Rapid innate immune defences against infection usually involve the recognition of invading pathogens by specific pattern recognition receptors recently attributed to the family of Toll-like receptors (TLR). TLRs constitute a major part of innate immune system, and have been characterised in different tissues and organs. Reports from our laboratory and others have demonstrated the existence of TLRs in the female reproductive tract, yet TLRs have not been explored completely in this system. There is little known about variation in TLR expression during the menstrual cycle and the effects that sex hormones may have on their expression and function. Here, we review recent information regarding the existence of TLRs in the female reproductive tract, their function in the maintenance of innate immune system in this tract and their potential role in pregnancy.

Schnelle angeborenen Immunabwehr gegen Infektionen beinhalten in der Regel die Anerkennung der eindringenden Krankheitserreger von bestimmten Muster-Anerkennung-Rezeptoren, die vor kurzem die Familie von Toll-Like Rezeptoren (TLR) zugeschrieben. TLR stellen einen wichtigen Teil des angeborenen Immunsystems und haben in verschiedenen Geweben und Organen gekennzeichnet. Berichte aus unserem Labor und andere zeigten die Existenz der TLRs in der weiblichen Fortpflanzungsorgane, dennoch TLRs haben in diesem System nicht v&ouml;llig erforscht worden. Es ist wenig bekannt &uuml;ber Variation in TLR Ausdruck w&auml;hrend des Menstruationszyklus und die Effekte, die Sexualhormone auf ihren Ausdruck und Funktion haben kann. Hier schauen wir uns den letzten Informationen &uuml;ber die Existenz der TLRs in der weiblichen Fortpflanzungsorgane, ihre Funktion bei der Aufrechterhaltung der angeborenen Infektionsabwehr in diesem Trakt und ihre m&ouml;gliche Rolle in der Schwangerschaft.

Report dal nostro laboratorio e altri hanno dimostrato l&#39;esistenza di TLR nel tratto riproduttivo femminile, ancora TLR non sono state completamente esplorate in questo sistema. TLRs constituyen una parte importante del sistema inmune innato y han sido caracterizados en &oacute;rganos y tejidos diferentes. Heterotrimeric G proteins play a key role in membrane-mediated cell-signalling and hormonal regulation. Our earlier studies gave evidence of G protein subunit Galpha(i2) being under hormonal regulation in human in vivo. In this study, we used immortalized human oviduct epithelial cell line OE-E6/E7 as a model to study the hormonal regulation of Galpha(i2). We aimed at clarifying whether estradiol or progesterone could individually regulate the expression of Galpha(i2) and its potential signalling partners. Furthermore, we aimed to investigate which sex hormone receptors could potentially mediate the gene regulation in OE-E6/E7 cell line. OE-E6/E7 cells were cultured for 5 days with different concentrations of estradiol or progesterone. Quantitative real-time polymerase chain reaction (Q-PCR) was performed using cDNA of the hormone-treated cells to reveal any changes in gene expression. The presence of potential receptor targets in these cells was studied using PCR. Our data clearly showed that low concentrations of estradiol up-regulated the expression of Galpha(i2) gene and down-regulated the expression of membrane progesterone receptor mPRalpha gene in OE-E6/E7 cell line. Progesterone had no significant effect on Galpha(i2) gene expression, but it caused up-regulation of mPRalpha gene expression. In conclusion, it appears that sex hormones regulate the expression of Galpha(i2) and mPRalpha genes in a reverse manner in OE-E6/E7 cells. Our results suggest that estrogen receptor ERbeta mediates the regulatory effects of estradiol in these cells.

Nostri studi precedenti ha dato prova di proteina G subunit&agrave; Galpha(i2) essere sotto regolazione ormonale in umani in vivo. In questo studio, abbiamo usato immortalata ovidotto umano delle cellule epiteliali linea OE-E6/E7 come un modello per studiare la regolazione ormonale di Galpha(i2). Abbiamo puntato chiarire se estradiolo o progesterone potrebbe regolare individualmente l&#39;espressione di Galpha(i2) e i suoi potenziali partner di segnalamento. Inoltre, abbiamo mirato a indagare quali recettori ormone sessuale potenzialmente potrebbero mediare la regolazione genica nella linea cellulare OE-E6/E7. In conclusione, sembra che gli ormoni sessuali regolano l&#39;espressione dei geni Galpha(i2) e mPRalpha in maniera inversa in cellule OE-E6/E7. I nostri risultati suggeriscono che estrogeno recettore ERbeta media gli effetti normativi dell&#39;estradiolo in queste cellule.

