Neuromuscular Activity

Neuromuscular activity is the coordinated signaling between the nervous system and skeletal muscles that produces movement and supports posture, expression, and other behavioral outputs. Motor neurons release acetylcholine at neuromuscular junctions, triggering muscle-fiber depolarization, calcium release, and actin–myosin contraction; the brain and spinal cord adjust motor-unit recruitment and firing to control force and timing. In behavioral research, electromyography and related measures reveal how muscle activation changes during locomotion, communication, learning, stress, and social interaction. Studying these patterns also helps connect observable behavior with neural control and identify functional changes associated with injury or neuromuscular disease.

The neuromuscular junction (NMJ) of Drosophila melanogaster is an important model system for studying normal synaptic function as well as perturbations to synaptic function found in certain neurological diseases. We present a protocol for dissection of the Drosophila larval motor system and immunostaining for active zone proteins within the NMJ.

This paper introduces a method for repeated measurements of ventilation and respiratory muscle activity in a freely behaving amyotrophic lateral sclerosis (ALS) mouse model throughout disease progression with whole-body plethysmography and electromyography via an implanted telemetry device.

A functional assessment of the neuromuscular junction (NMJ) can provide essential information on the communication between muscle and nerve. Here we describe a protocol to comprehensively evaluate both the NMJ and muscle functionality using two different muscle-nerve preparations, i.e. soleus-sciatic and diaphragm-phrenic.

Assessing Muscle Contraction with Neuromuscular Blocker Treatment

This video demonstrates assessing the muscle using cultured motor neurons, Schwann cells, and myotubes, followed by treatment with a neuromuscular blocker to inhibit contraction. Color-coded time-motion graphics show a color-coded time-motion graphic. Red indicates the fastest movement, and blue indicates the slowest.

Assessment of Neuromuscular Function Using Percutaneous Electrical Nerve Stimulation

We present a protocol to assess changes in neuromuscular function. Percutaneous electrical nerve stimulation is a non-invasive method that evokes muscular responses. Electrophysiological and mechanical properties of these responses permit the evaluation of neuromuscular function from brain to muscle (supra-spinal, spinal and peripheral levels).
