Structure of Lipids

Lipidy to grupa cząsteczek hydrofobowych, które obejmują trójglicerydy, które służą do magazynowania energii, oraz fosfolipidy, które są głównymi składnikami strukturalnymi błon komórkowych.

Lipidy są także budulcem wielu hormonów i ważnym składnikiem wszystkich błon komórkowych. Lipidy obejmują tłuszcze, oleje, woski, fosfolipidy i steroidy.

Ogólna struktura lipidów

Cząsteczka tłuszczu składa się z dwóch głównych składników – gliceryny i kwasów tłuszczowych. Glicerol to związek organiczny (alkohol) posiadający trzy atomy węgla, pięć wodorów i trzy grupy hydroksylowe (OH). Kwasy tłuszczowe mają długi łańcuch węglowodorów, do którego przyłączona jest grupa karboksylowa, stąd nazwa „kwas tłuszczowy”. Liczba atomów węgla w kwasie tłuszczowym może wynosić od 4 do 36. Najczęściej spotykane są te zawierające 12–18 atomów węgla. W cząsteczce tłuszczu kwasy tłuszczowe przyłączają się do każdego z trzech atomów węgla cząsteczki glicerolu za pomocą wiązania estrowego poprzez atom tlenu. Połączenie trzech kwasów tłuszczowych ze szkieletem glicerolu w reakcji odwodnienia tworzy triacyloglicerol. Trzy kwasy tłuszczowe w triacyloglicerolu mogą być podobne lub niepodobne.

Fosfolipidy to kolejny powszechny rodzaj lipidów. Jest cząsteczką amfipatyczną, co oznacza, że posiada część hydrofobową i hydrofilową. Łańcuchy kwasów tłuszczowych są hydrofobowe i nie mogą oddziaływać z wodą; podczas gdy grupa zawierająca fosforan jest hydrofilowa i oddziałuje z wodą. Hydrofilowe grupy główne fosfolipidów są zwrócone w stronę roztworu wodnego. Hydrofobowe ogony są zamaskowane w środku dwuwarstwy.

Kwasy tłuszczowe

Kwasy tłuszczowe mogą być nasycone lub nienasycone. W łańcuchu kwasów tłuszczowych, jeśli pomiędzy sąsiednimi atomami węgla w łańcuchu węglowodorowym występują tylko pojedyncze wiązania, kwas tłuszczowy jest nasycony. Kwas stearynowy jest przykładem nasyconego kwasu tłuszczowego.

Gdy łańcuch węglowodorowy zawiera wiązanie podwójne, kwas tłuszczowy jest nienasycony. Kwas oleinowy jest przykładem nienasyconego kwasu tłuszczowego. Większość tłuszczów nienasyconych ma postać płynną w temperaturze pokojowej i nazywane są olejami. Jeżeli w cząsteczce występuje jedno wiązanie podwójne, to jest to tłuszcz jednonienasycony (np. oliwa z oliwek), a jeśli w cząsteczce występuje więcej niż jedno wiązanie podwójne, to jest to tłuszcz wielonienasycony (np. olej rzepakowy). Długie, proste kwasy tłuszczowe z pojedynczymi wiązaniami zazwyczaj ściśle przylegają i są stałe w temperaturze pokojowej. Przykładami tłuszczów nasyconych są tłuszcze zwierzęce zawierające kwas stearynowy i palmitynowy (powszechne w mięsie) oraz tłuszcze z kwasem masłowym (powszechne w maśle).

Kwasy tłuszczowe można dodatkowo podzielić na Cis i trans. Cis i trans wskazują konfigurację cząsteczki wokół wiązania podwójnego. Jeśli wodory są obecne w tej samej płaszczyźnie, jest to tłuszcz cis. Jeśli atomy wodoru znajdują się w dwóch różnych płaszczyznach, jest to tłuszcz trans. Podwójne wiązanie cis powoduje zagięcie lub „załamanie”, które zapobiega ścisłemu upakowaniu kwasów tłuszczowych, utrzymując je w stanie ciekłym w temperaturze pokojowej. Przykładami tłuszczów nienasyconych są oliwa z oliwek, olej kukurydziany, olej rzepakowy i olej z wątroby dorsza. Tłuszcze nienasycone pomagają obniżyć poziom cholesterolu we krwi, podczas gdy tłuszcze nasycone przyczyniają się do tworzenia się płytek w tętnicach.

Tłuszcze trans

Przemysł spożywczy sztucznie uwodornia oleje, aby stały się półstałe i miały konsystencję pożądaną dla wielu przetworzonych produktów spożywczych. Podczas tego procesu wiązania podwójne o konformacji cis – w łańcuchu węglowodorowym mogą przekształcić się w wiązania podwójne o konformacji trans.

Margaryna, niektóre rodzaje masła orzechowego i tłuszcz piekarski to przykłady sztucznie uwodornionych tłuszczów trans. Najnowsze badania wykazały, że wzrost zawartości tłuszczów trans w diecie człowieka może prowadzić do podwyższenia poziomu lipoprotein o małej gęstości (LDL), czyli „złego” cholesterolu, co z kolei może prowadzić do odkładania się blaszek w tętnicach, powodując choroby serca.

Kwasy Omega

Niezbędne nienasycone kwasy tłuszczowe to te, których organizm ludzki potrzebuje, ale których nie syntetyzuje. Dlatego należy je uzupełniać poprzez dietę. Kwasy tłuszczowe omega-3 należą do tej kategorii i są jednymi z zaledwie dwóch znanych człowiekowi (drugim jest kwas tłuszczowy omega-6). Są to wielonienasycone kwasy tłuszczowe i należą do grupy omega-3, ponieważ podwójne wiązanie łączy trzeci węgiel od końca łańcucha węglowodorowego z sąsiednim węglem.

Kwas alfa-linolenowy jest przykładem kwasu tłuszczowego omega-3. Posiada trzy podwójne wiązania cis i w rezultacie ma zakrzywiony kształt. Łosoś, pstrąg i tuńczyk są dobrymi źródłami kwasów tłuszczowych omega-3. Badania wskazują, że kwasy tłuszczowe omega-3 zmniejszają ryzyko nagłej śmierci z powodu zawału serca, obniżają poziom trójglicerydów we krwi, obniżają ciśnienie krwi i zapobiegają zakrzepicy poprzez hamowanie krzepnięcia krwi. Zmniejszają także stany zapalne i mogą pomóc zmniejszyć ryzyko niektórych nowotworów u zwierząt.

Lipid molecules are built from hydrocarbon chains and a few key structural parts. Their shape helps explain why they behave differently from many other biological molecules. In high school biology, this structure is often linked to how lipids store energy and help form cell membranes.

Some lipids are made from fatty acids joined to other small molecules. Triglycerides contain glycerol and three fatty acids, while phospholipids contain glycerol, two fatty acids, and a phosphate group. These structural differences give each type a different role in living things.

Fatty acids can also differ from one another. Saturated fatty acids have only single bonds between carbon atoms, while unsaturated fatty acids have one or more double bonds. These bond patterns change the way lipid molecules pack together and help explain their physical properties.
