Cesium Chloride Gradient

Cesium chloride gradient centrifugation is a density-gradient separation technique that resolves biological molecules and particles according to buoyant density, making it useful for isolating and characterizing nucleic acids. In a concentrated cesium chloride solution, ultracentrifugation generates a stable density gradient; components move through the solution until they reach positions where their density matches that of the surrounding cesium chloride, often with DNA separated further by buoyant-density differences. The resulting bands can be collected for purification or analysis, supporting plasmid DNA isolation, viral particle characterization, genome studies, and assessment of nucleic-acid composition. Its high resolution remains valuable when closely related molecular species must be distinguished.
