Question 1

What are exosomes and how are they formed in cells?

Accepted Answer

Exosomes are lipid bilayer-enclosed vesicles ranging from 40 to 160 nanometers in diameter. They form when intralumenal vesicles and multivesicular bodies fuse with the plasma membrane, releasing exosomes extracellularly. This process enables cells to communicate with other cells by transmitting signals and molecules across biological barriers.

Question 2

What molecules do exosomes contain and why does this matter?

Accepted Answer

Exosomes contain diverse molecules including proteins, nucleic acids, lipids, and metabolites. The specific contents determine exosome function. For example, exosomes containing interferons prevent viral infections, while those with specific membrane proteins like tetraspanins may promote viral entry. This molecular diversity makes exosomes versatile intercellular communicators.

Question 3

How do exosomes enter recipient cells after being released?

Accepted Answer

Once released from their originating cell, exosomes enter recipient cells through multiple pathways including fusion, receptor-mediated endocytosis, macropinocytosis, and phagocytosis. These varied entry mechanisms allow exosomes to deliver their cargo to diverse cell types and tissues, facilitating widespread intercellular communication.

Question 4

What roles do exosomes play in maintaining cellular health and immunity?

Accepted Answer

Exosomes maintain cellular homeostasis and elicit immune responses depending on their tissue origin and molecular content. Leukocyte-derived exosomes containing interferon-alpha prevent viral infection by suppressing virus multiplication. These functions make exosomes critical regulators of both normal physiology and disease prevention.

Question 5

How are exosomes being used as diagnostic tools for disease detection?

Accepted Answer

Exosomes found in body fluids like plasma, cerebrospinal fluid, and serum contain disease-specific biomarkers. For example, elevated CD81 protein indicates hepatitis C infection, while specific microRNAs signal prostate cancer. These biomarkers enable early disease detection and monitoring through non-invasive exosome analysis.

Question 6

Why are researchers exploring exosomes as therapeutic delivery agents?

Accepted Answer

Exosomes are stable vesicles capable of crossing biological barriers and delivering molecules to specific cellular targets. Their natural ability to carry diverse cargo and enter recipient cells makes them ideal candidates for targeted drug delivery. This therapeutic potential is driving research into exosomes as precision medicine tools.

Question 7

Where are exosomes found in the body and what do they reveal about disease?

Accepted Answer

Exosomes are present in multiple body fluids including plasma, amniotic fluid, semen, and urine. Body-fluid-derived exosomes may contain disease biomarkers; for instance, Alzheimer's disease biomarkers appear at high levels in cerebrospinal fluid exosomes from affected patients. This distribution makes exosomes valuable for non-invasive disease diagnosis.
