Atomic absorption spectroscopy (AAS) depends on careful sample preparation and calibration. Samples must be turned into clear solutions before measurement. That often means extra treatment to dissolve materials such as soils, animal tissues, and minerals.

Common sample prep methods include hot mineral acids, wet ashing, combustion in closed containers, high-temperature ashing, and fusion with reagents. These steps help make the sample suitable for AAS analysis. Solutions that contain organic solvents, such as low-molecular-mass alcohols, esters, or ketones, can also increase absorbance. They do this by improving nebulizer efficiency and helping the solvent evaporate faster.

Immiscible solvents like methyl isobutyl ketone can be used to extract chelates of metallic ions. A chelate is a complex that helps separate the metal from other material in the sample. This can improve sensitivity and reduce interference from matrix components, which are the other substances present in the sample.

Calibration is especially important in AAS because absorbance does not always change linearly with concentration. For accurate results, scientists use periodic calibration curves and standard solutions that bracket the analyte concentration. The standard-addition method is also common. It helps correct for chemical and spectral interferences caused by the sample matrix.

Detection limits in AAS depend on the atomization technique. Flame atomization has detection limits of about 1 to 20 ng/mL. Electrothermal atomization is more sensitive, with detection limits around 0.002 to 0.01 ng/mL. Flame atomic absorption analysis usually has a relative error of a few percent. With special precautions, this can be reduced. Errors in electrothermal atomization are usually 5 to 10 times higher than in flame atomization.
