Lumen Like Structures

Lumen-like structures are engineered or cell-generated, hollow, cavity-containing arrangements that reproduce key features of biological tubes and ducts. In three-dimensional culture, cells organize around a fluid-filled space through coordinated polarity, cell–cell junction formation, cytoskeletal remodeling, and interactions with an extracellular matrix, producing a defined inner surface and surrounding tissue. Bioengineers use these structures to model vascular, epithelial, and glandular organization, assess transport across tissue barriers, and study how changes in matrix composition or signaling affect morphogenesis. Their development supports organoid design, disease modeling, drug testing, and the construction of more physiologically relevant engineered tissues.

Source: Li, J. et al. Development of a Larval Zebrafish Infection Model for Clostridioides difficile. J. Vis. Exp. (2020)This video demonstrates the microinjection-based delivery of fluorescently labeled Clostridioides difficile into the intestinal lumen of an anesthetized zebrafish larva. It outlines the steps involved in larval preparation, bacterial injection, and fluorescence microscopy–based confirmation of bacterial localization within the intestine.

Gold Immunostaining of Exosomal Sections: A Technique to Visualize Diagnostic Markers in Exosomal Lumen

2023

In this video, we demonstrate gold immunostaining of biomarkers localized within the lumen region of ultrathin exosomal sections. This technique enables early diagnosis of diseases by visualization of target molecules within the exosomes secreted by the diseased cells.

Source: Patras, K. A. & Doran, K. S. A Murine Model of Group B Streptococcus Vaginal Colonization. J. Vis. Exp. (2016)This video demonstrates the quantification of vaginal Group B Streptococcus (GBS) load in a mouse model using swabbing and chromogenic culture. It outlines the steps involved in sample collection, plating, and colony counting to quantify GBS load in the mouse vaginal lumen.

Gut Acidification Monitoring Assay: A Technique to Study the Acidification of the Intestinal Lumen in Drosophila using Bromophenol Blue Dye

2025

This video demonstrates an assay to monitor gut acidification in Drosophila flies. After the ingestion of food supplemented with a pH indicator dye, the acidification of the intestinal lumen results in a change in the color of the dye. Visual inspection of the digestive tract of the flies to examine the color change provides evidence of acidification.

Source: Roberto Leon, Department of Civil and Environmental Engineering, Virginia Tech, Blacksburg, VA It is rare nowadays that a whole year goes by without a major earthquake event wreaking havoc somewhere around the world. In some cases, like the 2005 Banda Ache earthquake in Indonesia, the damage involved large geographic areas and casualties in the six figures. In general, the number and intensity of earthquakes is not increasing, however, the vulnerability of the built environment is...
