Protein-Protein Interfaces

Muitos processos biológicos dependem das interações proteína-proteína. De fato, um grande número de proteínas necessidade de formar complexos de proteínas, ou oligómeros, para realizar suas funções. Muitas proteínas formam complexos para desempenhar suas funções, tornando as interações proteína-proteína (PPIs) essenciais para a sobrevivência de um organismo. A maioria das PPIs é estabilizada por numerosas forças químicas não covalentes fracas. A forma física das interfaces determina a forma como duas proteínas interagem. Muitas proteínas globulares têm formas semelhantes em suas superfícies, que formam um grande número de ligações fracas. Além disso, muitas PPIs ocorrem entre duas hélices ou entre uma fenda superficial e uma cadeia ou sequência polipeptídica. Vários métodos computacionais e bioquímicos são usados para estudar interfaces de proteínas. Métodos laboratoriais, como purificação por afinidade, espectrometria de massa e microarranjos de proteínas, podem ser usados para identificar novas interações. A co-imunoprecipitação de proteínas e a triagem de dois híbridos de levedura são amplamente utilizadas para fornecer evidências sobre se duas proteínas interagem in vitro. Protein binding surfaces and contacts help proteins recognize and attach to each other. These interfaces are the regions where two proteins meet and interact. They are important for many cell processes and for how proteins work together.

A protein-protein interface is not just a random touch point. It is shaped by the three-dimensional form of each protein. The matching surfaces allow a stable interaction to form when the proteins fit together well.
