&alpha;-Alkylation of Ketones via Enolate Ions

Ketone alkylation with enolate ions starts when a strong base such as lithium diisopropylamide, or LDA, removes an alpha proton. This forms an enolate ion, which is stabilized by resonance. In the resonance hybrid, negative charge is shared between the carbonyl oxygen and the alpha carbon.

An enolate is an ambident nucleophile, which means it can react at two different sites. It can attack through the carbonyl oxygen, called O-attack, or through the alpha carbon, called C-attack. In this reaction, C-attack is preferred.

C-attack is favored for two reasons. The positive partner of the base interacts strongly with the anionic oxygen, which makes electrophile approach less favorable. The product from C-attack is also more stable because it keeps the stronger C=O pi bond, while O-attack would leave the weaker C=C pi bond in place.

After the enolate reacts with an electrophile such as an alkyl halide, an alpha-alkylated ketone is formed by an SN2 pathway. This reaction works with primary alkyl, benzyl, and allylic halides. With secondary and tertiary alkyl halides, elimination usually happens instead of substitution.
