Branching Morphogenesis

Branching morphogenesis is a developmental process in which epithelial tissues repeatedly form and remodel branches, creating the complex architectures of organs such as the lungs, kidneys, and mammary glands. This process matters because branch patterning expands functional surface area while positioning specialized cells and ducts. During development, interactions between epithelial tips and surrounding mesenchyme, mediated by growth factors, extracellular matrix, and cell adhesion, regulate proliferation, migration, and directional outgrowth. Studying branching morphogenesis helps explain congenital abnormalities and tissue organization, while informing organoid models, regenerative medicine, and strategies for repairing or engineering branched organs.

In this video, we describe a qualitative spectrophotometric assay to determine the extent of branching in a glycogen sample with an uncharacterized structure. Comparing the absorbance maxima of the uncharacterized sample with that of polysaccharides with known branched structures indicates the presence of branching in the uncharacterized sample.
