białko i struktury białkowe

Białka są jednymi z najobficiej występujących cząsteczek organicznych w żywych systemach i mają najbardziej zróżnicowany zakres funkcji spośród wszystkich makrocząsteczek. Białka mogą być strukturalne, regulacyjne, kurczliwe lub ochronne. Mogą służyć w transporcie, magazynowaniu lub błonach; mogą też być toksynami lub enzymami. Ich struktury, podobnie jak funkcje, są bardzo zróżnicowane. Wszystkie są jednak polimerami aminokwasów ułożonymi w sekwencji liniowej.

Kształt białka ma kluczowe znaczenie dla jego funkcji. Na przykład enzym może wiązać specyficzny substrat w jego miejscu aktywnym. Jeśli to miejsce aktywne zostanie zmienione z powodu lokalnych zmian lub zmian w ogólnej strukturze białka, enzym może nie być w stanie związać się z substratem. Aby zrozumieć, w jaki sposób białko uzyskuje swój ostateczny kształt lub konformację, musimy zrozumieć cztery poziomy struktury białka: pierwszorzędową, drugorzędową, trzeciorzędową i czwartorzędową.

Struktura pierwszorzędowa

Może to prowadzić do niezliczonych poważnych problemów zdrowotnych, takich jak duszność, zawroty głowy, bóle głowy i bóle brzucha u osób dotkniętych tą chorobą.

Struktura drugorzędowa

Lokalne fałdowanie polipeptydu w niektórych regionach powoduje powstanie drugorzędowej struktury białka. Najbardziej powszechne są struktury helisy α i pofałdowanej płaszczyzny β. Obie struktury utrzymują swój kształt za pomocą wiązań wodorowych. Wiązania wodorowe tworzą się pomiędzy atomem tlenu w grupie karbonylowej w jednym aminokwasie i innym aminokwasem, który jest oddalony o cztery aminokwasy dalej w łańcuchu.

Każdy zwój spiralny w helisie alfa ma 3,6 reszt aminokwasowych. Grupy R polipeptydu (grupy wariantowe) wystają z łańcucha helisy α. W pofałdowanej płaszczyźnie β wiązania wodorowe między atomami w szkielecie łańcucha polipeptydowego tworzą „fałdy”. Grupy R są przyłączone do węgli i rozciągają się powyżej i poniżej fałd harmonijki. Pofałdowane segmenty są ustawione równolegle lub antyrównolegle względem siebie, a wiązania wodorowe tworzą się pomiędzy częściowo dodatnim atomem wodoru w grupie aminowej i częściowo ujemnym atomem tlenu w grupie karbonylowej szkieletu peptydowego. Struktury helisy α i pofałdowanej płaszczyzny β występują w większości białek kulistych i włóknistych i odgrywają ważną rolę strukturalną.

Struktura trzeciorzędowa

Unikalną trójwymiarową strukturą polipeptydu jest jego struktura trzeciorzędowa. Struktura ta jest częściowo spowodowana interakcjami chemicznymi zachodzącymi w łańcuchu polipeptydowym. Przede wszystkim interakcje między grupami R tworzą złożoną trójwymiarową strukturę trzeciorzędową białka. Charakter grup R w zaangażowanych aminokwasach może przeciwdziałać tworzeniu wiązań wodorowych, które opisaliśmy dla standardowych struktur drugorzędowych. Na przykład grupy R o podobnych ładunkach odpychają się, a grupy o różnych ładunkach przyciągają się (wiązania jonowe). Kiedy ma miejsce fałdowanie białka, hydrofobowe grupy R niepolarnych aminokwasów znajdują się we wnętrzu białka; podczas gdy hydrofilowe grupy R leżą na zewnątrz. Interakcje między łańcuchami bocznymi cysteiny tworzą wiązania disiarczkowe w obecności tlenu, jedynego wiązania kowalencyjnego, które tworzy się podczas fałdowania białka.

Wszystkie te interakcje, słabe i mocne, determinują ostateczny trójwymiarowy kształt białka. Kiedy białko traci swój trójwymiarowy kształt, może przestać działać.

Struktura czwartorzędowa

W naturze niektóre białka tworzą się z kilku polipeptydów lub podjednostek, a interakcja tych podjednostek tworzy strukturę czwartorzędową. Słabe interakcje między podjednostkami pomagają ustabilizować ogólną strukturę. Na przykład insulina (białko globularne) ma kombinację wiązań wodorowych i dwusiarczkowych, które powodują, że zbija się ona głównie w kształt kuli. Insulina zaczyna jako pojedynczy polipeptyd i traci pewne wewnętrzne sekwencje w obecności modyfikacji potranslacyjnej po utworzeniu wiązań dwusiarczkowych, które utrzymują razem pozostałe łańcuchy. Jedwab (białko włókniste) ma jednak strukturę pofałdowanej płaszczyzny β, która jest wynikiem wiązań wodorowych pomiędzy różnymi łańcuchami.

Proteins are essential molecules in living things, and their shape affects how they work. Protein structure is described at several levels, including primary, secondary, tertiary, and quaternary structure. These levels help explain how a protein is built and how its final form supports its job.

Primary structure is the sequence of amino acids in a protein. Secondary structure forms when parts of the chain fold into regular patterns. Tertiary structure is the overall three-dimensional shape of one protein chain. Quaternary structure appears when more than one protein chain joins together to make a working protein.

The video also shows how protein structure can be represented in different ways. A simple line drawing can show the amino acid sequence. Other models can show folding and the full 3D shape. These visual forms help students compare levels of structure and see why protein shape matters.
