Schlenk Lines Transfer of Solvents

1. Utilização de linhas Schlenk
1 Start Up

	<li>Limpe o espaço para a cabeça (e potencialmente o aparelho) de ar residual e umidade realizando 3 ciclos de enchimento de gás a vácuo/inerte: (1) abra a porta de reação ao vácuo, espere o vácuo completo e, em seguida, feche a porta de reação ao vácuo; (2) abrir lentamente a porta de reação ao gás inerte e, em seguida, esperar até que o borbulhador comece a borbulhar novamente. Abrir rapidamente a porta para gás inerte pode resultar em ar sendo sugado para dentro do sistema através do borbulhador. Feche o porto de reação ao gás inerte. (3) Repita 3 vezes o total.</li>

ATENÇÃO: se um líquido azul estiver presente na armadilha após a remoção, é possivelmente oxigênio líquido (B.P. -183 °C), que pode condensar a temperaturas de nitrogênio líquido (B.P. -196 °C) e pode formar misturas explosivas com compostos orgânicos ou causar uma rápida explosão de pressurização em um sistema fechado após o aquecimento. 7 No caso de presença de oxigênio líquido, substitua imediatamente a dewar para manter a armadilha fria, feche a faixa do capô da fumaça e informe outros da situação. Deixe a armadilha para cima e o sistema sob vácuo, e deixe-o lentamente aquecer e evaporar. A condensação do oxigênio líquido pode ser evitada garantindo que os coletores não vazem e limitando a exposição ao vácuo à atmosfera ambiente. Um evento semelhante pode ocorrer com argônio líquido (B. P. -186 °C), e deve-se tomar cuidado para não bombear volumes significativos de argônio através de uma armadilha LN2. 
</li>
	<li>Desligue a bomba de vácuo e desligue o sistema para a atmosfera ambiente, abrindo a saída de armadilha de solvente (se aplicável) ou uma porta no coletor de vácuo principal.</li>
	<li>Remova as armadilhas de solventes e descarte quaisquer voláteis presos no recipiente de resíduos apropriado.</li>
</ol>2. Secagem de Solventes/Reagentes de Hidrocarbonetos
ATENÇÃO: O metal de sódio reage violentamente com a água. Os radicais ketyl são perigosamente incompatíveis com alguns solventes, em particular solventes halogenados. As referências adequadas devem ser consultadas antes de escolher um dessecante apropriado para um determinado solvente. Siga as diretrizes em Purificação de Produtos Químicos laboratoriais. 8-9 
1 Preparação de um "Pote solvente" – 5 g Ph2CO por litro de solvente. 10
<ol><li>Consulte a Figura 1 para obter vidros necessários. Em atmosfera inerte, meça aproximadamente 1 cm5,6 Na metal (fio ou pedaço), corte-o em pedaços menores e coloque as peças em um frasco de fundo redondo de 500 mL com uma articulação de pescoço padrão 24/40.</li>
	<li>Pesar aproximadamente 1,25 g de Ph2CO e colocá-lo no frasco fundo redondo de 500 mL com o sódio.</li>
	<li>Coloque uma barra de agitação pesada no frasco de fundo redondo de 500 mL, em seguida, sele o frasco usando um adaptador 180° 24/40 que foi lubrificado com uma quantidade mínima de graxa de vácuo alto trabalho pesado. 	<li>Coloque um funil na parte superior da panela solvente(não use graxa!) e encha o funil com o solvente desejado. Usando uma agulha longa presa a uma linha de nitrogênio, bolha de nitrogênio através do solvente para desgas parcialmente.</li>
	<li>Mantendo o nitrogênio borbulhando, abra lentamente o adaptador de 180° para introduzir solvente no pote de solvente. Quando o nível do solvente no funil se aproximar do adaptador de 180°, feche o adaptador e remova o funil.  NOTA: O solvente pode ser introduzido de outras formas, com diferentes graus de rigor sem ar. Comumente, os usuários podem optar por encher a panela enquanto estão abertos ao ar e desgas mais tarde, ou podem optar por encher a panela com solvente pré-seco diretamente retirado de um sistema de purificação de solventes.
</li>
	<li>Mexa o pote de solvente por várias horas, durante o qual a solução ficará roxa profunda indicando a formação do ketil de benzofenona de sódio radical. No caso de a panela não ficar roxa profunda, desgas a solução (Seção 2.2) e mexa novamente.</li>
</ol>2 Congelamento da bomba-descongelamento para solvente de degas
ATENÇÃO: O nitrogênio líquido é comumente usado para "congelar" solventes para ciclos de congelamento da bomba.. É altamente recomendável não usar nitrogênio líquido a menos que seja absolutamente necessário e após consulta com os supervisores. O resfriamento de um sistema fechado a temperaturas líquidas de nitrogênio aumenta consideravelmente a chance de condensar oxigênio líquido se houver um vazamento no sistema. Além disso, o grande gradiente de temperatura aumenta a chance de quebra de vidro devido ao choque térmico. Em quase todos os casos, -78 °C é suficiente para resfriar um sistema de desgassing com perda mínima de solventes.
<ol style="margin-left: 40px;"><li>Inicie a linha Schlenk ou high vacuum (Seção 1.1) e conecte o pote de solvente ao coletor (Seção 1.1.6).</li>
