.). Izomery optyczne różnią się jedynie sposobem, w jaki wpływają na światło spolaryzowane i jak reagują z innymi izomerami optycznymi. W przypadku kompleksów koordynacyjnych wiele związków koordynacyjnych, takich jak [M(en)3]n+ [w których Mn+ jest centralnym jonem metalu, takim jak żelazo(III) lub kobalt(II)] tworzy enancjomery, jak pokazano na rysunku 2. Te dwa izomery będą reagować inaczej z innymi izomerami optycznymi. Na przykład helisy DNA są izomerami optycznymi i forma występująca w przyrodzie (prawoskrętny DNA) będzie wiązać się tylko z jednym izomerem [M(en)3]n+, a z drugim nie.

Jon [Co(en)2Cl2]+ wykazuje izomerię geometryczną (cis/trans), a jego izomer cis występuje w postaci pary izomerów optycznych (Rysunek 3).

Isomer shapes in coordination complexes can change a compound’s properties even when the chemical formula stays the same. Isomers are different chemical species with the same formula. In transition metal complexes, the atoms stay connected through the same types of bonds, but their arrangement in space can differ.

One common form is geometric isomerism. In these complexes, ligands (the ions or molecules attached to the metal) can sit in cis or trans positions. The octahedral [Co(NH3)4Cl2]+ ion has two geometric isomers. In the cis form, the two chloride ligands are next to each other. In the trans form, the chlorides are directly across from one another.

These geometric isomers can behave very differently. The two isomers of [Co(NH3)4Cl2]NO3 have different colors. The cis form is violet, and the trans form is green. They also differ in dipole moment, solubility, and reactivity. Polarity depends on bond dipoles and how they are arranged in space. In the cis isomer, the chloride ligands create more electron density on one side of the ion, so it is polar. In the trans isomer, matching ligands are opposite each other, so the dipoles cancel and the ion is nonpolar.

Another important category is optical isomerism, also called enantiomerism. Enantiomers are mirror images that cannot be superposed, like right and left hands. These optical isomers are important in organic chemistry and biochemistry because living systems often use one optical isomer but not the other. Unlike geometric isomers, enantiomers usually have nearly identical boiling points, polarity, and solubility. They differ in how they affect polarized light and how they react with other optical isomers.

Many coordination compounds also form enantiomers. A complex such as [M(en)3]n+, where M is a metal ion like iron(III) or cobalt(II) and en is ethylenediamine, can exist as a pair of non-superposable mirror images. These forms react differently with other optical isomers. For example, DNA helices are optical isomers, and natural right-handed DNA binds to only one isomer of [M(en)3]n+.

Some complexes show both geometric and optical isomerism. The [Co(en)2Cl2]+ ion has cis and trans forms. Its trans isomer has the chlorines 180° apart and has very different properties from the cis form. The cis isomer also exists as a pair of optical isomers. These mirror-image forms behave the same in most ways, except when they react with other enantiomers.
