Chicken Egg Windowing

Chicken egg windowing is a technique for accessing a developing avian embryo inside an intact fertilized egg, allowing researchers to observe and manipulate development without removing the embryo. The procedure involves creating a small opening in the eggshell, carefully exposing the underlying membranes and embryo, and resealing the window to maintain suitable conditions for continued incubation. This approach supports studies of embryology, organ formation, vascular development, and tissue patterning, while enabling targeted delivery of experimental materials or collection of developmental data. Because the embryo remains within its natural nutrient-rich environment, windowing provides a practical model for investigating biological processes over time.

Source: Brauer, R. et al., Influenza Virus Propagation in Embryonated Chicken Eggs. J. Vis. Exp. (2015)This video demonstrates the synthesis and harvest of the influenza virus in fertilized chicken eggs. The virus is inoculated into the allantoic cavity, where it replicates in epithelial cells and buds into the allantoic fluid, which is then collected and stored for downstream applications.

Source: Yates, J. G. E., et al. Production of High-Titer Recombinant Newcastle Disease Virus from Allantoic Fluid. J. Vis. Exp. (2022)The video demonstrates the purification and concentration of virus particles from clarified allantoic fluid collected from virus-infected embryonated chicken eggs. It showcases a series of filtration and separation steps, including depth filtration, tangential flow filtration, and density gradient ultracentrifugation to isolate intact viruses. The virus-rich...
