Microfabricated Devices

Microfabricated devices are miniaturized structures and systems produced with microscale manufacturing techniques, enabling precise control of fluids, cells, biomolecules, and mechanical forces. In bioengineering, processes such as photolithography, etching, and soft lithography pattern materials into channels, sensors, membranes, and three-dimensional scaffolds that guide transport and biological interactions. These devices support lab-on-a-chip platforms, microfluidic assays, tissue models, biosensors, and controlled drug delivery by reducing sample volumes and integrating multiple functions in compact formats. Their reproducible architecture helps researchers study cellular behavior under defined conditions and develop faster, more personalized approaches to diagnosis, disease modeling, and regenerative medicine.

Microfabrication via Photolithography

The fabrication of BioMEM devices is often done using a microfabrication technique called photolithography. This widely used method utilizes light to transfer a pattern onto a silicon wafer, and provides the basis for the fabrication of many types of BioMEM devices. This video presents the photolithography technique, shows how the process is performed in the cleanroom, and introduces some applications of the process.

Study of Cell Migration in Microfabricated Channels

A quantitative method to study spontaneous migration of cells in a one-dimensional confined microenvironment is described. This method takes advantage of microfabricated channels and can be used to study migration of large number of cells under different conditions in single experiments.

In this protocol, we demonstrate the fabrication of a microactuator array of vertically displaced posts on which the technology is based, and how this base technology can be modified to conduct high-throughput mechanically dynamic cell culture in both two-dimensional and three-dimensional culture paradigms.