	<li>Use banho seco de resfriamento de gelo/acetona para esfriar o conteúdo da panela para -78 °C. Isso pode congelar o solvente dependendo do seu ponto de congelamento.  NOTA: Um equívoco comum do "congelamento-bomba-degelo" é que o solvente deve ser congelado para desgas-lo. </li>
	<li>Se o ponto de congelamento do solvente estiver acima de -78 °C e o solvente estiver congelado, abra o pote de solvente para aspirar, abrindo o adaptador de 180°, a fim de remover o espaço para a cabeça do gás na panela.  Aguarde que o sistema volte ao vácuo total e, em seguida, feche o pote de solvente para aspirar.</li>
	<li>Deixe que o pote de solvente retorne à temperatura ambiente e observe se alguma bolha está se formando na mistura sólido/líquido. Se as bolhas estiverem presentes, o solvente não está totalmente desgaseado e as etapas 2.2.2-2.2.4 devem ser repetidas. Normalmente, três ciclos de "congelamento-bomba-degelo" são suficientes para desgasear.</li>
	<li>Se o ponto de congelamento do solvente estiver abaixo de -78 °C e o solvente tiver uma baixa pressão de vapor a -78 °C, em vez de realizar as etapas 2.2.3 e 2.2.4, o pote de solvente pode ser aberto ao vácuo por aproximadamente 5 minutos (ou até parar borbulhante) para desgas o sistema. Após 5 minutos, feche o adaptador de 180°. Uma pequena quantidade de solvente pode ser perdida para a armadilha da bomba de vácuo.</li>
	<li>Se o ponto de congelamento do solvente estiver abaixo de -78 °C e o solvente tiver maior pressão de vapor a -78 °C, duas opções estão disponíveis: realizar as etapas 2.2.3 e 2.2.4 e esfriar com nitrogênio líquido em vez do banho de -78 °C (menor perda de solvente, risco mais inerente) ou realizar o passo 2.2.5 para períodos mais curtos de tempo (perda mais solvente, mais seguro).</li>
</ol>3. Transferência de vácuo de solventes/produtos químicos
<ol><li>Seque um frasco de 500 mL recebendo frasco Straus e uma ponte de transferência de vácuo em forma de U em um forno de secagem (125 °C) por pelo menos 1 h.</li>
	<li>Conecte o frasco Straus e a panela de solvente à ponte em forma de U e unte todas as juntas levemente com graxa. Conecte a ponte em forma de U à linha de vácuo.  NOTA: Este sistema pesado deve ser suportado por tomadas de laboratório e clipes keck para evitar que ele caia fora da linha e quebre. O vácuo do coletor pode não ser suficiente para mantê-lo na linha.
</li>
	<li>Evacuar o sistema como descrito na etapa 1.1.6.</li>
	<li>Desgas o pote de solvente como descrito na seção 2.2.</li>
	<li>Feche a válvula superior da ponte U para fechar a configuração de transferência de vácuo a partir do vácuo dinâmico. O sistema deve agora estar sob vácuo estático com a válvula Straus aberta e o adaptador solvente 180° fechado.</li>
	<li>Use uma tomada de laboratório para levantar um acetona/gelo seco de -78 °C para esfriar o frasco straus receptor.  ATENÇÃO: O resfriamento de um sistema fechado com nitrogênio líquido é perigoso, pois há uma chance significativa de condensar oxigênio líquido se houver um vazamento. Embora possa ser mais rápido para a transferência de vácuo a temperaturas de nitrogênio líquido, não vale a pena a troca em segurança. Essa prática é proibida em nossos laboratórios. Se um solvente não pode ser transferido a um -78 °C, é sugerido usar um vácuo melhor ou considerar a destilação térmica.
</li>
	<li>Ligue a agitação magnética para agitar a panela solvente, depois abra lentamente a torneira do adaptador de 180° na panela de solvente, de modo que o líquido na panela solvente não ferva rapidamente na articulação u.</li>
	<li>Em breve, o solvente deve começar a condensar no frasco receptor. Aguarde até que o pote de solvente esteja quase seco ou a quantidade desejada de solvente tenha sido coletada. </ol>4. Testando o solvente coletado em um porta-luvas de nitrogênio
1 Preparação da solução de ketyl
<ol><li>Pese 0,137 g de Ph2CO e mais 0,028 g de na metal em um frasco de 20 mL.</li>
	<li>Adicione 20 mL de THF ao frasco e uma pequena barra de rebuliço, depois tampe o frasco e mexa durante a noite. Isso deve gerar uma solução roxa profunda de 0,0337 M Na/Ph2CO ketyl radical com um pequeno excesso de Na permanecendo na parte inferior do frasco.</li>
</ol>2 Titulação do solvente coletado
<ol><li>Pipeta aproxima 4 mL do solvente a ser testado em um frasco de 4 mL.</li>
	<li>Use uma pipeta Pasteur para soltar cuidadosamente uma única gota de solução de THF de CO Na/Ph2roxo no frasco.</li>
	<li>Mexa suavemente o frasco com uma ponta de pipeta limpa e analise a cor resultante. Solvente que é 10 ppm H2O ou menos deve permanecer roxo pálido. (Nãoazul)
</li>
	<li>Se o solvente reagir com ph2CO radical (por exemplo, solventes halogenados como diclorometano), teste o solvente com titulação de Karl Fischer ou, no caso de solventes deuterados, espectrometria de 1,2,3H NMR.</li>

