Soil bacteria can be counted and grown using dilution and plating methods. Surface soils contain inorganic and organic particles that form secondary aggregates. The spaces between these aggregates hold air and water. That mix creates a good habitat for bacteria, and soil often contains more than 1 million bacteria per gram.

Bacteria are the simplest microorganisms and are known as prokaryotes. A special group within this group is the actinomycetes. Actinomycetes are bacteria with a filamentous shape, meaning they form multiple cells linked together into hyphae. They are often treated as a separate group because of this structure.

This experiment uses glycerol case media to select for actinomycete colonies during dilution and plating. Actinomycetes usually make up about 10% of the total bacterial population. Bacteria and actinomycetes live in every environment on Earth, but soil has an unmatched abundance and diversity of these microbes.

These microbes matter to people in many ways. They affect what people eat, drink, breathe, and touch. Some bacterial species can cause disease, while others produce natural products that help people. Actinomycetes are especially important because they produce antibiotics such as streptomycin.

Soil bacteria also support nutrient cycling, plant growth, and the breakdown of organic contaminants. They are highly diverse because they differ in how they get energy and food. Some are heterotrophic and use organic compounds like glucose. Others are autotrophic and use inorganic compounds like elemental sulfur.

Bacteria also differ in how they respire. Aerobic bacteria use oxygen. Anaerobic bacteria use combined forms of oxygen, such as nitrate or sulfate. Some bacteria can use either oxygen or combined forms of oxygen, and these are called facultative anaerobes.

Soil Pore Space and Water Content

Soil pore space holds water and air. Soil moisture content is the amount of water present in soil. This water sits in the spaces between soil aggregates and inside the aggregates themselves.
