Alzheimer's disease (AD) is an age-related progressive neurodegenerative disease commonly found among elderly. In addition to cognitive and behavioral deficits, vision abnormalities are prevalent in AD patients. Recent studies investigating retinal changes in AD double-transgenic mice have shown altered processing of amyloid precursor protein and accumulation of Î²-amyloid peptides in neurons of retinal ganglion cell layer (RGCL) and inner nuclear layer (INL). Apoptotic cells were also detected in the RGCL. Thus, the pathophysiological changes of retinas in AD patients are possibly resembled by AD transgenic models. The retina is a simple model of the brain in the sense that some pathological changes and therapeutic strategies from the retina may be observed or applicable to the brain. Furthermore, it is also possible to advance our understanding of pathological mechanisms in other retinal degenerative diseases. Therefore, studying AD-related retinal degeneration is a promising way for the investigation on (1) AD pathologies and therapies that would eventually benefit the brain and (2) cellular mechanisms in other retinal degenerations such as glaucoma and age-related macular degeneration. This review will highlight the efforts on retinal degenerative research using AD transgenic mouse models.

Neurodegeneration of the retina in mouse models of Alzheimer's disease: what can we learn from the retina?

La maladie d&#x26;#39;Alzheimer (AD) est une li&#xE9;e &#xE0; l&#x26;#39;&#xE2;ge maladie neurod&#xE9;g&#xE9;n&#xE9;rative progressive trouve couramment chez les personnes &#xE2;g&#xE9;es. En plus de d&#xE9;ficits cognitifs et comportementaux, troubles de la vision sont fr&#xE9;quents chez les patients AD. Des &#xE9;tudes r&#xE9;centes modifications de la r&#xE9;tine de l&#x26;#39;enqu&#xEA;te dans AD double-souris transg&#xE9;niques ont montr&#xE9; la transformation modifi&#xE9;e de la prot&#xE9;ine pr&#xE9;curseur amylo&#xEF;de et l&#x26;#39;accumulation de &#x3B2;-amylo&#xEF;des peptides dans les neurones de la couche des cellules ganglionnaires r&#xE9;tiniennes (RGCL) et la couche int&#xE9;rieure nucl&#xE9;aire (INL). Les cellules apoptotiques ont &#xE9;galement &#xE9;t&#xE9; d&#xE9;tect&#xE9;s dans le RGCL. Ainsi, les modifications physiopathologiques de la r&#xE9;tine chez les patients AD sont peut-&#xEA;tre ressemblait par des mod&#xE8;les transg&#xE9;niques AD. La r&#xE9;tine est un mod&#xE8;le simple du cerveau dans le sens que certains changements pathologiques et des strat&#xE9;gies th&#xE9;rapeutiques de la r&#xE9;tine peuvent &#xEA;tre observ&#xE9;es ou applicable au cerveau. En outre, il est &#xE9;galement possible de faire progresser notre compr&#xE9;hension des m&#xE9;canismes pathologiques dans d&#x26;#39;autres maladies de d&#xE9;g&#xE9;n&#xE9;rescence r&#xE9;tinienne. Par cons&#xE9;quent, l&#x26;#39;&#xE9;tude AD li&#xE9;e d&#xE9;g&#xE9;n&#xE9;rescence r&#xE9;tinienne est une voie prometteuse pour l&#x26;#39;enqu&#xEA;te sur les pathologies AD (1) et les th&#xE9;rapies qui finirait par profiter du cerveau et (2) les m&#xE9;canismes cellulaires dans d&#x26;#39;autres d&#xE9;g&#xE9;n&#xE9;rescences r&#xE9;tiniennes telles que le glaucome et la d&#xE9;g&#xE9;n&#xE9;rescence maculaire li&#xE9;e &#xE0; l&#x26;#39;&#xE2;ge. Neurod&#xE9;g&#xE9;n&#xE9;rescence de la r&#xE9;tine dans les mod&#xE8;les murins de la maladie d&#x26;#39;Alzheimer: Que pouvons-nous savoir de la r&#xE9;tine?

Alzheimer-Krankheit (AD) ist eine altersbedingte progressive Neurodegenerative Krankheit, die h&auml;ufig bei &auml;lteren Menschen gefunden. Neben kognitiven und verhaltensbezogenen Defizite sind Vision Anomalien bei AD-Patienten weit verbreitet. J&uuml;ngste Studien untersuchen retinale Ver&auml;nderungen im AD Doppel-transgene M&auml;use haben ver&auml;nderte Verarbeitung von Amyloid-Vorl&auml;uferprotein und Anh&auml;ufung von &beta;-Amyloid-Peptide in Neuronen der retinalen Ganglienzellen Zellschicht (RGCL) und nukleare Innenschicht (INL) gezeigt. Apoptotic Zellen wurden auch bei der RGCL festgestellt. Somit sind die pathophysiologischen Ver&auml;nderungen der Netzhaut bei AD-Patienten m&ouml;glicherweise von AD transgenen Modellen &auml;hnelte. Die Netzhaut ist ein einfaches Modell des Gehirns in dem Sinne, dass einige pathologische Ver&auml;nderungen und therapeutische Strategien aus der Netzhaut beobachtet werden k&ouml;nnen oder auf das Gehirn. Dar&uuml;ber hinaus ist es auch m&ouml;glich, unser Verst&auml;ndnis der pathologischen Mechanismen in anderen degenerativen Netzhauterkrankungen voranzutreiben. Studium AD-bezogene Netzhautdegeneration ist daher eine vielversprechende M&ouml;glichkeit zur Untersuchung auf (1) AD Krankheiten und Therapien, die schlie&szlig;lich die Gehirn und (2) zellul&auml;re Mechanismen in anderen retinalen Degenerationen wie Glaukom und die altersbedingte Makula-Degeneration profitieren w&uuml;rden. Diese Bewertung wird die Anstrengungen auf die Netzhaut degenerative Forschung unter Verwendung von AD transgener Mausmodelle hervorheben.