Prote&iacute;nas heterotrim&eacute;ricas G juegan un papel clave en la se&ntilde;alizaci&oacute;n celular mediada por membrana y regulaci&oacute;n hormonal. Nuestros estudios anteriores dieron pruebas de G prote&iacute;na subunidad Galpha(i2) ser bajo regulaci&oacute;n hormonal en humanos in vivo. En este estudio, utilizamos inmortalizadas oviducto humano c&eacute;lula epitelial l&iacute;nea OE-E6/E7 como modelo para estudiar la regulaci&oacute;n hormonal de la Galpha(i2). Apuntamos a clarificar si el estradiol o la progesterona individualmente podr&iacute;a regular la expresi&oacute;n de Galpha(i2) y sus potenciales socios de se&ntilde;alizaci&oacute;n. Adem&aacute;s, decidimos investigar qu&eacute; receptores de hormonas sexuales potencialmente podr&iacute;an mediar la regulaci&oacute;n g&eacute;nica en la l&iacute;nea celular de OE-E6/E7. OE-E6/E7 las c&eacute;lulas fueron cultivadas por 5 d&iacute;as con diferentes concentraciones de estradiol o la progesterona. La reacci&oacute;n en cadena de la polimerasa en tiempo real cuantitativa (Q-PCR) fue realizada usando cDNA de las c&eacute;lulas de hormonas para revelar las alteraciones en la expresi&oacute;n g&eacute;nica. Se estudi&oacute; la presencia de objetivos potenciales receptores en estas c&eacute;lulas mediante PCR. Nuestros datos demostraron claramente que bajas concentraciones de estradiol para arriba-regulaci&oacute;n la expresi&oacute;n del gen Galpha(i2) y regulaci&oacute;n la expresi&oacute;n del gene en la l&iacute;nea celular de OE-E6/E7 mPRalpha del receptor de membrana progesterona. Progesterona no tuvo efectos significativos sobre la expresi&oacute;n g&eacute;nica de Galpha(i2), pero caus&oacute; para arriba-regulaci&oacute;n de la expresi&oacute;n g&eacute;nica de mPRalpha. En conclusi&oacute;n, parece que las hormonas sexuales regulan la expresi&oacute;n de los genes Galpha(i2) y mPRalpha de manera inversa en las c&eacute;lulas de la OE-E6/E7. Nuestros resultados sugieren que eso receptor de estr&oacute;geno ERbeta media los efectos reglamentarios de estradiol en estas c&eacute;lulas.

The notion of a gamete recognition system that alerts females to the presence of gametes in their reproductive tract profoundly influences our understanding of the physiology of events leading to conception and the bearing of offspring. Here, we show that the female responds to gametes within her tract by modulating the environment in which pregnancy is initially established. We found distinct alterations in oviductal gene expression as a result of sperm and oocyte arrival in the oviduct, which led directly to distinct alterations to the composition of oviductal fluid in vivo. This suggests that either gamete activates a cell-type-specific signal transduction pathway within the oviduct. This gamete recognition system presents a mechanism for immediate and local control of the oviductal microenvironment in which sperm transport, sperm binding and release, capacitation, transport of oocytes, fertilization, and early cleavage-stage embryonic development occur. This may explain the mechanisms involved in postcopulatory sexual selection, where there is evidence suggesting that the female reproductive tract can bias spermatozoa from different males in the favour of the more biologically attractive male. In addition, the presence of a gamete recognition system explains the oviduct's ability to tolerate spermatozoa while remaining intolerant to pathogens.

