Parkinson's disease (PD) is a common neurodegenerative disorder that results from the selective loss of midbrain dopaminergic neurons. Misfolding and aggregation of the protein alpha-synuclein, oxidative damage, and proteasomal impairment are all hypotheses for the molecular cause of this selective neurotoxicity. Here, we describe a Saccharomyces cerevisiae model to evaluate the misfolding, aggregation, and toxicity-inducing ability of wild-type alpha-synuclein and three mutants (A30P, A53T, and A30P/A53T), and we compare regulation of these properties by dysfunctional proteasomes and by oxidative stress. We found prominent localization of wild-type and A53T alpha-synuclein near the plasma membrane, supporting known in vitro lipid-binding ability. In contrast, A30P was mostly cytoplasmic, whereas A30P/A53T displayed both types of fluorescence. Surprisingly, alpha-synuclein was not toxic to several yeast strains tested. When yeast mutants for the proteasomal barrel (doa3-1) were evaluated, delayed alpha-synuclein synthesis and membrane association were observed; yeast mutant for the proteasomal cap (sen3-1) exhibited increased accumulation and aggregation of alpha-synuclein. Both sen3-1and doa3-1 mutants exhibited synthetic lethality with alpha-synuclein. When yeasts were challenged with an oxidant (hydrogen peroxide), alpha-synuclein was extremely lethal to cells that lacked manganese superoxide dismutase Mn-SOD (sod2Delta) but not to cells that lacked copper, zinc superoxide dismutase Cu,Zn-SOD (sod1Delta). Despite the toxicity, sod2Delta cells never displayed intracellular aggregates of alpha-synuclein. We suggest that the toxic alpha-synuclein species in yeast are smaller than the visible aggregates, and toxicity might involve alpha-synuclein membrane association. Thus, yeasts have emerged effective organisms for characterizing factors and mechanisms that regulate alpha-synuclein toxicity.

Ici, les auteurs d&eacute;crivent le mod&egrave;le pour &eacute;valuer le mauvais repliement, agr&eacute;gation, Saccharomyces cerevisiae et de capacit&eacute; induisant la toxicit&eacute; de l&#39;alpha-synucl&eacute;ine de type sauvage et trois mutants (A30P, A53T et A30P/A53T) et nous comparer r&egrave;glement de ces propri&eacute;t&eacute;s prot&eacute;asomes dysfonctionnelle et par le stress oxydatif. Les deux mutants de doa3-1 sen3-1 et pr&eacute;sentaient une l&eacute;talit&eacute; synth&eacute;tique avec alpha-synucl&eacute;ine. Lorsque les levures ont &eacute;t&eacute; contest&eacute;es avec un oxydant (eau oxyg&eacute;n&eacute;e), alpha-synucl&eacute;ine &eacute;tait extr&ecirc;mement l&eacute;tal pour les cellules qui manquaient de dismutase de superoxyde de Mangan&egrave;se Mn-SOD (sod2Delta), mais pas aux cellules qui manquait de cuivre, zinc superoxyde dismutase Cu, Zn-SOD (sod1Delta). Malgr&eacute; la toxicit&eacute;, les cellules sod2Delta jamais affichent agr&eacute;gats intracellulaires de l&#39;alpha-synucl&eacute;ine. Hier beschreiben wir eine Saccharomyces-Cerevisiae-Modell zur Bewertung der misfolding, Aggregation, und Toxizit&auml;t-induzierende F&auml;higkeit von Wildtyp Alpha-Synuclein und drei Mutanten (A30P, A53T und A30P/A53T), und wir vergleichen Verordnung dieser Eigenschaften durch gest&ouml;rte Proteasomes und durch oxidativen Stress. Wir fanden prominente Lokalisierung von Wildtyp und A53T Alpha-Synuclein in der N&auml;he von der Plasmamembran bekannte in-vitro-Lipid-Bindung F&auml;higkeit zu unterst&uuml;tzen. Beide sen3-1und doa3-1 Mutanten ausgestellt synthetischen Letalit&auml;t mit Alpha-Synuclein. Wenn Hefen mit einem Oxidationsmittel (Wasserstoffperoxid) in Frage gestellt wurden, Alpha-Synuclein war extrem t&ouml;dlich auf Zellen, die Mangan-Superoxid-Dismutase Mn-SOD (sod2Delta) fehlten, aber nicht auf Zellen, die fehlte, Kupfer, Zink-Superoxid-Dismutase Cu, Zn-SOD (sod1Delta). Trotz der Toxizit&auml;t angezeigt sod2Delta Zellen nie intrazellul&auml;re Aggregate von Alpha-Synuclein. Wir schlagen vor, da&szlig; die toxische Alpha-Synuclein-Arten in der Hefe kleiner als die sichtbaren Aggregate sind und Toxizit&auml;t z. Alpha-Synuclein-Membran-Verband b. wird. So entstanden die Hefen wirksame Organismen f&uuml;r die Charakterisierung von Faktoren und Mechanismen, die Alpha-Synuclein Toxizit&auml;t zu regulieren.

