O potencial da membrana em repouso é a diferença no potencial elétrico entre o interior e o exterior da membrana celular de neurónios em repouso quando eles não estão a ser estimulados. Normalmente, o valor é em torno de 70 milivolts, o que significa que é mais negativo por dentro. As membranas celulares são seletivamente permeáveis porque a maioria dos iões e moléculas não podem se difundir passivamente entre elas.Elas geralmente exigem proteínas transmembranares, como canais iónicos, para permitir que eles passem. Quando um neurónio está em repouso, canais de potássio são o tipo principal de canais iónicos que estão abertos. Outra proteína transmembranar, a bomba de sódio-potássio, usa energia para mover continuamente o sódio para fora da célula e o potássio para dentro.Esta ação cria um gradiente de concentração com maior concentração de potássio dentro do que fora. A força de difusão então faz com que iões de potássio se movam para baixo do seu gradiente de concentração através dos canais de potássio abertos e para fora da célula.  A diferença relativa na carga elétrica, ou tensão, entre o interior e o exterior de uma membrana celular, é chamada de potencial de membrana. É gerado por diferenças na permeabilidade da membrana a vários iões e as concentrações desses iões através da membrana.
 
 <h4>O Interior de um Neurónio é Mais Negativo</h4>
 
 O potencial de membrana de uma célula pode ser medido inserindo um microelétrodo em uma célula e comparando a carga a um elétrodo de referência no fluido extracelular. O potencial de membrana de um neurónio em repouso—ou seja, um neurónio que não está atualmente a receber ou enviar mensagens—é negativo, tipicamente em torno de -70 milivolts (mV). Isso é chamado de potencial de repouso da membrana. O valor negativo indica que o interior da membrana é relativamente mais negativo do que o exterior—está polarizado. O potencial de repouso resulta de dois fatores principais: permeabilidade seletiva da membrana, e diferenças na concentração de iões dentro da célula em comparação com o exterior.
 
 <h4>Permeabilidade da Membrana</h4>
 
 As membranas celulares são seletivamente permeáveis porque a maioria dos iões e moléculas não podem atravessar a bicamada lipídica sem ajuda, muitas vezes de proteínas de canais de iões ao longo da membrana. Isso porque os iões carregados não se podem difundir através do interior hidrofóbico não carregado das membranas. Os iões intra e extracelulares mais comuns encontrados no tecido nervoso são potássio (K+), sódio (Na+), cloreto (Cl-), e cálcio (Ca2+). Quando um neurónio está em repouso, os canais de potássio (K+) são o principal tipo de canal de iões aberto—permitindo que o K+ migre através da membrana. Essa permeabilidade, juntamente com as grandes concentrações intracelulares, tornam o potencial de membrana de repouso do neurónio determinado principalmente pelo movimento de K+.
 
 <h4>Bombas Criam Gradientes de Concentração</h4>
 
 Diferenças na concentração de iões entre o interior e o exterior dos neurónios devem-se principalmente à atividade da bomba de sódio-potássio (Na+/ K+)—uma proteína transmembranar que bombeia continuamente três iões de Na+ para fora da célula por cada dois iões de K+ que bombeia para dentro. Isto estabelece gradientes de concentração, com maior concentração de iões Na+ fora dos neurónios e maior concentração de iões K+ dentro.
 
 Uma vez que a membrana é principalmente permeável a K+ em repouso—devido aos canais de K+ abertos—o K+ pode difundir o seu gradiente de concentração para a região de menor concentração, fora da célula. Essas cargas positivas que deixam a célula, combinadas com o facto de existirem muitas proteínas negativamente carregadas dentro da célula, fazem com que o interior seja relativamente mais negativo.
 
 Eventualmente, a difusão de K+ para o exterior é equilibrada pela repulsa eletrostática de cargas positivas acumuladas fora da célula, e o equilíbrio eletroquímico é atingido. O efeito líquido é o potencial de repouso negativo observado. O potencial de repouso é muito importante no sistema nervoso porque mudanças no potencial da membrana—como o potencial de ação—são a base para a sinalização neural.
 
 <h4>Cuidado com o Peixe-Balão</h4>
 
 O peixe-balão não é frequentemente encontrado em muitos menus de alimentos marinhos fora do Japão, em parte porque eles contêm uma neurotoxina potente. A tetrodotoxina (TTX) é um bloqueador muito seletivo de canais de sódio dependentes de voltagem que é letal em doses mínimas. A dose letal média (LD50) para murganhos é de 334 μg/kg, em comparação com 8,5 mg/kg para cianeto de potássio. Ele também serviu como uma ferramenta essencial na investigação em neurociências. A toxina bloqueia o fluxo de Na+ para dentro da célula quando o canal abre. Ele, portanto, interrompe os potenciais de ação—mas não o potencial de repouso da membrana—e pode ser usado para silenciar a atividade neuronal. The Resting Membrane Potential

Neurons keep a negative resting charge across their membrane. This electrical difference, called the membrane potential, comes from how ions move and how the membrane lets them pass. In a resting neuron, the inside is typically around -70 millivolts and is more negative than the outside, so the cell is polarized.

Two main factors create this resting state. First, the membrane is selectively permeable, which means most ions and molecules cannot cross the lipid bilayer without help from ion channels. Second, ion concentrations are different inside and outside the cell. In nervous tissue, common ions include potassium (K+), sodium (Na+), chloride (Cl-), and calcium (Ca2+).

At rest, potassium channels are the main channels that are open. This makes potassium the ion that most strongly shapes the resting membrane potential. Potassium can move across the membrane more easily than many other ions, and its movement helps set the neuron’s negative charge.

The sodium-potassium pump creates the ion gradients that support this process. This transmembrane protein pumps three Na+ ions out of the cell for every two K+ ions it pumps in. As a result, Na+ builds up outside the neuron, and K+ builds up inside.

Because the membrane is mostly permeable to K+ at rest, potassium diffuses out of the cell down its concentration gradient. As positive charges leave, the inside becomes more negative. Negatively charged proteins inside the cell also contribute to this effect. Eventually, the outward movement of K+ is balanced by the electrical push from the positive charge outside the cell, and electrochemical equilibrium is reached.

This resting potential is important because changes in membrane potential are the basis of neural signaling. One example is the action potential, which depends on these voltage changes. The pufferfish toxin tetrodotoxin (TTX) blocks voltage-gated sodium channels and stops Na+ from entering when the channel opens. It disrupts action potentials, but it does not change the resting membrane potential, so it can be used to silence neuronal activity.