Linhas de schlenk e linhas de vácuo elevados são usadas para excluir umidade e oxigênio de reações, executando reações sob uma leve sobrepressão de gás inerte (geralmente N2 ou Ar) ou sob vácuo. A transferência de vácuo foi desenvolvida como um método separado de solventes (outros reagentes voláteis) de agentes de secagem (ou outros agentes não ativos) e os dispensa a vasos de reação ou armazenamento, mantendo um ambiente livre de ar. Certas reações químicas devem ser mantidas livres de água e oxigênio. Uma linha Schlenk é um coletor duplo usado no manuseio seguro de reagentes sensíveis ao ar e à umidade. Para começar, coloque os copos e reagentes em um porta-luvas sob atmosfera inerte. Meça aproximadamente um centímetro cúbico de metal de sódio e corte-o em pedaços menores. Coloque as peças em um balão de fundo redondo de 500 mL com uma junta de gargalo cônico padrão. Pesar 1,25 g de benzofenona e colocá-lo no balão de fundo redondo com o sódio. Enquanto mantém o nitrogênio borbulhando, abra lentamente o 180? adaptador para introduzir solvente no pote de solvente. Quando o nível de solvente no funil se aproximar do adaptador, feche o adaptador e remova o funil. Mexa a panela por várias horas. A solução ficará roxa profunda, indicando a formação do radical benzofenona cetil de sódio. A formação do radical significa que o solvente é seco e livre de oxigênio. Se a panela não ficar roxa profunda, desgaseifique a solução. Seque um frasco de recepção Straus de 500 mL e uma ponte de transferência a vácuo em forma de U em um forno de secagem. Um frasco Straus é um balão de fundo redondo de dois pescoços. Um pescoço é rosqueado para permitir a conexão de uma válvula macho. Cubra todas as juntas levemente com graxa a vácuo e prenda a ponte em forma de U à linha de vácuo. Aguarde até que o pote de solvente esteja quase seco ou até que a quantidade desejada de solvente tenha sido coletada. Feche a torneira do balão receptor e do pote de solvente. O balão selado agora pode ser removido do sistema. Para desligar o sistema, primeiro feche todas as portas do coletor e desligue o fluxo de gás inerte. Em seguida, remova a armadilha de solvente e os Dewars. Tome muito cuidado se algum líquido azul estiver presente na armadilha, pois pode ser oxigênio líquido. Schlenk lines and high vacuum lines are used to move solvents without exposing them to moisture or oxygen. They do this by keeping reactions under a slight overpressure of inert gas, usually nitrogen or argon, or by using vacuum.

Vacuum transfer is a separate method for isolating solvents and other volatile reagents from drying agents or other nonvolatile materials. It lets users dispense clean solvent into a reaction vessel or storage vessel while the air-free environment is maintained. The method works in a way that is similar to thermal distillation, because the solvent is vaporized and then condensed in a second receiving vessel.

A key advantage of vacuum transfer is that the low pressure in Schlenk and high vacuum line manifolds lowers boiling points to room temperature or even below. This makes cryogenic distillations possible. It can also be a safer alternative to thermal distillation when collecting air-free, moisture-free solvents.

After transfer, the collected solvent can be checked for water content. Common tests include Karl Fischer titration, titration with a Na/Ph2CO solution, and 1H NMR spectroscopy.

Reflux Setup for Heating Reactants Safely

A reflux setup lets chemists heat reactants without losing material to evaporation. Many chemical experiments need elevated temperatures before a reaction starts. When the reactants or solvent have low boiling points, heating can cause them to escape as vapor. A reflux system prevents that loss by condensing the vapor that forms during heating.
