Bifc Validation

BiFC validation is a fluorescence-based method for testing whether two proteins interact in cells, linking molecular binding to cellular location. It uses bimolecular fluorescence complementation, in which a fluorescent protein is divided into two nonfluorescent fragments and each fragment is fused to a candidate partner; interaction brings the fragments together, allowing the fluorophore to reconstitute and emit detectable fluorescence. Researchers compare the resulting signal with appropriate controls and examine where complexes form. In biology, BiFC validation supports studies of protein networks, signaling pathways, and subcellular localization while helping assess interaction specificity and cellular function.

We have developed a technique to test protein-protein interactions in plant. A yellow fluorescent protein (YFP) is split into two non-overlapping fragments. Each fragment is cloned in-frame to a gene of interest via Gateway system, enabling expression of fusion proteins. Reconstitution of YFP signal only occurs when the inquest proteins interact.
