Question 1

Why is instrument calibration important in analytical chemistry?

Accepted Answer

Instrument calibration ensures accurate measurements by eliminating determinate errors and establishing a reliable reference standard. Without calibration, instruments produce inconsistent or incorrect results. Calibration adjusts an instrument's measured signal using a known standard, making it essential for producing trustworthy data in research, manufacturing, and testing laboratories.

Question 2

How does air buoyancy affect analytical balance measurements?

Accepted Answer

Air buoyancy makes weighted objects appear lighter in air than in a vacuum because air density reduces the effective weight. This determinate error is resolved by applying a buoyancy correction that accounts for the object's density, calibration weight density, and air density under standard laboratory conditions. The disparity between object and calibration weight densities directly influences measurement error.

Question 3

What is the relationship between concentration and absorbance in spectrophotometer calibration?

Accepted Answer

According to the Beer-Lambert law, absorbance and concentration have a direct linear relationship. A calibration curve is created by measuring absorbance of standard solutions with known concentrations, producing a straight line. Once established, this curve allows determination of unknown analyte concentrations by measuring their absorbance values and referencing the calibration curve.

Question 4

How are standard solutions used to calibrate a spectrophotometer?

Accepted Answer

Standard solutions with known analyte concentrations are prepared across a range from low to high values. Each solution is measured for absorbance using the spectrophotometer, which shines light through and measures absorption. The absorbance values are plotted against known concentrations to establish a calibration curve used for analyzing unknown samples.

Question 5

What external factors can affect analytical balance calibration?

Accepted Answer

External factors like air buoyancy significantly impact analytical balance accuracy. Air buoyancy causes objects to appear lighter than their true weight in a vacuum. Regular calibration against known standard weights compensates for these environmental effects, ensuring the balance maintains precision and provides accurate mass readings despite varying laboratory conditions.

Question 6

How does calibration differ between analytical balances and spectrophotometers?

Accepted Answer

Analytical balance calibration compares measurements against known standard weights to correct for mass determination errors. Spectrophotometer calibration involves creating a calibration curve from standard solutions with known concentrations, then measuring absorbance to determine unknowns. Each instrument requires a unique calibration process tailored to its design and measurement function.

Question 7

Why is density important when correcting for buoyancy errors in mass measurement?

Accepted Answer

Density differences between the object being weighed and calibration weights directly influence buoyancy error magnitude. Objects with lower density experience greater buoyancy effects than denser objects. The buoyancy correction formula accounts for object density, calibration weight density, and air density to calculate the true weight and eliminate this determinate error.
