As células interagem com uma rede de moléculas secretadas localmente ao redor da membrana plasmática, chamada de matriz extracelular ou MEC.

Cada complexo de adesão compreende receptores transmembranares e proteínas adaptadoras citosólicas que ligam os filamentos do citoesqueleto com ligantes na MEC.
A integrina, o receptor de adesão mais proeminente, e seu adaptador talin podem se ligar a qualquer uma das três principais classes de moléculas de MEC: proteoglicanos, que amortecem a célula; colágeno, que fornece resistência mecânica; e proteínas solúveis multiadesivas, como a fibronectina, que reticulam os receptores de adesão com outros componentes da matriz.
As integrinas unem os filamentos do citoesqueleto com uma forma especializada da MEC na lâmina basal que reveste a superfície externa da membrana celular.
O complexo de adesão resultante interage com as moléculas de ECM e transmite sinais mecânicos e químicos para regular o crescimento, migração e diferenciação celular.

Overview of Cell-Matrix Interactions

A matriz extracelular, ou MEC, mantém as células unidas para formar um tecido e permite que as células dentro do tecido se comuniquem. A MEC compreende proteínas como fibronectina, colágeno, laminina, etc. A proteína mais abundante neste espaço é o colágeno. As fibras de colágeno são entrelaçadas com moléculas de proteínas contendo carboidratos chamadas proteoglicanos. A MEC permite a migração celular e fornece um andaime estrutural na adesão celular que ancora a célula quando as proteínas da matriz extracelular interagem com os receptores de proteínas transmembrana. Um ligante da MEC se liga ao receptor e altera sua conformação. O receptor, por sua vez, altera a conformação dos microfilamentos posicionados logo abaixo da membrana plasmática. Estas alterações conformacionais induzem sinais químicos dentro da célula que atingem o núcleo e “ligam” ou “desligam” a transcrição de secções específicas de DNA , afetando a produção de proteínas associadas, alterando assim as atividades celulares. As integrinas são os receptores transmembrana mais proeminentes que fazem a mediação das junções célula-matriz, como adesões focais e hemidesmossomos. The extracellular matrix, or ECM, is the material that helps hold cells together in a tissue. It also lets cells in that tissue communicate with one another. The ECM contains proteins such as fibronectin, collagen, and laminin. Collagen is the most abundant protein in this space.

Collagen fibers are woven together with proteoglycans, which are protein molecules with carbohydrates attached. This mix gives the ECM its structure. It also supports cell migration and provides a scaffold for cell adhesion. At these adhesion sites, ECM proteins connect with transmembrane protein receptors and help anchor the cell.

When an ECM ligand binds to a receptor, the receptor changes shape. This change also affects the microfilaments just under the plasma membrane. The shape changes trigger chemical signals inside the cell that travel to the nucleus. There, they can turn specific DNA sections on or off and change the proteins the cell makes.

These signals can alter cellular activities. Integrins are the main transmembrane receptors that mediate cell-matrix junctions such as focal adhesions and hemidesmosomes. On the inside of the cell, integrins bind to actin and intermediate filaments. On the ECM side, they bind to fibronectin and collagen.
