Sample Isolation

Sample isolation is the process of separating a target biological component from a complex specimen so it can be examined, measured, or used in downstream experiments. In biological techniques, isolation commonly combines physical or chemical steps such as filtration, centrifugation, selective lysis, affinity capture, or microdissection, using differences in size, density, solubility, surface markers, or other properties to enrich the material of interest while limiting contamination. Careful control of collection conditions, temperature, handling, and separation parameters helps preserve sample integrity and biological activity. Isolated cells, tissues, nucleic acids, proteins, or microorganisms support microscopy, molecular analysis, diagnostics, culture, and reproducible research.

Source: Cross, T., et al. An Optimized Enrichment Technique for the Isolation of Arthrobacter Bacteriophage Species from Soil Sample Isolates. J. Vis. Exp.(2015)This video demonstrates the isolation of bacteriophages from a soil sample. A soil sample is collected and suspended in a phage buffer and allowed to settle the sediment. The extract is filtered, and nutrients are added. A vacuum filtration is done to remove microbial contaminants. The filtrate with phages is aliquoted into flasks, and...

Source: Laboratories of Dr. Ian Pepper and Dr. Charles Gerba - The University of Arizona Demonstrating Author: Luisa Ikner The quality of water destined for use in agricultural, recreational, and domestic settings is of great importance due to the potential for outbreaks of waterborne disease. Microbial agents implicated in such events include parasites, bacteria, and viruses that are shed in high numbers in the feces of infected people and animals. Transmission to new and susceptible hosts may...

(2015).This video demonstrates the isolation of Arthrobacter bacteriophages from a soil sample using enrichment and plaque assay techniques. It highlights the infection, amplification, and visualization of phages as clear lysis zones formed on a bacterial lawn.
