Eine Zeit, die Anspritzung System aufgebaut, das Hohe Aufnahmequalit&auml;t kann Raman-Spektren von stark fluoreszierende biologischen Proben w&auml;hrend des Betriebs unterhalb der Schwellenwerts von Lichtsch&auml;den. Mit einem kollinear gating Geometrie und Sorgfalt zur Erhaltung von macht, haben wir rein optischen Wechsel eine Pikosekunde gating einmal mit 5 %-Spitze Anspritzung Effizienz erreicht. Die Energie pro Impuls in diesem Instrument ist mehr als 3 Gr&ouml;&szlig;enordnungen schw&auml;cher als die vorherigen Berichte. Mit diesem System haben wir auf einer Stichprobe bestehend aus perylen gel&ouml;st in Toluol und die stengel der Pflanze Jasminum Multiflorum, letzteren Fall besonders wichtig f&uuml;r das Studium der Pflanzen, die Herstellung von Zellulose Biokraftstoffen Proof-of-Concept-Versuche durchgef&uuml;hrt. In beiden F&auml;llen wurde ein hoher SNR-Spektrum der hoch-die Wellenzahl Region des Spektrums in der Gegenwart eine &uuml;berw&auml;ltigende Fluoreszenz im Hintergrund aufgenommen.

Entwicklung eines Zeit-Gating f&uuml;r Raman-Spektroskopie von biologischen Proben.

&Egrave; stato costruito un tempo gating sistema che &egrave; in grado di registrare ad alta qualit&agrave; spettri Raman di campioni biologici altamente fluorescenti durante il funzionamento di sotto della soglia di fotodanneggiamento. Usando una geometria controllo collineare e particolare attenzione al risparmio energetico, abbiamo raggiunto all-optical switching con un una volta controllo picosecondo e 5% picco gating efficienza. L&#39;energia per impulso in questo strumento &egrave; pi&ugrave; di 3 ordini di grandezza pi&ugrave; deboli rispetto a precedenti relazioni. Con questo sistema abbiamo eseguito esperimenti di prova su un campione composto di perilene sciolto in toluene e il gambo di una pianta di Jasminum multiflorum, quest&#39;ultimo caso &egrave; particolarmente importante per lo studio delle piante utilizzate nella produzione di biocarburanti cellulosici. In entrambi i casi, &egrave; stato registrato un elevato spettro SNR della regione alta-numero d&#39;onda dello spettro in presenza di un&#39;opprimente sfondo di fluorescenza.

Un sistema de llenado tiempo se ha construido de forma que es capaz de registrar los espectros de alta calidad de Raman altamente fluorescentes muestras biol&#xF3;gicas mientras se opera por debajo del umbral de da&#xF1;o solar. Utilizando una geometr&#xED;a de puerta alineados, y la atenci&#xF3;n a la conservaci&#xF3;n de energ&#xED;a, hemos logrado todas las &#xF3;pticas de conmutaci&#xF3;n con un tiempo de puerta un picosegundo y el 5% de eficiencia pico de puerta. La energ&#xED;a de un impulso en este instrumento es de m&#xE1;s de 3 &#xF3;rdenes de magnitud m&#xE1;s d&#xE9;bil que los informes anteriores. Utilizando este sistema, hemos realizado la prueba de concepto experimentos sobre una muestra compuesta de perileno disolvi&#xF3; en tolueno, y el tallo de una planta Jasminum multiflorum, el &#xFA;ltimo caso es particularmente importante para el estudio de las plantas utilizadas en la producci&#xF3;n de biocombustibles celul&#xF3;sicos. En ambos casos, un alto espectro SNR de la regi&#xF3;n de alto n&#xFA;mero de onda del espectro se registr&#xF3; en la presencia de un fondo de fluorescencia abrumador.

A time gating system has been constructed that is capable of recording high quality Raman spectra of highly fluorescing biological samples while operating below the photodamage threshold. Using a collinear gating geometry and careful attention to power conservation, we have achieved all-optical switching with a one picosecond gating time and 5% peak gating efficiency. The energy per pulse in this instrument is more than 3 orders of magnitude weaker than previous reports. Using this system we have performed proof-of-concept experiments on a sample composed of perylene dissolved in toluene, and the stem of a Jasminum multiflorum plant, the latter case being particularly important for the study of plants used in production of cellulosic biofuels. In both cases, a high SNR spectrum of the high-wavenumber region of the spectrum was recorded in the presence of an overwhelming fluorescence background.

Un syst&#xE8;me de d&#xE9;clenchement de temps a &#xE9;t&#xE9; construit qui est capable d&#x26;#39;enregistrer des spectres Raman de qualit&#xE9; tr&#xE8;s fluorescent &#xE9;chantillons biologiques pendant le fonctionnement en dessous du seuil photodommage. En utilisant une g&#xE9;om&#xE9;trie de d&#xE9;clenchement colin&#xE9;aires et une attention particuli&#xE8;re &#xE0; la conservation d&#x26;#39;&#xE9;nergie, nous avons r&#xE9;alis&#xE9; tout-optique de commutation avec un temps de d&#xE9;clenchement d&#x26;#39;une picoseconde et 5% l&#x26;#39;efficacit&#xE9; de d&#xE9;clenchement de pointe. L&#x26;#39;&#xE9;nergie par impulsion dans cet instrument est plus de 3 ordres de grandeur plus faible que les pr&#xE9;c&#xE9;dents rapports. Gr&#xE2;ce &#xE0; ce syst&#xE8;me, nous avons effectu&#xE9; une preuve de concept des exp&#xE9;riences sur un &#xE9;chantillon compos&#xE9; de p&#xE9;ryl&#xE8;ne dissous dans du tolu&#xE8;ne, et la tige d&#x26;#39;une plante Jasminum multiflorum, le dernier cas &#xE9;tant particuli&#xE8;rement importante pour l&#x26;#39;&#xE9;tude des plantes utilis&#xE9;es dans la production de biocarburants cellulosiques. Dans les deux cas, un spectre de haute SNR de la r&#xE9;gion haute-nombre d&#x26;#39;ondes du spectre a &#xE9;t&#xE9; enregistr&#xE9;e en pr&#xE9;sence d&#x26;#39;une base &#xE9;crasante de fluorescence.

This paper presents an endoscopic configuration for measurements of tissue autofluorescence using two-photon excitation and time-correlated single photon counting detection through a double-clad photonic crystal fiber (DC-PCF) without pre-chirping of laser pulses. The instrument performance was evaluated by measurements of fluorescent standard dyes, biological fluorophores (collagen and elastin), and tissue specimens (muscle, cartilage, tendon). Current results demonstrate the ability of this system to accurately retrieve the fluorescence decay profile and lifetime of these samples. This simple setup, which offers larger penetration depth than one-photon-based techniques, may be combined with morphology-yielding techniques such as photoacoustic and ultrasound imaging.