In mammals, Toll-like receptors (TLRs) are the principal family of innate immune pattern recognition receptors (PRRs). The main function for TLRs is the detection of molecular patterns associated with invading pathogens. We investigated TLR expression and function in three established human endometrial epithelial cell lines, including hTERT-EEC, HEC-1B and Ishikawa cells, and clarified the application of these endometrial cell lines as in vitro models for studying TLR expression and function in the female reproductive tract. TLR gene expression was examined by RT-PCR and protein localization by immunohistochemistry. Our results showed that TLR expression in these cell lines is comparable to published literature on TLR expression in primary human endometrial tissue. TLR function was investigated by the detection of IL-6 and IL-8 production by ELISA in response to TLR2, TLR3, TLR5, TLR7 and TLR9 ligands. We found that hTERT-EEC cells were responsive to TLR5 ligand and HEC-1B cells respond to TLR3 and TLR5 ligands. In contrast, Ishikawa cells respond only to PMA/I which was used as a positive control for IL-8 production. Finally, we investigated the influence of flagellin as a TLR5 stimulant on TLR5 expression in these cell lines by QPCR. Our results showed that the endometrial cell lines showed a tendency for increased TLR5 expression in response to flagellin stimulation and in hTERT-EEC cells this tendency was statistically significant. These results suggest that hTERT-EEC, HEC-1B and Ishikawa cell lines can be used as in vitro models to investigate innate immune responses of endometrial cells in the female reproductive tract.

Die wichtigste Funktion f&uuml;r TLRs ist die Erkennung von molekularen Mustern, die eindringenden Krankheitserregern zugeordnet. Wir untersuchten TLR-Expression und Funktion in drei etablierten humane endometrial Epithelzellen Linien, einschlie&szlig;lich hTERT-EWG, HEC-1B und Ishikawa-Zellen, und verdeutlichte die Anwendung diese endometrial Zelllinien in Vitro Modelle zur Untersuchung der TLR-Expression und Funktion in der weiblichen Fortpflanzungsorgane. TLR-Genexpression wurde durch RT-PCR und Protein-Lokalisierung durch Immunhistochemie untersucht. Nei mammiferi, recettori Toll-like (TLR) sono la principale famiglia di recettori di riconoscimento di pattern immunitario innato (PRRs). La funzione principale per TLR &egrave; l&#39;individuazione di pattern molecolari associati a patogeni. En mam&iacute;feros, receptores Toll-like (TLRs) son la principal familia de receptores de reconocimiento de patr&oacute;n inmune innata (PRRs). La funci&oacute;n principal de TLRs es la detecci&oacute;n de patrones moleculares asociados a pat&oacute;genos invasores. Investigamos TLR expresi&oacute;n y funci&oacute;n en tres l&iacute;neas de c&eacute;lulas epiteliales endometrial humana establecidas, incluyendo hTERT-CEE, HEC-1B y c&eacute;lulas de Ishikawa y clarifica la aplicaci&oacute;n de estas l&iacute;neas de c&eacute;lulas endometriales como en vitro modelos para el estudio de expresi&oacute;n TLR y funci&oacute;n en el tracto reproductor femenino. Expresi&oacute;n g&eacute;nica TLR fue examinado por RT-PCR y prote&iacute;na de localizaci&oacute;n mediante inmunohistoqu&iacute;mica. Nuestros resultados mostraron que expresi&oacute;n TLR en estas l&iacute;neas celulares es comparable a la literatura publicada sobre expresi&oacute;n TLR en primario del tejido endometrial humano. Funci&oacute;n TLR fue investigada por la detecci&oacute;n de la producci&oacute;n de IL-6 e IL-8 por ELISA en respuesta a ligandos TLR2, TLR3, TLR5, TLR7 y TLR9. Se encontr&oacute; que las c&eacute;lulas hTERT-CEE fueron sensibles a TLR5 ligando y las c&eacute;lulas de HEC-1B responden a ligandos TLR3 y TLR5. En contraste, las c&eacute;lulas de Ishikawa responden s&oacute;lo a PMA / I que se utiliz&oacute; como control positivo para la producci&oacute;n de IL-8. Finalmente, se investig&oacute; la influencia de flagelina como estimulante TLR5 TLR5 expresi&oacute;n en estas l&iacute;neas celulares por QPCR. Nuestros resultados mostraron que las l&iacute;neas de c&eacute;lulas endometriales demostraron una tendencia de aumento de la expresi&oacute;n TLR5 en respuesta a la estimulaci&oacute;n de flagelina y en c&eacute;lulas hTERT-CEE esta tendencia fue estad&iacute;sticamente significativa. Estos resultados sugieren que hTERT-CEE, HEC-1B y l&iacute;neas celulares de Ishikawa pueden utilizarse como en vitro modelos para investigar innata respuesta inmune de las c&eacute;lulas del endometrio en el tracto reproductor femenino.
