Apoplast Washing Fluid

Apoplast washing fluid is the liquid recovered from the extracellular spaces of plant tissues, where it contains soluble proteins, metabolites, hormones, ions, and signaling molecules that influence plant physiology. Researchers typically obtain it by infiltrating tissue with a solution under vacuum and then centrifuging the tissue to collect fluid displaced from the apoplast while minimizing cellular damage. Analysis of apoplast washing fluid provides a window into the plant cell wall environment and helps assess nutrient movement, defense responses, pathogen interactions, and stress signaling. Because its composition changes during infection or environmental stress, it supports studies of plant immunity, cellular communication, and extracellular biochemical processes.

(2022)This video demonstrates a syringe-based pressure method for extracting apoplastic bacteria, which proliferate and form microcolonies within the apoplast, the intercellular space between plant cells. The method yields debris-free bacterial samples suitable for downstream single-cell analysis.

The Apoplast and Symplast

2026

Plant growth depends on its ability to take up water and dissolved minerals from the soil. The root system of every plant is equipped with the necessary tissues to facilitate the entry of water and solutes. The plant tissues involved in the transport of water and minerals have two major compartments - the apoplast and the symplast. The apoplast includes everything outside the plasma membrane of living cells and consists of cell walls, extracellular spaces, xylem, phloem, and tracheids. The...

Washing, Drying, and Ignition of Precipitates

After filtration, the precipitate is washed to remove coprecipitated impurities and any remaining mother liquor. Colloidal precipitates, such as silver chloride, are washed with an electrolyte (such as dilute nitric acid) to prevent the peptization of the precipitate. In the case of slightly soluble precipitates, the wash solution contains a common ion to reduce solubility. Lead sulfate, which is slightly soluble in water, is washed with dilute sulfuric acid. Similarly, wash solutions may be...