Oltre ai deficit cognitivi e comportamentali, sono prevalenti in pazienti AD anomalie di visione. Recenti studi indagando i cambiamenti della retina nei topi transgenici doppio annuncio hanno dimostrato alterata elaborazione della proteina precursore dell&#39;amiloide e accumulo di peptidi &beta;-amiloide nei neuroni di strato delle cellule gangliari della retina (RGCL) e strato nucleare interno (INL). Il RGCL sono state individuate anche le cellule apoptotic. Cos&igrave;, i cambiamenti patofisiologici delle retine in pazienti AD sono forse somigliava da modelli transgenici AD. La retina &egrave; un semplice modello del cervello nel senso che possono essere osservate alcune alterazioni patologiche e strategie terapeutiche dalla retina o applicabile al cervello. Inoltre, &egrave; anche possibile avanzare la nostra comprensione dei meccanismi patologici in altre malattie degenerative della retina. Pertanto, studiando la degenerazione retinica correlati AD &egrave; un modo promettente per l&#39;inchiesta su (1) AD patologie e terapie che gioverebbe alla fine i meccanismi cerebrali e cellulari (2) in altre degenerazioni retiniche come il glaucoma e la degenerazione maculare legata all&#39;et&agrave;. La enfermedad de Alzheimer (EA) es una relacionada con la edad la enfermedad neurodegenerativa progresiva que se encuentran com&#xFA;nmente en los ancianos. Adem&#xE1;s de los d&#xE9;ficits cognitivos y de comportamiento, anomal&#xED;as en la visi&#xF3;n son frecuentes en los pacientes con EA. Estudios recientes que investigan cambios en la retina en AD doble ratones transg&#xE9;nicos han demostrado que el procesamiento alterado de la prote&#xED;na precursora de amiloide y la acumulaci&#xF3;n de p&#xE9;ptidos amiloides &#x3B2;-en las neuronas de la capa de c&#xE9;lulas ganglionares retinianas (RGCL) y la capa nuclear interna (INL). Las c&#xE9;lulas apopt&#xF3;ticas se detectaron tambi&#xE9;n en el RGCL. Age-related neurodegeneration in the brain and retina is complicated. It comprises a series of events encompassing different modes of degeneration in neurons, as well as inflammation mediated by glial cells. Systemic inflammation and risk factors can contribute to disease progression. Age-related conditions such as Alzheimer's disease (AD), Parkinson's disease (PD) and Age-related Macular Degeneration (AMD) affect patients for 5 to 20 years and are highly associated with risk factors such as hyperhomocysteinaemia, hypercholesterolaemia, hypertension, and symptoms of mood disorder. The long duration of the degeneration and the wide array of systemic factors provide the opportunity for nutraceutical intervention to prevent or delay disease progression. Small molecules such as phenolic compounds are candidates for neuroprotection because they have anti-oxidant activities and can modulate intracellular signaling pathways. Bigger entities such as oligosaccharides and polysaccharides have often been neglected because of their complex structure. However, certain big molecules can provide neuroprotective effects. They may also have a wide spectrum of action against risk factors. In this review we use an integrative approach to the potential uses of nutraceutical products to prevent age-related neurodegeneration. These include direct effects of phenolic compounds and polysaccharides on neurons to antagonize various neurodegenerative mechanisms in AD, PD and AMD, and indirect effects of these compounds on peripheral disease-related risk factors.

From small to big molecules: how do we prevent and delay the progression of age-related neurodegeneration?

Relacionada con la edad neurodegeneraci&#xF3;n en el cerebro y la retina es complicado. Se compone de una serie de eventos que abarcan los diferentes modos de degeneraci&#xF3;n en las neuronas, as&#xED; como la inflamaci&#xF3;n mediada por c&#xE9;lulas gliales. Los factores sist&#xE9;micos de inflamaci&#xF3;n y el riesgo puede contribuir a la progresi&#xF3;n de la enfermedad. Relacionados con la edad las condiciones como la enfermedad de Alzheimer (EA), enfermedad de Parkinson (EP) y relacionada con la edad degeneraci&#xF3;n macular (DMAE) afecta a los pacientes de 5 a 20 a&#xF1;os y est&#xE1;n altamente relacionados con los factores de riesgo tales como la hiperhomocisteinemia, la hipercolesterolemia, la hipertensi&#xF3;n y los s&#xED;ntomas de trastorno del humor. La larga duraci&#xF3;n de la degeneraci&#xF3;n y la amplia gama de factores sist&#xE9;micos que tengan la posibilidad de intervenci&#xF3;n de nutrac&#xE9;uticos para prevenir o retrasar la progresi&#xF3;n de la enfermedad. Las mol&#xE9;culas peque&#xF1;as, tales como los compuestos fen&#xF3;licos son los candidatos para la neuroprotecci&#xF3;n, ya que tienen propiedades anti-oxidantes y puede modular las actividades de v&#xED;as de se&#xF1;alizaci&#xF3;n intracelular. Entidades de mayor tama&#xF1;o, como los oligosac&#xE1;ridos y polisac&#xE1;ridos, han sido descuidados debido a su compleja estructura. Sin embargo, ciertas mol&#xE9;culas grandes puede ofrecer efectos neuroprotectores. Tambi&#xE9;n pueden tener un amplio espectro de acci&#xF3;n contra los factores de riesgo. En esta revisi&#xF3;n se utiliza un enfoque integral a los usos potenciales de los productos nutrac&#xE9;uticos para prevenir la neurodegeneraci&#xF3;n relacionada con la edad.
