Bowman's Capsule

Bowman’s capsule is a double-walled, cup-shaped structure at the beginning of each nephron that surrounds the glomerulus and forms the renal corpuscle. Blood pressure drives ultrafiltration across the glomerular filtration barrier, allowing water and small dissolved substances to enter the capsule’s urinary space while retaining blood cells and most plasma proteins; the resulting filtrate then passes into the proximal tubule. Studying Bowman’s capsule clarifies how kidneys initiate urine formation, regulate the composition of body fluids, and maintain homeostasis. Changes in its structure or filtration barrier can contribute to kidney disorders and provide important clues for understanding renal function.

By centrifuging a transposon mutant library through the gradient, bacteria with different capsule densities localize to distinct layers. This protocol provides instruction for negative staining virus samples which can easily be used in BSL-2, -3, or -4 laboratories. It includes the use of an innovative processing capsule, which protects the transmission electron microscopy grid and provides the user easier handling in the more turbulent environments within biocontainment.