Enfermedad de Parkinson (EP) es una neurodegeneraci&oacute;n com&uacute;n que resulta de la p&eacute;rdida selectiva de las neuronas dopamin&eacute;rgicas de mesenc&eacute;falo. Mal plegamiento y agregaci&oacute;n de la prote&iacute;na alfa-sinucle&iacute;na, da&ntilde;o oxidativo y deterioro proteasomal son todas las hip&oacute;tesis para la causa molecular de esta neurotoxicidad selectiva. Aqu&iacute;, describimos un modelo de Saccharomyces cerevisiae para evaluar el mal plegamiento, agregaci&oacute;n, y capacidad inductora de toxicidad de tipo alfa-sinucle&iacute;na y tres mutantes (A30P, A53T y A30P/A53T) y comparar la regulaci&oacute;n de estas propiedades proteasomas disfuncional y estr&eacute;s oxidativo. Encontramos localizaci&oacute;n prominente de tipo salvaje y A53T alfa-sinucle&iacute;na, cerca de la membrana plasm&aacute;tica, apoyando la conocida capacidad de uni&oacute;n a l&iacute;pidos in vitro. Por el contrario, A30P era sobre todo citopl&aacute;smico, mientras que A30P/A53T muestran ambos tipos de fluorescencia. Sorprendentemente, alfa-sinucle&iacute;na no era t&oacute;xico para varias cepas de levadura probadas. Cuando mutantes de la levadura para el ca&ntilde;&oacute;n proteasomal (doa3-1) fueron evaluados, retrasada alfa-sinucle&iacute;na s&iacute;ntesis y membrana Asociaci&oacute;n fueron observados; mutantes de la levadura para la tapa proteasomal (sen3-1) exhibieron mayor acumulaci&oacute;n y agregaci&oacute;n de alfa-sinucle&iacute;na. Genetic defects in the dystrophin-associated protein complex (DAPC) are responsible for a variety of pathological conditions including muscular dystrophy, cardiomyopathy, and vasospasm. Conserved DAPC components from humans to Caenorhabditis elegans suggest a similar molecular function. C. elegans DAPC mutants exhibit a unique locomotory deficit resulting from prolonged muscle excitation and contraction. Here we show that the C. elegans DAPC is essential for proper localization of SLO-1, the large conductance, voltage-, and calcium-dependent potassium (BK) channel, which conducts a major outward rectifying current in muscle under the normal physiological condition. Through analysis of mutants with the same phenotype as the DAPC mutants, we identified the novel islo-1 gene that encodes a protein with two predicted transmembrane domains. We demonstrate that ISLO-1 acts as a novel adapter molecule that links the DAPC to SLO-1 in muscle. We show that a defect in either the DAPC or ISLO-1 disrupts normal SLO-1 localization in muscle. Consistent with observations that SLO-1 requires a high calcium concentration for full activation, we find that SLO-1 is localized near L-type calcium channels in muscle, thereby providing a mechanism coupling calcium influx with the outward rectifying current. Our results indicate that the DAPC modulates muscle excitability by localizing the SLO-1 channel to calcium-rich regions of C. elegans muscle.

