Question 1

What is the HPA axis and how does it respond to stress?

Accepted Answer

The HPA axis is the main pathway involved in stress response, consisting of the hypothalamus, pituitary gland, and adrenal gland. When a stressor is encountered, the hypothalamus activates the pituitary gland, which releases adrenocorticotropic hormone (ACTH). This stimulates the adrenal gland to produce cortisol, the primary stress hormone responsible for physiological effects like increased blood pressure and body temperature.

Question 2

How does the resident-intruder task model social stress in rodents?

Accepted Answer

The resident-intruder task uses naturalistic aggressive behavior to generate social stress. An adult male rodent's territory is established by pairing him with a female. On test day, a novel male is introduced into the resident's home cage. The intruder experiences stress from the resident's aggression, creating a model of social stress that researchers can measure and analyze through video recording and behavioral scoring.

Question 3

What behaviors are scored when analyzing resident-intruder interactions?

Accepted Answer

Researchers record aggressive behaviors displayed by the resident, including rearing, moving towards, lateral threat, clinch attack, chase, keep down, and upright posture. Submissive behaviors displayed by the intruder include moving away, submissive posture, flight, and defensive upright posture. Additional categories such as social and exploratory behaviors can also be noted and analyzed.

Question 4

How can the resident-intruder task be adapted to study postpartum depression?

Accepted Answer

A variation introduces a novel male into the home cage of a female and her litter, providing social stress. After the male is removed, maternal behavior is assessed. Stressed females typically show depressed maternal care. Nesting behavior and marble burying are also observed as indicators of stress susceptibility or resiliency in the mother.

Question 5

What is an ethogram and why is it important in stress research?

Accepted Answer

An ethogram is a detailed list of behaviors to be recorded during an experiment. In stress research, developing an ethogram before the resident-intruder task ensures consistent and systematic observation of specific aggressive and submissive behaviors. This standardized approach allows researchers to accurately measure and compare behavioral responses across different experimental conditions.

Question 6

How do researchers induce and measure social stress in humans?

Accepted Answer

Researchers induce social stress by asking participants to deliver a job interview speech or solve arithmetic problems in front of an audience. Scientists then measure heart rate during the test and examine cortisol levels in saliva samples collected before and after the task. These physiological parameters reveal how social stress affects both cardiovascular and neuroendocrine responses.

Question 7

What does cortisol measurement reveal about stress responses?

Accepted Answer

Cortisol is the main stress hormone produced by the adrenal gland during the stress response. High cortisol levels circulating in the bloodstream indicate active stress. By measuring cortisol in saliva samples or blood, researchers can quantify the physiological intensity of stress responses in both animal models and human participants, providing objective data on stress effects.
