Rabbit Calvarial Model

The rabbit calvarial model is an in vivo experimental system that uses the rabbit skull to study bone healing and evaluate bioengineered materials. Researchers create standardized defects in the calvarial bone and place scaffolds, grafts, cells, or therapeutic agents within them, then assess tissue formation through imaging, histology, and mechanical or molecular analyses. Because the relatively accessible skull supports controlled comparisons between treated and untreated sites, this model helps measure osteogenesis, vascularization, biomaterial integration, and defect repair. Its findings guide the development of bone substitutes, tissue-engineered constructs, and regenerative strategies before translation to larger animal studies or clinical research.

Here we present a surgical protocol in rabbits with the aim to assess bone substitution materials in terms of bone regeneration capacities. By using PEEK cylinders fixed onto rabbit skulls, osteoconduction, osteoinduction, osteogenesis and vasculogenesis induced by the materials may be evaluated either on live or euthanized animals.