Most human genes contain multiple alternative splice sites believed to extend the complexity and diversity of the proteome. However, little is known about how interactions among alternative exons regulate protein function. We used the Caenorhabditis elegans slo-1 large-conductance calcium and voltage-activated potassium (BK) channel gene, which contains three alternative splice sites (A, B, and C) and encodes at least 12 splice variants, to investigate the functional consequences of alternative splicing. These splice sites enable the insertion of exons encoding part of the regulator of K(+) conductance (RCK)1 Ca(2+) coordination domain (exons A1 and A2) and portions of the RCK1-RCK2 linker (exons B0, B1, B2, C0, and C1). Exons A1 and A2 are used in a mutually exclusive manner and are 67% identical. The other exons can extend the RCK1-RCK2 linker by up to 41 residues. Electrophysiological recordings of all isoforms show that the A1 and A2 exons regulate activation kinetics and Ca(2+) sensitivity, but only if alternate exons are inserted at site B or C. Thus, RCK1 interacts with the RCK1-RCK2 linker, and the effect of exon variation on gating depends on the combination of alternate exons present in each isoform.

Alternatively spliced domains interact to regulate BK potassium channel gating.

Exons A1 et A2 sont utilis&#xE9;s d&#x26;#39;une mani&#xE8;re mutuellement exclusive et sont identiques 67%. Alternative splicing is critical for diversifying eukaryotic proteomes, but the rules governing and coordinating splicing events among multiple alternate splice sites within individual genes are not well understood. We developed a quantitative PCR-based strategy to quantify the expression of the 12 transcripts encoded by the Caenorhabditis elegans slo-1 gene, containing three alternate splice sites. Using conditional probability-based models, we show that splicing events are coordinated across these sites. Further, we identify a point mutation in an intron adjacent to one alternate splice site that disrupts alternative splicing at all three sites. This mutation leads to aberrant synaptic transmission at the neuromuscular junction. In a genomic survey, we found that a UAAAUC element disrupted by this mutation is enriched in introns flanking alternate exons in genes with multiple alternate splice sites. These results establish that proper coordination of intragenic alternative splicing is essential for normal physiology of slo-1 in vivo and identify putative specialized cis-regulatory elements that regulate the coordination of intragenic alternative splicing.

Alternatives Splei&szlig;en ist entscheidend f&uuml;r die Diversifizierung der eukaryotischen Proteome, aber die Regeln, die EZB und die Koordination der Splei&szlig;en Veranstaltungen unter mehreren Alternativen Splei&szlig;-Standorten innerhalb einzelner Gene sind nicht gut verstanden. Wir entwickelt eine quantitative PCR-basierte Strategie zur Quantifizierung der Ausdruck der 12 Abschriften, die durch den Fadenwurm Caenorhabditis Elegans Slo-1-Gen kodiert, enth&auml;lt drei alternative Splice-Sites. Bedingte Wahrscheinlichkeit basierende Modelle verwenden, zeigen wir, dass Splei&szlig;en Veranstaltungen &uuml;ber diese Standorte hinweg koordiniert werden. Dar&uuml;ber hinaus identifizieren wir eine Punktmutation in einem Intron grenzte an einen alternativen Splei&szlig;-Platz, das alternative Splei&szlig;en an allen drei Standorten st&ouml;rt. Splicing alternativo &egrave; fondamentale per diversificare ai proteomi eucariotici, ma le norme che disciplinano e coordinare eventi tra pi&ugrave; siti di splicing alternativo all&#39;interno di singoli geni di splicing non sono ben comprese. Abbiamo sviluppato una strategia basata su PCR quantitativa per quantificare l&#39;espressione di 12 trascrizioni codificata dal gene slo-1 Caenorhabditis elegans, contenente tre siti di splicing alternativo. Splicing alternativo es fundamental para la diversificaci&#xF3;n de proteomas eucariotas, pero las normas que rigen y coordinaci&#xF3;n de eventos de empalme entre los m&#xFA;ltiples sitios de empalme alternativo dentro de los genes individuales no se comprenden bien. Se desarroll&#xF3; una PCR cuantitativa basada en la estrategia para cuantificar la expresi&#xF3;n de los 12 transcripciones codificadas por el Caenorhabditis elegans SLO gen-1, que contiene tres sitios de empalme alternativo. Uso de condicionales basados &#x200B;&#x200B;en modelos de probabilidad, nos muestran que los eventos de splicing se coordinan a trav&#xE9;s de estos sitios. Adem&#xE1;s, se identifica una mutaci&#xF3;n puntual en un intr&#xF3;n adyacente a un sitio de empalme alternativo que rompe splicing alternativo en los tres sitios. Esta mutaci&#xF3;n da lugar a la transmisi&#xF3;n sin&#xE1;ptica aberrante en la uni&#xF3;n neuromuscular.
